Researchers can share an AI-designed protein more safely by treating release as a staged decision, not a choice between publishing everything and publishing nothing. First assess the sequence and the surrounding information—such as structure, function claims, expression context, protocols, optimization data, and metadata—then obtain appropriate institutional review and screen sequences and recipients when synthesis or transfer is involved. Choose the least restrictive release that still serves the scientific purpose, and document the decision so it can be revisited if the design or circumstances change.

What does it mean to share an AI-designed protein safely?

Safe sharing means enabling a legitimate scientific purpose while avoiding unnecessary disclosure of details that could make harmful synthesis, expression, evasion, or optimization easier. It is a risk-management process, not a guarantee that misuse is impossible. The appropriate balance depends on the design, what is already known, the likely recipients, the release channel, and the consequences of misuse.

A protein design is more than an amino-acid sequence. Its structure, claimed function, expression context, protocols, optimization data, troubleshooting information, and metadata can all affect how useful the package is to another person. A sequence alone may be insufficient to reproduce a result; combined with operational context, it may lower barriers to execution. Assess the complete information package rather than treating the sequence as the only sensitive asset.

The same distinction matters when considering screening. Much current policy and industry guidance concerns synthetic nucleic acid sequences and the providers who synthesize them. A protein sequence is not itself a nucleic acid order, but a design may be translated into a synthesis request or shared alongside information that supports one. Researchers should therefore identify where synthesis or transfer occurs and use the relevant screening and institutional processes at those points.

How should a research team assess a design before release?

Start with a written inventory and a plausible-misuse assessment. The goal is to identify which parts of the package are needed for independent scientific scrutiny and which would materially reduce the effort or expertise needed for a harmful application.

1. Inventory the information package

List the sequence, structure, function claims, expression system, protocols, optimization parameters, troubleshooting notes, and metadata. Include supplementary files, repository records, presentations, and information embedded in model or workflow outputs if they reveal operational details. Track versions: a revised design or added protocol can change the disclosure risk even if the project title stays the same.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

Separate information by purpose. For example, a reviewer may need enough evidence to evaluate a function claim, while an external reader may not need every optimization detail to understand the scientific result. Do not assume that removing one file makes a package safe if the same information remains in a figure, caption, repository history, or another attachment.

2. Consider plausible harms and enabling detail

Ask what harmful function could plausibly be associated with the design, how readily it could be synthesized or expressed, what expertise and equipment would be needed, whether the material could aid screening evasion or further optimization, and how serious the consequences might be. Record both the reasoning and uncertainties; a confident-looking score should not conceal gaps in what the team knows.

Assess the contribution of the whole package, not just whether a sequence resembles a known sequence of concern. A function claim, host information, or troubleshooting detail may alter practical usefulness. Conversely, a prediction or structural resemblance alone does not establish that a protein will have a particular activity. Keep claims proportionate to the evidence and avoid treating computational outputs as proof of biological function.

3. Bring in the right reviewers

Route the proposed release through the institution’s biosafety and dual-use review process, and involve legal, export-control, or information-security specialists when applicable. Identify a decision owner who can reconcile scientific value, safety concerns, and local policy. Requirements differ by country, institution, funder, and design, so this article is a governance aid rather than legal advice.

Review should happen early enough to affect the release plan, not only after a manuscript or dataset is already public. If the assessment identifies unresolved concerns, reviewers can help determine whether to narrow the package, restrict access temporarily, or seek additional expertise. Keep a record of the decision, its rationale, approvals, and any conditions attached to release.

Which release option should researchers choose?

There is no single release mode that fits every design. Compare the scientific benefit and reproducibility retained with misuse plausibility, consequence severity, reversibility, access controls, monitoring capability, and whether a delay would enable safer review.

Release mode When it may fit Main consideration
Open publication When review finds that broad disclosure is justified and the full package does not create an unacceptable added risk. Public disclosure is difficult to reverse; consider all associated files and metadata, not only the article text.
Controlled-access repository When legitimate researchers need access but unrestricted distribution is not necessary. Set clear eligibility and access rules, and ensure the institution can manage requests and keep appropriate records.
Vetted-recipient transfer When a specific collaboration or validation purpose calls for sharing with identifiable recipients. Verify recipient legitimacy and transfer only the information needed for the stated purpose.
Partial release When selected results can be shared while sensitive operational details are withheld or redacted. Check that the remaining material still supports meaningful scientific interpretation and does not expose the withheld detail elsewhere.
Delayed release When time is needed for review, screening, or a safer sharing arrangement. Define a review date and conditions for lifting the restriction; a delay should not become an undocumented indefinite hold.

These options can be combined. A paper might describe the scientific finding openly while a repository provides a vetted route to selected data. The design choice should follow the research purpose and documented assessment, rather than convenience or a blanket assumption that every AI-generated sequence must be either public or secret.

What should be screened before synthesis or transfer?

When synthesis or distribution is possible, screening should address both the sequence and the people or organizations involved. HHS guidance recommends sequence screening, customer and recipient legitimacy checks, responsible handling and transfer, follow-up on sequences of concern, and records. Researchers should use an appropriate provider or institutional process and determine who is responsible for each check before initiating an order or transfer.

Screening is not merely a single database lookup. NIST describes a broader approach involving standards, databases, tools, testing, and secure-transfer work to identify and track sequences of concern. In practice, researchers should establish how a concern is escalated, who can resolve it, and how the final disposition is recorded. Do not treat a screening result as a substitute for institutional review or recipient verification.

HHS’s synthetic nucleic acid screening page says its 2024 framework is being revised or replaced under the May 5, 2025 executive order. The Federal Register framework describes recommendations for provider screening, customer identity verification, follow-up, and documentation; check current official guidance and local procedures before relying on a particular version. The International Gene Synthesis Consortium lists a 2026 Harmonized Screening Protocol v3.1, another indication that relevant industry practices are updated over time. These sources describe guidance and protocols; they do not by themselves establish the legal requirements in every jurisdiction.

How much technical detail belongs in a paper or dataset?

Release enough detail to support the legitimate scientific purpose, but assess whether each operational element adds avoidable capability. This is a targeted review, not a rule to strip papers of reproducibility. The relevant question is whether a particular detail is necessary for independent evaluation or validation, and whether its benefit outweighs the added risk in the specific context.

  • Function claims: State what has been established and distinguish predictions from validated observations. Do not let a computational prediction imply demonstrated activity.
  • Protocols and troubleshooting: Review whether the exact operational detail is essential to the audience’s scientific purpose or would substantially lower the barrier to harmful execution.
  • Host and expression context: Consider whether including this information changes practical usability, especially when combined with sequence and procedural detail.
  • Optimization data: Check whether parameters or design iterations reveal how to improve a sensitive property or circumvent a safeguard.
  • Metadata and version history: Inspect repository records, supplementary files, and prior versions for details removed from the final manuscript but still accessible elsewhere.

If detail is withheld or redacted, explain the boundary in terms that preserve scientific clarity where possible. A controlled route for qualified access may be more useful than removing information without an alternative, but it requires a workable access process and ongoing recordkeeping. Reviewers should consider whether the public material still allows readers to understand the study’s scope, limitations, and basis for its claims.

What do recent evaluations say about AI-generated sequences and screening?

AI-generated designs make screening a moving-target problem, but recent evaluations do not support the claim that structural similarity alone establishes retained biological activity or reliable screening evasion. NIST’s 2026 program page describes 2025 work evaluating 30,000 enzyme variants and a dataset containing approximately 40,000 AI-generated enzyme variants assembled with Profluent Bio and Align Foundation. These are NIST-reported program figures, not a universal performance benchmark for all protein designs or screening systems.

NIST also reports an evaluation of synthetic homologs whose predicted structures were similar to native templates. Structural similarity did not necessarily preserve activity, and the evaluation did not demonstrate reliable rewriting that both maintained activity and evaded screening. The practical lesson is to avoid two opposite errors: assuming a structurally similar design is functionally equivalent, and assuming a screening system will catch every future design. Sequence screening, recipient checks, expert review, and careful disclosure decisions address different parts of the risk.

The Responsible Biodesign Workshop 2025 recommended next-generation screening algorithms for AI-generated sequences of concern and collaboration with synthesis providers. That recommendation points to continuing development, not a guarantee that current tools are comprehensive. Teams should keep screening within a broader process that can respond to concerns, document decisions, and reassess when designs or threat information change.

What records should a team keep, and when should it reassess?

Maintain a traceable record sufficient to explain what was shared, with whom, under what review, and why. HHS guidance calls for documentation of screening, follow-up, transfers, and final disposition. A practical record should include:

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.
  • The versioned inventory of the sequence and associated information considered for release.
  • The assessment rationale, uncertainties, and relevant approvals or conditions.
  • Screening results and any follow-up or final disposition when synthesis or transfer is involved.
  • The recipient list, access decisions, transfer details, and access logs where applicable.
  • The release version, review date for any temporary restriction, and an incident contact.

Reassess when the design changes, new protocols or metadata are added, the recipient or transfer route changes, or relevant threat information and guidance are updated. A decision made for one version or recipient should not automatically be treated as approval for a materially different package or transfer.

NIST’s biosecurity program page reports monthly baseline testing begun in August 2025 and describes collaborations with IBBIS, MITRE, and SecureDNA. Those details illustrate that screening standards and testing are active areas of work; they are not a reason to assume a particular provider’s process meets every institution’s needs. Teams should confirm the screening responsibilities, escalation path, and recordkeeping expectations that apply to their own release.

Frequently Asked Questions

Can a journal require a protein sequence to be deposited publicly?

Publication and data-sharing rules vary by journal, funder, and institution. If a deposit requirement conflicts with a documented safety concern, raise the issue with the relevant editor and institutional reviewers before submission rather than assuming a policy exception exists.

Does a predicted structure prove that an AI-designed protein works?

No. A predicted structure is evidence about a model’s structural prediction, not direct proof of biological activity. Claims should distinguish computational predictions from experimental validation.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

Who is responsible for checking a sequence before a synthesis order?

Responsibilities can be shared among the ordering institution, the researcher, and the synthesis provider, depending on the applicable process. Agree on who performs screening, handles a flagged order, verifies relevant parties, and retains records before placing the order.

How often should a team revisit a sharing decision?

Revisit it when a material change occurs, such as a modified design, added operational information, a new recipient, or updated threat guidance. For a temporary restriction, set a review date when it is imposed so the decision does not simply persist without reconsideration.
