Test the finished, processed print with the cell type and exposure route that match its intended use. A cytotoxicity assay can show whether that specific material-and-process combination harms cells under stated conditions; it cannot prove that a material is universally biocompatible, chemically harmless, sterile, or suitable for every culture system.

What does it mean for a 3D-printed material to be safe for cell culture?

It means the final printed article performs acceptably in a defined cell-based evaluation for a particular application—not that the material has a blanket safety status. Cell response can vary with the cell model, exposure route, material formulation, print and post-processing steps, and sterilization method.

Set the question narrowly: for example, whether a particular finished printed component, processed by a stated workflow, causes an adverse response in the cell type that will contact it. Record the biological endpoint you will use, such as cell morphology, damage, growth, or cellular metabolism. There is no single cytotoxicity endpoint that answers every use case.

Keep the conclusions within the limits of the test. A result from one cell type or exposure method does not establish performance with other cell types, culture conditions, or applications. A cell assay also does not establish sterility or provide a complete inventory of chemicals that may leach from the print.

What should I define before testing?

Define the intended use and the test article before choosing an assay. The test should represent the finished component—or specimens produced through the same manufacturing and processing route—not merely the raw resin or filament.

Record the complete workflow

  • Material formulation and, where available, the relevant lot or batch.
  • Printer, print parameters, and build orientation or other settings that distinguish the tested article.
  • Support materials and their removal method.
  • Cleaning, washing, curing, coatings, and other post-processing.
  • Disinfection or sterilization method, if the intended workflow uses one.
  • Storage conditions between processing and testing.
  • The cell type, expected contact or exposure, and duration relevant to the intended culture use.

These details define what your result applies to. If a material or processing step changes, the old test may no longer represent the article you plan to use. ISO 10993-12:2021 addresses sample selection and preparation, controls, reference materials, and extract preparation; its listing shows Amendment 1 published in 2025.

Make the test article representative

A coupon made from the same feedstock is not automatically equivalent to a finished device. Differences in print settings, support removal, washing, curing, surface coatings, storage, or sterilization can change what remains on or in the article and what cells encounter. Document the route from printing to exposure, and use that same route for every specimen intended to support the decision. If the culture will contact a surface directly, the tested surface should have the same relevant finish and treatment as the production article.

Define the exposure question before selecting specimens. A component that only touches the outside of a culture vessel raises a different question from a scaffold that remains in contact with cells, and an extract-only screen answers a different question from direct placement. The method, article geometry, and processing state should therefore be planned together rather than selected independently.

Which exposure method should I use?

Choose an exposure method based on how cells will encounter the printed article and what the test needs to detect. ISO 10993-5:2009 describes direct contact, indirect contact or diffusion, and testing with a liquid extract; its official listing says the edition was reviewed and confirmed in 2022 and remains current.

Method What it evaluates When it can be useful
Direct contact Cells exposed to the material itself in the test setup. When the intended use involves cell contact with the printed surface and the setup can make that contact meaningfully.
Indirect contact or diffusion Effects when material-associated substances reach cells without placing the test article directly on them. When diffusion-mediated exposure is relevant or direct placement is unsuitable for the specimen or cell system.
Liquid extract Effects of substances released from a specimen into a selected extraction liquid. When the question concerns releasable substances under the chosen extraction conditions.

These approaches are not interchangeable. An extract test depends on how the extract was prepared, while direct and indirect contact depend on the geometry and exposure arrangement. State the chosen method and its conditions so readers can understand the exposure the cells actually received. ISO 10993-5 provides a set of options, not one universally required assay or exposure method.

Match the exposure to the intended use

Use direct contact when the cell layer or cell-laden construct will actually touch the printed surface and the setup allows meaningful contact. Use an indirect or diffusion arrangement when substances can reach cells without the article being placed on them, or when direct placement would confound the observation. Use an extract when the practical concern is what the specimen releases into a selected liquid under documented conditions. A negative extract result does not rule out an effect caused by the physical surface, and a direct-contact response does not identify which constituent caused it.

For every approach, record the specimen identity, extraction or contact arrangement, exposure duration, and the state of the article at the moment of exposure. Without those details, two nominally identical tests may be exposing cells to different chemical or physical conditions.

How do I set up a useful cell-based test?

Use a fit-for-purpose assay with controls that help distinguish material effects from problems in the test setup. Pair quantitative measurements with visual inspection when that adds useful context.

  1. Prepare representative specimens. Use the material and workflow defined for the intended application, including post-processing and any intended sterilization or disinfection.
  2. Prepare the exposure. For an extract test, document the extraction conditions and include an extraction blank. For contact tests, document how specimens are positioned relative to the cells.
  3. Include suitable controls. Use appropriate negative and positive controls as well as the extraction blank where extracts are used. Controls help show whether the assay can distinguish an acceptable response from a harmful one under the test conditions.
  4. Choose and state the endpoint. Identify whether you will assess morphology, cell damage, growth, cellular metabolism, or another justified biological response. Do not treat one endpoint as a universal substitute for all others.
  5. Apply a defined exposure and readout schedule. State the exposure duration and the time and method used to assess the endpoint, so the result can be interpreted in context.
  6. Inspect and report the response. Report the quantitative result alongside relevant visual observations, including the test conditions and control performance.

The available standards information does not prescribe a universal cell line, exposure duration, assay kit, or pass threshold for every cell-culture use. Select these to fit the biological question and the applicable requirements rather than implying that one protocol covers every printed material.

Use controls to expose test problems

An extraction blank shows whether the liquid, container, or handling procedure contributes a response independent of the printed specimen. Negative and positive controls indicate whether the cell system and assay are behaving as expected and whether the test can discriminate an acceptable response from a harmful one. If controls do not perform as intended, a result from the print is difficult to interpret. Report control observations rather than merely stating that controls were included.

Do not rely on one readout alone

Cell morphology can reveal changes that a single numerical measurement misses, while measurements of damage, growth, or metabolism provide quantitative evidence. The endpoint should follow the biological question. For example, a material may not produce an immediate visible change yet still reduce growth or metabolic activity under the stated exposure. Conversely, a visual change needs to be interpreted with the assay and controls rather than treated as proof of a specific chemical cause.

What can make a result misleading?

  • Testing raw feedstock only: The resin or filament is not the same test article as a washed, cured, coated, disinfected, or sterilized print.
  • Changing the workflow after testing: A new printer setting, support-removal method, curing step, coating, storage interval, or sterilization method can make earlier evidence less representative.
  • Using an unmatched cell model: A result in one cell type should not be presented as a result for every culture type.
  • Confusing sterility with cytotoxicity: Cells can remain viable in a contaminated system, and a sterile article can still release cytotoxic substances. These are separate questions.
  • Treating a treatment comparison as a universal recipe: A method that reduced toxicity for one polymer may not do so for another formulation or print process.
  • Overlooking the exposure route: Extract, indirect, and direct-contact results answer different questions and should not be substituted for one another without justification.

These pitfalls are reasons to describe the material-and-process combination precisely, not reasons to label every printed material unsafe. A narrowly stated result is more useful than an unsupported universal claim.

When should I investigate leachables or chemical composition?

A cell-based cytotoxicity result is a biological screen, not a chemical inventory. If the intended use or observed response makes the identity or amount of released substances important to risk assessment, consider chemical characterization in addition to the cell assay.

ISO 10993-18:2020 provides a stepwise framework for identifying and, where needed, quantifying material constituents to support hazard identification and risk estimation. The ISO listing says the edition was confirmed in 2025 and that Amendment 1 was published in 2022. Chemical characterization supports a risk assessment; it does not by itself demonstrate that a printed article is suitable for a particular culture.

In a 2023 study, Huang and colleagues found leachable toxicity in the three stereolithography polymers they tested. Flexibility and treatment method affected the observed response. Soxhlet solvent extraction was the most effective treatment among those evaluated in that study, but that finding is specific to its materials and methods—not a general detoxification recipe for other polymers or workflows.

Consider chemical follow-up when a biological result is adverse, when the article has several constituents or processing aids, or when the intended exposure makes released substances especially consequential. The purpose is to identify and, where needed, quantify constituents so that hazards and estimated exposure can be considered together. Chemical data and cell data answer complementary questions; neither replaces the other.

Why do cell type and processing change the result?

Cell models and processing steps can change what a test shows, so interpret published findings as evidence about the tested combinations rather than guarantees about a whole material family.

Study What was evaluated Reported finding and scope
Winkler and colleagues, 2022 Extraction media from a heat-resistant polyacrylate, evaluated with L929 mouse fibroblasts, HEK 293E cells, and yeast. No negative effect was reported for L929 cells and yeast under the study conditions, while HEK 293E viability or growth was reduced. The study also found no different cell effects between autoclaving and ethanol disinfection for the specific polyacrylate and cell systems tested.
Licata and colleagues, 2024 3D-printed plastics considered for bioreactors, including photoresins and thermoplastics. The tested photoresins affected human induced pluripotent stem cells but not primary human dermal fibroblasts. Among the thermoplastics tested, only Nylon 12 withstood autoclaving. These results are limited to the materials and conditions in that study.
Huang and colleagues, 2023 Three stereolithography polymers and methods intended to address leachable cytotoxicity. Leachable toxicity was reported in the tested materials, with observed responses affected by flexibility and treatment method. The comparative treatment finding applies only to the methods and polymers evaluated.
“Evaluating the Cytotoxicity of Commercial Materials for Additive Manufacturing of Soft Tissues,” 2026 Formlabs Biomed Elastic 50A, Biomed Flexible 80A, and Biomed Durable resin, tested with human dermal fibroblasts and human mesenchymal stromal cells. The study reported minimal cytotoxicity after its tested printing and post-processing conditions. It does not establish results for other resins, lots, cell types, or workflows.

The contrast between cell models is consequential: a material that appears acceptable with one cell type may produce a different response with another. Likewise, a result before sterilization does not establish how the intended sterilization process affects the article or cells.

The Winkler finding about autoclaving and ethanol disinfection should be read narrowly. It means those processes produced no different cell effects for the specified heat-resistant polyacrylate and cell systems in that evaluation. It does not show that every resin tolerates autoclaving, that ethanol removes all relevant leachables, or that either process is appropriate for your article.

Licata’s observation that only Nylon 12 among the tested thermoplastics withstood autoclaving is likewise a compatibility result for that set of materials and conditions. If heat sterilization is required, assess whether the candidate material survives it and whether the post-process article still gives the intended cell response.

How should I compare candidate printed materials?

Compare evidence at the level of the intended application, not by material name alone. Give greatest weight to whether the evidence matches your target cell type, exposure route, and required processing.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.
  • Target-cell response: Does the evidence use the cell type you plan to culture, or a justified relevant model?
  • Exposure match: Is the evidence from direct contact, indirect contact, or extracts, and does that reflect the exposure you need to assess?
  • Chemical evidence: Is there information about releasable substances or chemical characterization where the risk question calls for it?
  • Workflow compatibility: Was the tested article cleaned, cured, coated, disinfected, or sterilized in the same way you intend to process yours?

A published result for a named resin or polymer can help identify a promising candidate, but it is not a substitute for checking whether the study’s cell model and manufacturing route represent your own use. A material with favorable fibroblast data may still require testing with stem cells, epithelial cells, or another target model, and a material that survives one disinfection method may not survive another.

What do the ISO standards establish—and what do they not?

The ISO 10993 documents offer structured methods for biological evaluation and sample preparation, but they do not turn one cell assay into proof of broad safety or regulatory clearance. They concern medical-device materials; a laboratory cell-culture setup may use them as a reference, while the intended application and applicable requirements determine what evaluation is appropriate.

Standard Relevant scope Edition status in the cited ISO listing
ISO 10993-5 In vitro cytotoxicity test methods, including direct contact, indirect contact, and extracts. 2009 edition; reviewed and confirmed in 2022, and listed as current.
ISO 10993-12 Sample preparation, reference materials, controls, and extracts. 2021 edition; Amendment 1 published in 2025.
ISO 10993-18 Chemical characterization within a risk-management process. 2020 edition; confirmed in 2025, with Amendment 1 published in 2022.

If you are writing a formal protocol or evaluation plan, the official ISO listing for ISO 10993-5 identifies purchasable formats. Obtaining the standard can guide test design; it does not establish that a particular print is suitable for cell culture. The standards are frameworks for selecting and documenting methods, not certificates attached to a resin name.

How should I report the result?

Write the conclusion so another laboratory can tell exactly what was tested and how far the finding can reasonably be generalized. Include the details that define the test article, exposure, and biological result.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.
  • Material, specimen, and relevant formulation or lot information.
  • Printer and print settings, plus cleaning, curing, support removal, coatings, storage, and sterilization or disinfection.
  • Cell type and exposure method, including extraction conditions when applicable.
  • Exposure duration, assay or endpoint, and how the response was assessed.
  • Extraction blank and positive and negative controls, with their observed performance.
  • Quantitative findings and relevant morphology or other visual observations.
  • A conclusion limited to the tested cell model, exposure, processing state, and conditions.

State what the test did not examine when that limitation matters—for example, a chemical inventory, another cell type, a different sterilization method, or long-term exposure. This prevents readers from treating a narrow result as a product-wide guarantee.

Prefer a conclusion such as “the processed specimens did not produce the measured adverse response in this cell model under the stated exposure conditions” over the unqualified claim “the material is biocompatible.”

Frequently Asked Questions

Can a supplier’s biocompatibility claim tell me whether a print will work in my culture?

Only if the underlying evaluation matches the relevant formulation, print and post-processing route, cell type, and exposure. Ask what specimens and endpoints were tested and treat evidence from a different workflow as limited rather than as a guarantee for your setup.

Does a cell-culture cytotoxicity test establish regulatory approval?

No. A cytotoxicity result is one part of a biological evaluation, and the ISO 10993 standards discussed here address medical-device materials. Applicable regulatory requirements depend on the intended application; a single assay does not establish clearance.

Should I repeat testing after changing a print or post-processing step?

A change can make earlier results less representative, especially if it changes the formulation, surface, cleaning, curing, or sterilization state. Assess whether existing evidence still covers the finished article and its intended exposure before relying on it.

Can the same printed material behave differently in two culture systems?

Yes. Published studies summarized here reported different responses across cell models, including distinct results for L929, HEK 293E, and yeast in one study, and for induced pluripotent stem cells versus dermal fibroblasts in another. That is why evidence should be matched to the cells you intend to use.

Does passing an extract test prove that direct contact is safe?

No. Extract testing evaluates substances released under its extraction conditions. Direct contact also involves the physical printed surface and a different exposure arrangement, so use the method that reflects the intended culture contact.

Is autoclaving a reliable way to make any printed biomaterial safe?

No. Sterilization compatibility and cytotoxicity are separate questions. One study found no different cell effects after autoclaving versus ethanol disinfection for a specific heat-resistant polyacrylate, while another reported that only Nylon 12 among its tested thermoplastics withstood autoclaving. Those findings do not generalize to every resin.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

When is chemical characterization worth adding to the cell assay?

Add it when the identity or amount of released constituents matters to the risk assessment, such as after an adverse biological result or when the formulation and processing involve several potential constituents. ISO 10993-18 provides a framework for identifying and, where needed, quantifying them.

Is Soxhlet extraction a universal detoxification step?

No. Huang and colleagues found it most effective among the treatments they tested for three stereolithography polymers in 2023. The result is specific to those polymers and methods; another material or workflow may behave differently.

The Bottom Line

Test the finished print after every processing step that defines its use, with a cell model and exposure route that represent the intended culture. Use controls, report the endpoint and conditions, investigate leachables when the risk question requires it, and limit the conclusion to the material, workflow, cells, and exposure actually evaluated.
