Three Constraints That Make Origin-of-Life Chemistry Testable

The strongest origin-of-life theories do not ask only whether a reaction is chemically possible. They ask whether it fits early Earth, can happen without an experimenter, and leads toward chemistry recognizably related to life. A 2022 review by Malaterre, Jeancolas, and Nghe names these tests historical adequacy, natural spontaneity, and similarity to life-as-we-know-it.

Those constraints narrow an otherwise enormous search through possible molecules and environments. They also clarify why researchers sometimes relax a constraint: exploratory chemistry can reveal mechanisms and building blocks that are later tested against geological timing, realistic concentrations, and biological relevance. Matt Powner’s work on aqueous ligation of α-aminonitriles illustrates that strategy, examining whether peptide bonds could emerge from plausible amino-acid precursors rather than from modern laboratory assumptions.

What constraints should an origin-of-life theory satisfy?

“Origin of life” is not one reaction waiting to be discovered. It is a historical explanation for how nonliving chemistry on the young Earth could have produced systems capable of heredity, metabolism, compartmentalization, or evolution. A proposed pathway therefore has to answer two different questions: can the chemistry work, and does it belong in the world that actually existed?

The review The Origin of Life: What Is the Question? (Malaterre, Jeancolas, and Nghe, 2022) separates three dimensions that are often mixed together:

  • Historical adequacy: whether the pathway is compatible with the early Earth’s chronology, materials, environments, and geological history.
  • Natural spontaneity: whether the transition can proceed through naturally occurring processes rather than an experimenter’s intervention or an engineered supply of energy and reagents.
  • Similarity to life-as-we-know-it: whether the proposed chemistry resembles terrestrial biology in its molecules, information systems, metabolism, and environmental requirements.

These dimensions do not always point to the same experiment. A reaction can be highly plausible in water yet use compounds that were unavailable on early Earth. Another can fit a geological setting but require repeated purification, precise dosing, or a laboratory catalyst. A third can be entirely plausible for a hypothetical alien biology while offering little evidence about the ancestry of terrestrial life.

Why adding constraints helps

Without constraints, the search space is combinatorial: countless solvents, minerals, energy sources, temperatures, concentrations, molecular families, and reaction sequences can be combined. Most combinations are impossible to test in a human lifetime. Constraints remove options that cannot answer the historical question, making experiments more focused and their results easier to interpret.

Constraints are not a demand that every exploratory experiment reproduce a complete early-Earth scene. They are filters applied according to the question being asked. A fact-establishing experiment may deliberately relax one dimension to determine whether a reaction class is possible at all. An explanation-seeking experiment must eventually restore the relevant historical and environmental conditions.

Historical adequacy: could this have happened on early Earth?

Historical adequacy asks whether a proposed pathway belongs to the interval between Earth’s formation and the earliest evidence for life. The review summarizes initial planetary accretion at approximately 4.56 billion years ago and the earliest undisputed traces of living cells at approximately 3.42 billion years ago. A chemical scenario must fit somewhere within that broad window, while respecting what is known—or still uncertain—about the planet’s surface and atmosphere.

The chronology test

A pathway should specify when each stage could occur. If a mineral, atmospheric gas, or ocean condition appeared only after the proposed reaction, that sequence fails historically even if the reaction works in a flask. Conversely, uncertainty in the early geological record can justify a range of possible settings, but not an unlimited choice of conditions.

The materials test

Every reactant needs a plausible source. Researchers ask whether a compound could form geologically, arrive through atmospheric or extraterrestrial chemistry, or accumulate in a local environment. “Plausible” does not mean abundant everywhere; a transient pool, mineral surface, wet-dry margin, or protected pore may be enough if the pathway can operate there before the material is destroyed or diluted.

The concentration and stability test

Many laboratory demonstrations use purified reagents at concentrations far above those expected in an open ocean. Historical adequacy therefore requires attention to dilution, competing reactions, hydrolysis, temperature, pH, and the lifetime of intermediates. A compound that survives for minutes under controlled conditions may not support a multi-step pathway in nature. Local concentration mechanisms—evaporation, freezing, adsorption, or flow through pores—must be part of the explanation rather than an invisible laboratory convenience.

The setting test

“Early Earth” was not a single reactor. Ocean water, volcanic land, hydrothermal systems, impact-generated environments, mineral surfaces, and freshwater basins would have imposed different chemical constraints. A credible account identifies the setting that supplies reactants and energy, removes waste, and protects useful products. It also states which atmospheric and ocean assumptions are required, because those conditions remain an active area of geological interpretation.

Natural spontaneity: can nature do the work?

Natural spontaneity is stricter than showing that a reaction can be made to occur. In a laboratory, a chemist may add a purified catalyst, adjust pH between steps, exclude oxygen, remove products, illuminate a sample, or replenish a reagent. Those interventions can establish chemical capability, but they are not themselves evidence that the same transition occurred without an agent.

What counts as intervention?

An experiment can still be relevant while using intervention. The key is to distinguish the intervention that reveals a mechanism from the intervention that supplies the mechanism. For example, preparing a starting material in advance may be reasonable for a first test of downstream reactivity. Claiming a historical pathway, however, requires showing how that material could arise and persist naturally.

Spontaneity also includes energy and cycling. A process may be naturally driven by sunlight, geothermal gradients, redox differences, wet-dry cycles, freezing, or mineral catalysis. It should not depend on an unreported energy source or on a sequence of manual transfers that nature has no obvious way to perform.

Why negative results matter

When a reaction fails under plausible conditions, that result can eliminate an attractive but unrealistic route. Failure may reveal that an intermediate hydrolyzes too quickly, that a side reaction dominates, or that the required concentration cannot be achieved. Such constraints prevent researchers from building long pathways out of individually impressive but mutually incompatible steps.

Similarity to life-as-we-know-it: how much biology must be reproduced?

The third dimension asks how closely the target resembles terrestrial life. Biology on Earth uses a familiar set of molecular classes, genetic polymers, amino acids, lipids, cofactors, and energy currencies. A theory that produces these or closely related systems has a direct connection to our ancestry. A theory based on alternative genetic materials, metabolisms, solvents, or habitats may still demonstrate that life-like organization is chemically possible, but it addresses a broader question than the specific origin of terrestrial life.

When relaxing similarity is useful

Researchers may relax the life-similarity constraint to explore chemical principles that biology does not currently use. Alternative polymers can reveal which features are necessary for information storage. Unfamiliar catalytic cycles can show how energy coupling might work without modern enzymes. These results can guide the search for plausible ancestral chemistry even when the final system must converge on terrestrial biochemistry.

The danger of changing the question

A relaxed model should be labeled as exploratory rather than presented as a reconstruction of Earth’s history. Demonstrating a self-organizing chemical network in an unusual solvent does not establish that such a network preceded terrestrial cells. It establishes a possibility that may later be tested against geological and biological constraints.

Could peptides form in water before biology?

Peptides are chains of amino acids linked by amide bonds. Modern biology makes peptide synthesis look routine because ribosomes, enzymes, activated substrates, and controlled cellular compartments handle the difficult chemistry. Before biology, none of those systems could be assumed. The central question is whether peptide-like products could arise in water from compounds that early-Earth chemistry could plausibly supply.

The aminonitrile approach

In 2019, Matt Powner’s group reported aqueous ligation of α-aminonitriles as a route toward peptides under prebiotically plausible conditions. Aminonitriles are accepted abiotic precursors related to amino-acid chemistry. Instead of beginning with the activated reagents commonly chosen for modern synthetic chemistry, the work examined whether these plausible precursors could participate in bond-forming reactions in water.

The significance is methodological as much as chemical. The experiment connects a building-block family to a larger molecular class—peptides—while keeping the solvent and starting materials within a prebiotic frame. It does not by itself establish that peptides were the first informational or catalytic polymers, nor that the tested conditions describe every early-Earth environment. It shows that peptide-forming reactivity can be investigated without assuming biological machinery.

What a realistic peptide claim must still establish

  • How α-aminonitriles could be generated and supplied in the relevant environment.
  • Whether their concentrations and lifetimes are compatible with repeated ligation.
  • Which side reactions compete in water and whether useful products remain stable.
  • Whether the products accumulate, participate in catalysis, or influence compartment formation rather than appearing only as trace analytical signals.
  • How the chemistry fits a specific geological setting and sequence of events.

These questions illustrate why “peptides form in water” is an incomplete conclusion. The chemically important result is a route worth testing under increasingly demanding constraints; the historical conclusion requires the additional environmental evidence.

How do scientists know a prebiotic reaction was realistic?

Realism is assessed by comparing a reaction with five practical axes. No single axis decides the origin question, but together they expose where a proposal is strong and where it needs further work.

Axis Question to ask Warning sign
Early-Earth chronology and geology Could the materials and setting exist between Earth’s formation and the earliest cells? The pathway requires a mineral, atmosphere, or environment with no historical basis.
Spontaneity Can naturally available energy and cycles drive the transition? Manual reagent additions, purification, or pH changes are essential but unexplained.
Similarity to terrestrial life Does the product connect to the molecular systems used by Earth life? The result answers a general life-possibility question but is presented as Earth’s history.
Aqueous chemical plausibility Do water, concentration, stability, and competing reactions permit the process? A high-concentration, purified demonstration is treated as an ocean-scale event.
Experimental tractability Can the mechanism and products be measured in controlled, repeatable tests? The proposed network is so complex that no step can be isolated or falsified.

This framework helps compare experiments without pretending that every proposal has the same goal. A study may score highly on tractability while intentionally relaxing historical adequacy. Another may use a geologically realistic setting but produce products too poorly characterized to support a strong mechanistic claim.

Does origin-of-life research have to recreate early Earth exactly?

No. Exact reconstruction is impossible because the early Earth was heterogeneous and incompletely known. The useful standard is conditional fidelity: an experiment should reproduce the features that matter for its claim and disclose which features it leaves out.

If the claim is “this reaction is chemically possible in water,” an aqueous test with defined reagents may be appropriate. If the claim is “this pathway could have operated on early Earth,” the study must add geological sourcing, realistic concentrations, environmental cycling, and a natural route to each starting material. If the claim is “this chemistry could lead toward terrestrial life,” the products must be connected to life’s molecular systems rather than merely to generic self-organization.

Why relax constraints if the goal is historical origin?

Relaxation is a controlled research strategy, not a retreat from history. A fully constrained model can be too complicated to diagnose: if a multi-component experiment fails, researchers may not know which assumption caused the failure. By relaxing one dimension at a time, they can establish reaction mechanisms, identify useful intermediates, and determine which measurements are feasible.

The next step is to reintroduce constraints deliberately. A reaction first demonstrated with purified α-aminonitriles can be tested with plausible precursor-generation chemistry. A pathway first studied at high concentration can be examined under dilution, cycling, or mineral contact. An alternative polymer can be compared with the stability and copying requirements of terrestrial genetic systems. Each added constraint turns a broad possibility into a sharper historical test.

A practical checklist for evaluating a new origin-of-life claim

  1. Define the question. Is the study testing chemical possibility, a prebiotic mechanism, or a historical explanation?
  2. Map the timeline. Identify when Earth, the proposed environment, each reactant, and the earliest cells existed.
  3. Inventory the inputs. For every reagent, state its natural source, expected concentration, stability, and transport mechanism.
  4. Identify interventions. Separate laboratory conveniences from processes that nature could plausibly provide.
  5. Specify the setting. Name the water body, mineral surface, atmospheric condition, temperature range, and cycling process being modeled.
  6. Measure products and losses. Quantify useful products alongside degradation and side reactions.
  7. Connect to biology carefully. Explain whether the result bears on terrestrial molecules or only on life-like chemistry in general.
  8. Restore constraints. After a relaxed demonstration, test the mechanism under the historical conditions relevant to the claimed scenario.

Matt Powner captures the reason for this systems approach: “Understanding the origins of life is obviously a huge problem that implicates lots of different science.” He also notes that thinking across molecule classes “forces you to think about more chemistry” and encourages researchers to diversify both their questions and the molecules they study. That breadth is most useful when paired with explicit limits.
