Choose the printer wavelength first, then screen for water compatibility, cell safety and curing behavior. For a 405 nm printer making aqueous, cell-laden hydrogels, lithium phenyl-2,4,6-trimethylbenzoylphosphinate (LAP) is usually the most sensible first candidate. For approximately 365 nm ultraviolet systems, Irgacure 2959 remains a useful comparator, provided its limited water solubility and ultraviolet dose are acceptable. The right choice is not a universal “best” molecule: initiator concentration, irradiance, exposure time, resin composition, photo-absorber, cell type and post-cure treatment all change the result.

A good screen therefore measures more than whether a construct gels. Compare gel time, conversion, cure depth, feature fidelity, viability during and after printing, swelling, degradation, pore structure, modulus and residuals in the exact bioink and printer you plan to use.

What does a photoinitiator do in a bioprinting resin?

A photoinitiator (PI) absorbs light and produces reactive species—usually free radicals—that start polymerization of light-sensitive groups such as methacrylates. In vat photopolymerization (VPP), digital light processing (DLP), stereolithography and related methods, the PI determines how efficiently each projected layer turns liquid precursor into a solid network.

For cell-laden materials, the PI is part of a coupled biological and optical system. The initiator must absorb where the printer actually emits, dissolve or remain uniformly dispersed in the aqueous formulation, generate radicals quickly enough for the exposure schedule, and avoid unacceptable damage to encapsulated cells. A formulation that cures rapidly but leaves high residuals, excessive swelling or poor viability is not a successful biomaterial.

How should I match a photoinitiator to the printer wavelength?

Measure or confirm the emission spectrum and irradiance at the resin plane, not merely the nominal wavelength printed on the machine. Select an initiator whose absorption overlaps that spectrum; a mismatch lowers radical generation and can produce incomplete or inconsistent cure.

Printer or light band Initiators reported in the literature Practical implications
Approximately 365 nm Irgacure 2959 Established UV option with low reported toxicity, but poor water solubility and ultraviolet exposure can complicate cell-laden inks.
380–405 nm LAP; VA-086; Na-TPO (380–450 nm activity) LAP is a strong first screen for aqueous 405 nm workflows. VA-086 has very low reported toxicity but can release nitrogen bubbles.
300–500 nm Riboflavin (vitamin B2) Very water-soluble and low-toxicity type-II system; often paired with a co-initiator such as triethanolamine (TEOA).
Approximately 452 nm Ruthenium complexes with sodium persulfate The persulfate partner is required; the metal complex alone is not a complete initiating system.
Approximately 528 nm Eosin Y Visible-light type-II chemistry that requires an appropriate co-initiator and careful dose control.
520–550 nm Rose Bengal Another visible-light type-II option; co-initiator choice and exposure strongly affect cell response.

The wavelength bands above come from a 2026 review by Ng and colleagues. They are screening guides, not guarantees: absorption depends on solvent, pH, concentration and other resin components. A photo-absorber added to control cure depth can further reduce the light available to the PI, so measure the complete formulation rather than relying on a datasheet spectrum.

Is LAP or Irgacure 2959 better for cell-laden 405 nm printing?

For many aqueous 405 nm bioinks, start with LAP. The 2026 review describes it as a type-I initiator absorbing at 380–405 nm, with good water solubility, low reported toxicity and high reactivity. It generally gels faster than Irgacure 2959, although LAP cures more slowly at 405 nm than at 365 nm. Irgacure 2959 is better aligned with approximately 365 nm UV and remains useful when that optical setup and formulation are already validated.

Criterion LAP Irgacure 2959
Typical spectral fit 380–405 nm; a natural first screen for 405 nm printers Near 365 nm
Water behavior Good water solubility reported Poor water solubility; may require more difficult formulation or higher concentration
Cure behavior Highly reactive; faster gelation than Irgacure 2959 in the cited review; 405 nm is slower than 365 nm Usable UV curing, but slower or less practical when the ink is aqueous and the printer is 405 nm
Cell-related concern Still sensitive to concentration, irradiance and exposure time; higher radical load can reduce viability Low reported toxicity, but UV exposure and poor dissolution are practical drawbacks for encapsulated cells
Observed material differences in a GelMA study At high concentrations, better viability during printing than Irgacure 2959 Slightly larger pores, faster degradation and greater swelling in the cited comparison

In a 2020 head-to-head study of GelMA vascular-like constructs by Heqi Xu and the Texas Tech University team, both initiators produced good overall viability during a 60-minute printing period at 0.3% and 0.5% (w/v). At 0.7% and 0.9% (w/v), viability during that printing period was much higher with LAP than with Irgacure 2959. Viability after crosslinking was not strongly determined by initiator type in that experiment, so the result should be treated as formulation- and protocol-specific rather than a universal safety threshold.

Which candidate fits a specific biomaterial workflow?

LAP for aqueous 405 nm hydrogels

LAP combines the optical fit, water solubility and reactivity that make it a practical first screen for many 405 nm cell-laden hydrogels. Begin with a concentration matrix rather than assuming that faster gelation is always better. As concentration or print time rises, radical exposure rises too; that can improve layer locking while harming cells or changing network structure.

Irgacure 2959 for approximately 365 nm UV systems

Irgacure 2959 has extensive use and low reported toxicity, but its poor water solubility can cause incomplete dissolution or force a higher loading. The ultraviolet dose must be validated with the target cell type. A formulation that looks clear by eye can still have local concentration gradients or precipitated material that alter cure and toxicity.

Riboflavin with a co-initiator

Riboflavin (vitamin B2) is a naturally derived type-II initiator with excellent water solubility, very low reported toxicity and broad 300–500 nm absorption. Type-II chemistry needs a co-initiator. In a 2022 study, Chia-Chun Wang and colleagues at National Tsing Hua University used a vitamin B2/triethanolamine (B2/TEOA) system in biodegradable PGSA formulations. The reported films sustained cell proliferation for 10 days and showed low immune responses 14 days after implantation in that study, while DLP printing required 100 seconds per layer. Those results describe one PGSA-B2/TEOA formulation and exposure schedule, not a general clinical performance claim.

VA-086

VA-086 is reported for 385–405 nm use with very low toxicity and high cell viability. Its decomposition can release nitrogen, however, creating bubbles that become defects, scatter light and reduce dimensional accuracy. Degassing and bubble inspection are therefore part of the process, not optional finishing steps.

Na-TPO and visible-light systems

Na-TPO is water-soluble and active at 380–450 nm, making it another candidate for near-405 nm systems. Ruthenium/sodium persulfate systems absorb around 452 nm and require the persulfate co-reactant. Eosin Y (about 528 nm) and Rose Bengal (520–550 nm) operate in visible light and require carefully selected co-initiators and dose control. Type-II systems should be screened as complete pairs, including co-initiator concentration and oxygen conditions.

What concentration should I test first?

Use literature values to define a screening window, not as universal operating limits. A 2023 review table lists Irgacure 2959 at 0.05–0.7% (w/v), LAP at 0.05–0.5% (w/v) and VA-086 at 0.5–1.5% (w/v). The suitable point inside—or outside—those ranges depends on optical power, layer thickness, resin solids, photoinhibitors, exposure time and cell sensitivity.

Initiator Reported review range How to use the number
Irgacure 2959 0.05–0.7% (w/v) Screen near the lower end first in cell-laden aqueous inks because solubility and UV dose are limiting factors.
LAP 0.05–0.5% (w/v) Build a low-to-moderate matrix around the optical dose needed for complete cure; do not assume the upper end is safer because cure is faster.
VA-086 0.5–1.5% (w/v) Include bubble inspection and degassing in every condition because nitrogen release can undermine prints.

For each candidate, prepare several concentrations with the same polymer, cell density, layer thickness, exposure and post-cure. Record the actual mass, final volume and whether the percentage is w/v. Keep stock solutions protected from light and document storage time, temperature and any filtration or sterilization step; these details can change effective concentration.

How do I run a photoinitiator screening experiment?

  1. Characterize the printer. Confirm the nominal wavelength, measure irradiance at the resin plane with an appropriate calibrated meter, and record optics, pixel size, layer thickness and exposure time. Do not substitute the lamp’s rated power for delivered irradiance.
  2. Shortlist by absorption. For 405 nm, begin with LAP or Na-TPO; for approximately 365 nm, include Irgacure 2959; for visible systems, evaluate the complete riboflavin, ruthenium, Eosin Y or Rose Bengal chemistry with its required co-initiator.
  3. Make a concentration matrix. Use at least three concentrations spanning a conservative low point, a literature-centered point and a higher point only if the cell and material controls justify it. Include a no-initiator control where practical to detect dark curing or background polymerization.
  4. Measure optical and kinetic behavior. Determine gel time, working curve or cure depth, conversion and the minimum exposure that locks each layer. Check whether a photo-absorber changes penetration or creates an under-cured base.
  5. Print standardized test pieces. Use thin lattices, channels and overhangs as well as bulk coupons. Measure dimensional error, unsupported-feature collapse, layer adhesion and surface tack.
  6. Test cells during the real exposure. Assess viability immediately after printing, after washing or medium exchange, and at later time points. The Xu study shows why concentration and total printing time must be analyzed together: viability declined as both increased.
  7. Characterize the cured network. Measure swelling, degradation rate, elastic modulus, pore size and residual monomer or initiator. In the Xu comparison, Irgacure-cured samples had slightly larger pores, faster degradation and greater swelling despite similar post-crosslinking viability.
  8. Repeat with the intended post-cure. A rinse, secondary illumination or thermal step can change conversion and cell exposure. Use the same washing volume, duration, light dose and medium that the final protocol will use.
  9. Document reproducibility. Track batch, initiator purity, stock age, oxygen handling, temperature, cell passage, irradiance and software exposure settings. A setting that works once is not a process until it is repeatable.

How can I balance cure speed, resolution and cell viability?

Think of exposure as a budget shared by three outcomes: enough radicals to solidify each voxel, enough optical contrast to preserve features, and a low enough chemical and light dose to protect cells. Increasing PI concentration or exposure can shorten gel time, but it can also increase radical damage, conversion-induced shrinkage and heat. Reducing dose may preserve viability while causing tacky surfaces, layer delamination or loss of fine channels.

  • For higher resolution: reduce unintended light penetration with a validated photo-absorber, optimize exposure and keep the initiator uniformly dissolved. Verify that the absorber does not starve deeper layers of radicals.
  • For faster throughput: first improve irradiance and exposure uniformity, then adjust PI concentration in small increments. Faster gelation can alter conversion, swelling and feature fidelity, so remeasure the final network.
  • For fragile cells: test shorter exposures, lower radical-generating concentration and cooler processing, while checking whether incomplete cure leaves toxic residuals or unstable constructs.
  • For thick or highly scattering inks: measure cure depth through the actual cell-containing formulation. Nominal absorption data from a dilute solution may overestimate penetration.
  • For long prints: include cumulative exposure in the design. A concentration that is acceptable for a five-minute print may not be acceptable when the same cells remain under illumination for an hour.

What post-print properties must be checked?

Live/dead staining immediately after printing is only one endpoint. Initiator choice can change the network even when early viability appears similar. At minimum, compare:

  • Conversion and residual monomer or initiator after washing;
  • Equilibrium swelling and dimensional change;
  • Hydrolytic or enzymatic degradation and mass loss;
  • Elastic modulus in the hydrated state;
  • Pore size, interconnectivity and transport;
  • Surface chemistry and protein or cell adhesion;
  • Feature fidelity after swelling, degradation and any secondary cure;
  • Cell proliferation and phenotype over the intended culture period; and
  • Immune response in the relevant model, rather than inferring it from short-term viability.

The B2/TEOA PGSA study reported proliferation through 10 days and low immune responses at 14 days post-implantation under its specific formulation and model. Such time points are useful examples of the duration needed to reveal behavior that an immediate viability assay cannot capture.

What common problems indicate a poor initiator choice?

The resin stays tacky or layers separate

Check wavelength overlap, irradiance at the resin plane, oxygen inhibition, initiator dissolution, exposure time and cure depth. A photo-absorber or high cell and polymer loading may be attenuating light. Increase dose only after confirming that the initiator is actually absorbing and uniformly distributed.

The print cures, but channels close or dimensions are oversized

Excessive penetration or scattering can expose neighboring voxels. Lower exposure, reduce initiator concentration, adjust the photo-absorber or change layer thickness, then remeasure conversion so that improved resolution does not create an under-cured core.

Cells die during a long print

Separate concentration from cumulative exposure. Compare the same PI at shorter and longer print durations, record temperature and irradiance, and evaluate cells immediately and after washing. The high-concentration results in the 2020 GelMA comparison demonstrate that a condition tolerated at 0.3–0.5% (w/v) may not be tolerated at 0.7–0.9% (w/v).

Bubbles appear in the cured part

For VA-086, investigate nitrogen release first. Degas the precursor, minimize agitation and inspect the resin before exposure. Bubbles can also originate from mixing or cell handling, so include a resin-only control.

What are the safety and translation limits?

Published cell-viability and implantation results do not establish clinical approval for a commercial photoinitiator formulation. Purity, residuals, sterilization compatibility, endotoxin control, storage stability and manufacturing consistency require independent qualification. Treat every cited concentration and exposure as specific to its formulation, wavelength, dose, cell type and assay.

Before translational work, define an analytical method for residual initiator and unreacted monomer, validate washing and post-cure, and establish acceptance criteria for mechanical and biological performance. Keep research-use labeling distinct from clinical claims, and confirm the regulatory status of the exact chemical grade and complete formulation in the jurisdiction where it will be used.

Which decision rule should I use?

Use this order: wavelength fit, complete dissolution or dispersion, cell-dose window, cure kinetics, then long-term network performance. For a 405 nm aqueous hydrogel, LAP is the most practical starting screen; compare it with Na-TPO or another wavelength-matched option if the initial matrix fails. For a 365 nm setup, include Irgacure 2959 while explicitly controlling UV dose and solubility. Choose a different system only when its optical and biological trade-offs are demonstrated in the actual printer and bioink.

Frequently Asked Questions

Can I use the same photoinitiator concentration for every 3D bioprinter?

No. Delivered irradiance, emission spectrum, optics, layer thickness, resin absorbance and exposure software differ between machines. Recalibrate a concentration and exposure whenever the printer, formulation or cell type changes.

Does a water-soluble photoinitiator automatically make a bioink cell-safe?

No. Solubility prevents one formulation problem but does not remove radical, light or residual toxicity. Cell response must be measured at the combined concentration, irradiance and cumulative exposure used for printing.

Why might a sample with good immediate viability fail later?

Incomplete conversion, residual chemicals, altered swelling, fast degradation or unsuitable pore structure can impair cells after the first assay. Include washing controls and multi-day proliferation, phenotype and material testing.

Are visible-light photoinitiators always safer than ultraviolet initiators?

Not necessarily. Visible-light systems can reduce UV exposure, but type-II chemistry needs co-initiators and still creates radicals whose dose must be controlled. Compare the complete pair, wavelength and exposure rather than judging by color alone.
