What Is Electron Beam Imaging? How Electron Microscopy Works

Electron-beam imaging uses focused electrons to reveal a specimen’s surface or internal structure. Learn how SEM, TEM and STEM differ and what their detectors can show.

Electron-beam imaging uses a focused beam of electrons to create images of a specimen. Depending on whether the beam scans a surface or passes through a thin sample, electron microscopy can reveal surface texture, internal structure and—in combination with analytical detectors—elemental or bonding-related information.

What is electron microscopy?

Electron microscopy is a family of imaging methods that uses electrons rather than visible light to probe a specimen. An electron source produces and accelerates the beam in a vacuum; electromagnetic lenses and apertures shape and focus it. As the beam meets the specimen, electrons are transmitted, scattered or emitted. Detectors measure selected signals, and software maps them to image data.

The term “electron-beam imaging” describes this general approach, not one single microscope mode. The U.S. Department of Veterans Affairs’ overview describes typical accelerating energies of 5–100 keV, but that is a general range from its 2017 explanation, not a specification for every instrument. How does an electron microscope work?

  1. Generate and accelerate electrons. An electron source produces the beam, which is accelerated inside the instrument’s vacuum column.
  2. Focus and direct the beam. Electromagnetic lenses and apertures control the beam’s shape and path.
  3. Interact with the specimen. Depending on the imaging mode and sample, electrons pass through, scatter within, or emerge from the specimen.
  4. Collect a signal. Detectors record chosen transmitted, scattered or emitted electrons. The specific signal used affects what features stand out.
  5. Build the image. In scanning modes, the beam moves across the specimen point by point; software associates the detector signal at each position with image pixels. Arizona State University Eyring Materials Center: Electron microscopy techniques

What is the difference between SEM, TEM and STEM?

Mode How the beam is used Best suited to
SEM (scanning electron microscopy) A focused beam scans the specimen surface. Detectors can collect emitted signals, including secondary electrons, to form an image. Surface shape and texture; spatially resolved composition when suitable analytical detectors are available.
TEM (transmission electron microscopy) Electrons pass through an electron-transparent specimen to form an image or diffraction pattern. Internal structure, defects, and structural or compositional variation in thin specimens.
STEM (scanning transmission electron microscopy) A fine probe scans across a thin specimen. Detectors measure transmitted or scattered electrons at each position. Localized transmission contrast and analysis across a thin specimen.

These distinctions are about geometry and signal, not a simple ranking. STEM combines scanning with transmission: the probe raster-scans a thin sample, while detector geometry determines which scattered electrons contribute to the image. Bright-field, dark-field and high-angle annular dark-field modes therefore emphasize different scattering information. Indiana University and ETH Zurich describe this scanning and detector-based approach in their STEM resources: Indiana University Electron Microscopy Center: Scanning Transmission Electron Microscopy (STEM) and ETH Zurich: Scanning Transmission Electron Microscopy (STEM).

What can electron-beam imaging reveal?

  • Surface features: SEM can show topography and texture, and can support spatially resolved composition measurements when paired with suitable detectors.
  • Internal structure: TEM and STEM can examine features inside electron-transparent samples, including biological structures, crystals and defects.
  • Elemental information: Energy-dispersive X-ray spectroscopy (EDS, also called EDX) can identify elements in a specimen and, depending on the setup and measurement, map their distribution.
  • Composition and bonding clues: Electron energy-loss spectroscopy (EELS) can provide compositional information and additional insight into bonding or oxidation state.
  • Structure and three-dimensional information: Diffraction can reveal structural information, while tomography can be used to reconstruct three-dimensional features from multiple views.

These are possible techniques, not guaranteed outputs from every electron microscope. ASU’s facility overview describes biological TEM applications such as cell interiors and biological complexes, as well as materials applications including crystal defects, structure and composition. It also lists capabilities such as elemental mapping, diffraction and 3D imaging for particular facility instruments. ASU Eyring Materials Center: Electron microscopy techniques

What should you consider when choosing an imaging mode?

  • Choose SEM when the question is about a surface. It scans the surface and forms images from signals emitted or scattered there.
  • Choose TEM when you need a transmission image or diffraction pattern. The specimen must be thin enough for electrons to pass through.
  • Consider STEM for localized measurements across a thin specimen. Its scanning probe and selectable detector signals can provide spatially resolved transmission contrast or analytical data.
  • Match the detector to the question. Image contrast depends on which signal is collected and, in STEM, on detector geometry.

TEM and STEM require electron-transparent specimens, while conventional SEM is used to image specimen surfaces. Sample preparation and instrument configuration therefore matter as much as the microscope’s label. Resolution and analytical performance also depend on the specimen, instrument and operating conditions; atomic-resolution images or chemical maps are not automatic outcomes.
