Proposed Alternative Nutraceutical Delivery Methods: Which Format Fits Which Nutrient?

The most credible alternatives to conventional nutraceutical capsules are oral thin films, liposomes and other lipid nanoparticles, and transdermal systems such as patches or microneedles. None is universally superior: performance depends on the nutrient’s dose, solubility, stability, target tissue, formulation and human evidence. Reviews published in 2023–2024 describe genuine formulation advantages, but they do not prove that every commercial strip, liposomal drink, nanoparticle or patch produces better health outcomes than a conventional product.

What “alternative nutraceutical delivery” means

Nutraceutical delivery is the engineering of how a vitamin, mineral, botanical compound, antioxidant or other bioactive reaches the body. The familiar route is a tablet or capsule that disintegrates in the stomach and intestine. Alternative delivery research tries to solve specific weaknesses in that route: poor water solubility, limited intestinal permeability, chemical breakdown from acid or enzymes, first-pass metabolism in the liver, unpleasant taste, swallowing difficulty or the need for a controlled release profile.

As an Amazon Associate I earn from qualifying purchases.

The proposed platforms are not interchangeable. An oral thin film still delivers a substance through the mouth, but it can dissolve on the tongue or against the buccal or sublingual mucosa. A liposome or lipid nanoparticle is a carrier that may be swallowed or used in another route. A transdermal patch attempts to move an ingredient across the skin into local tissue or the bloodstream. Nanoformulations also include nasal, inhaled and ocular routes, each with different anatomical and regulatory requirements.

Reviews from 2023 and 2024 consistently describe platform-level benefits rather than a single winning product. The right question is therefore not “Which format absorbs best?” but “Which delivery problem does this nutrient have, and does the proposed formulation solve it in a validated model and at a practical dose?”

At-a-glance comparison of the main proposals

Delivery approach How it is used Problems it may address Main limitations
Oral thin film Ultra-thin sheet placed on the tongue or buccal/sublingual mucosa Swallowing difficulty, portability, rapid disintegration, flexible dosing and possible reduction of first-pass exposure Limited film capacity, moisture and stability concerns, taste masking, dose-uniformity and scale-up challenges
Liposome Vesicle with lipid bilayers carrying hydrophilic, lipophilic or amphiphilic ingredients; usually swallowed in a food, liquid or capsule Protection from degradation, improved dispersion of poorly soluble compounds and potential changes in bioaccessibility Stability, leakage, loading efficiency, manufacturing cost and uncertain translation from laboratory models to human outcomes
Solid lipid nanoparticle (SLN) Nanoparticle with a solid lipid core Protection of antioxidant nutraceuticals and potential improvement in solubility, permeability and gastrointestinal stability Limited drug-loading space for some molecules, possible expulsion during storage and formulation-specific safety questions
Nanostructured lipid carrier (NLC) Particle made from a blend of solid and liquid lipids Greater loading flexibility and protection for selected poorly soluble compounds Particle stability, oxidation, scale-up and the need to validate each lipid mixture
Transdermal system Patch, gel, vesicle, microneedle or assisted device applied to skin Avoidance of gastrointestinal degradation and possible reduction of oral first-pass metabolism The skin is a strong barrier; molecule size, lipophilicity, dose and test model determine whether meaningful systemic delivery is possible
Other nanoformulations Nanosuspensions, microemulsions, micelles, nanocapsules or nanoliposomes delivered by oral, nasal, inhaled, transdermal or ocular routes Solubility, dispersion, protection and route-specific release control Complex manufacturing, route-specific tolerability, limited human evidence and regulatory characterization requirements

Oral thin films: the most practical non-pill concept

How a film delivers a nutrient

An oral thin film is a very thin polymeric sheet containing the active ingredient and excipients. It is placed on the tongue or against the cheek, under the tongue or another area of oral mucosa. Saliva hydrates the sheet, the matrix disintegrates, and the ingredient is swallowed or absorbed through the oral lining. The exact balance between mucosal absorption and gastrointestinal absorption depends on the ingredient, film design, contact time and how much saliva is produced.

A 2024 Biotechnology Advances review called oral thin films “innovative dosage forms that have gained tremendous attention for the delivery of nutraceuticals.” The attraction is practical as well as pharmacokinetic: a film is light, discreet and usable without water. Rapid disintegration can help people who have difficulty swallowing tablets, although rapid disappearance does not by itself prove greater systemic absorption.

Where films can help

  • Portability: Individual films can be carried without a bottle of liquid and may suit travel or on-the-go use.
  • Flexible dosing: Manufacturers can vary the amount per sheet or, in some designs, combine layers with different release behavior.
  • Potential first-pass bypass: Ingredient absorbed through oral mucosa can partially avoid gastrointestinal transit and hepatic first-pass metabolism. The proportion that follows this route must be measured for the specific formulation.
  • Acceptability: A dissolving sheet can be easier to take than a large tablet, provided its taste, mouthfeel and residue are acceptable.

What limits an oral film

Films have finite surface area and thickness, so they are better suited to potent ingredients or modest doses than to bulky mineral doses. Hygroscopic ingredients can draw in moisture and weaken the sheet. Taste masking is a formulation problem for bitter botanicals and minerals. Uniform distribution of the active must be demonstrated across production batches, and packaging must protect the film from humidity. The 2024 review literature emphasizes formulation and quality-by-design work because a laboratory sheet is not automatically a manufacturable, dose-accurate product.

Consumers should distinguish a research prototype from a marketed “vitamin strip.” A commercial film should identify the amount of each active per strip, serving instructions, storage conditions, lot information and the jurisdiction’s required labeling. “Dissolves quickly” describes a physical property, not a clinical outcome.

Liposomes, solid lipid nanoparticles and nanostructured lipid carriers

Liposomes

Liposomes are closed vesicles made from lipid bilayers. Their structure can accommodate water-soluble compounds in an aqueous interior, fat-soluble compounds within the bilayer and amphiphilic compounds at the interface. That versatility explains why vitamins are frequent targets in food and cosmetic research.

A liposome may protect a sensitive nutrient from environmental conditions or alter how it disperses in the gastrointestinal tract. However, encapsulation does not guarantee that the nutrient reaches the bloodstream intact. Researchers must measure loading efficiency, leakage during storage, digestion behavior and bioaccessibility—the fraction released in a form available for absorption. A systematic review of vitamin liposomes identifies scale-up expense, stability, loading and bioaccessibility as persistent challenges.

SLNs and NLCs

Solid lipid nanoparticles use a solid lipid matrix to carry a compound. Nanostructured lipid carriers combine solid and liquid lipids, creating a less-ordered matrix that can provide additional room for some actives. Reviews describe both as ways to address low solubility, poor permeability, pH or enzyme degradation and gastrointestinal instability, especially for antioxidant nutraceuticals.

The benefit is formulation-specific. Changing the lipid type, particle size, surfactant, active concentration or storage temperature can alter release and digestion. A particle that improves apparent solubility in a laboratory medium may not improve absorption in people. Oxidation of the lipid phase, aggregation, active expulsion during storage and differences between simulated digestion and human digestion all require testing.

How the lipid systems differ

Characteristic Liposome SLN NLC
Basic structure One or more phospholipid bilayers around an aqueous compartment Solid lipid matrix Mixture of solid and liquid lipids
Suitable cargo Hydrophilic, lipophilic and amphiphilic ingredients Selected lipophilic or compatible ingredients Selected ingredients needing more flexible matrix loading
Potential advantage Versatile compartmentalization and protection Solid matrix protection and controlled release potential Less-crystalline matrix can increase loading flexibility
Key development question Does the vesicle remain intact or release the nutrient at the intended stage? Does storage cause crystallization or active expulsion? Do lipid blending and liquid-lipid content remain stable over time?

Can nutrients be delivered through the skin?

Yes, but “can be applied to skin” and “can deliver an effective systemic dose through skin” are different claims. The outer stratum corneum is designed to block molecular passage. A nutrient must have suitable size, lipophilicity, concentration and stability, and the formulation must keep it in contact with skin long enough to cross. Results also vary between intact human skin, damaged skin, animal skin, reconstructed skin and cell experiments.

An Annual Reviews discussion of transdermal strategies notes that oral feeding can expose natural molecules to gastrointestinal degradation and presents transdermal delivery as a possible alternative. That rationale supports research; it does not establish that a particular nutrient patch works in humans.

Proposed transdermal carriers and enhancers

Technology Purpose in a transdermal design Important qualification
Liposomes Flexible lipid vesicles intended to interact with or carry cargo toward the skin Skin penetration depends on vesicle composition and the active ingredient
Ethosomes and transethosomes More deformable or alcohol-containing vesicles designed to improve passage through the skin barrier Alcohol, deformability and excipients can affect irritation and stability
Transfersomes Highly elastic vesicles proposed to squeeze through skin pathways Performance is formulation- and model-dependent
SLNs and NLCs Lipid nanoparticles used for skin residence, protection or penetration Particle deposition does not necessarily equal bloodstream delivery
Nanoemulsions Fine oil-and-water dispersions that can improve spreading and solubilization Small droplets alone do not overcome every barrier or prove systemic exposure
Dendrimers Branched nanoscale polymers that can bind or carry selected compounds Material safety and clearance must be established for each design
Microneedles Arrays that create microscopic pathways through the stratum corneum Needle geometry, insertion, pain, sterility and dose capacity are critical
Ultrasound Acoustic energy used to temporarily increase skin permeability Requires controlled equipment and validated energy settings
Iontophoresis Low electrical current used to assist movement of charged or suitable molecules Skin tolerance, electrode design and current control matter
Jet injectors High-velocity liquid stream used to breach the skin without a conventional needle Device regulation, dose control and user training become part of the product

Why nutrient patches often overpromise

Many vitamins and botanical compounds require doses that are difficult to move through intact skin. A patch can contain an ingredient and still deliver little beyond the surface. A credible claim should state whether evidence comes from an in-vitro diffusion cell, ex-vivo human skin, an animal study or a controlled human trial, and should report the delivered dose rather than only the patch’s total content. Skin redness, itching, sensitization and adhesive reactions are practical safety issues even when systemic absorption is low.

Other nanoformulations and routes

Reviews also discuss nanosuspensions, microemulsions, nanoliposomes, micelles and nanocapsules. These are formulation architectures rather than guarantees of superior absorption. A nanosuspension reduces particle size for a poorly soluble solid. A microemulsion uses carefully selected oil, water, surfactant and cosurfactant components. A micelle can solubilize hydrophobic compounds, while a nanocapsule surrounds cargo with a polymeric or lipid shell.

Researchers have proposed these systems for oral, nasal, inhalational, transdermal and ocular delivery. Each route changes the design brief:

  • Nasal: The formulation must remain in contact with nasal mucosa without causing unacceptable irritation or rapid clearance.
  • Inhaled: Aerodynamic particle size, device performance, lung deposition and pulmonary safety are central, so an oral supplement cannot simply be repackaged for inhalation.
  • Ocular: Sterility, preservative choice, residence time and protection of the eye take priority over ordinary supplement convenience.
  • Oral: Digestion, bile, mucus, intestinal permeability and microbiome interactions can change the carrier before absorption.

Do liposomal vitamins or oral strips absorb better?

Sometimes a formulation can increase measured bioaccessibility or exposure, but the evidence does not support a universal superiority claim. Liposomal and nanoparticle studies often begin with physicochemical or simulated-digestion tests, then progress—if successful—to cell, animal or limited human studies. Oral-film studies may show faster disintegration or a different absorption pathway without demonstrating a better long-term nutritional outcome.

“Absorption” also has several meanings. A formulation may dissolve more readily, survive digestion, cross an artificial membrane, raise a blood concentration or improve a tissue level. Those endpoints are not equivalent. A higher short-term blood concentration is not automatically a better clinical result, and a nutrient with an already adequate status may provide no additional benefit from increased exposure.

When evaluating a claim, look for five details:

  1. The exact active and dose: Vitamin C, vitamin D, a carotenoid and a botanical extract have different solubility and stability problems.
  2. The comparator: The meaningful comparison is the same active and dose in a conventional formulation, not an unrelated product.
  3. The study model: Human randomized trials provide a different level of evidence from simulated digestion or animal work.
  4. The measured endpoint: Bioaccessibility, pharmacokinetics, tissue concentration and clinical outcomes answer different questions.
  5. The formulation identity: Results for one carrier, particle size and excipient system cannot automatically be transferred to another brand or batch.

  1. Define the target site. Decide whether the goal is local mouth, stomach, intestine, skin, bloodstream, nasal tissue, lung or eye exposure. A local effect may not require systemic delivery.
  2. Determine the required dose. High-dose minerals and bulky extracts may exceed the practical capacity of a thin film or patch. A potent compound may be easier to formulate in either format.
  3. Characterize the molecule. Record water solubility, lipophilicity, molecular size, pH sensitivity, oxidation risk and susceptibility to enzymes. These properties determine whether a lipid carrier, film or skin strategy addresses a real bottleneck.
  4. Choose the protection mechanism. Use a carrier when the principal problem is degradation or poor dispersion; consider a mucosal route when swallowing or first-pass metabolism is the issue; consider a skin system only when penetration data support it.
  5. Check dose accuracy and release. Ask how much active is present per unit, how uniform units are, how release was measured and how storage affects potency.
  6. Rank evidence by model. Separate in-vitro, ex-vivo, animal and human evidence. A review-level platform advantage is not proof for a particular supplement.
  7. Assess tolerability. Check oral irritation and taste for films, gastrointestinal effects for lipid systems, and adhesive or sensitization reactions for patches. Device-assisted routes add procedural risks.
  8. Verify manufacturing and regulation. Nanoparticle size distribution, encapsulation, residual solvents, microbial quality, sterility where relevant and stability data must be controlled. Regulatory classification and permitted claims differ by jurisdiction.
  9. Match the format to adherence. A theoretically efficient product that a person dislikes, forgets or cannot store correctly may deliver less nutrient in practice than a conventional product used consistently.

Manufacturing and quality controls that matter

Alternative delivery systems add quality attributes beyond the label amount. A lipid product may require particle-size distribution, polydispersity, surface charge, encapsulation efficiency, leakage testing, oxidation monitoring and digestion or release studies. A thin film requires thickness, tensile strength, disintegration time, moisture content, content uniformity and packaging protection. A transdermal device requires adhesive performance, dose delivery across skin, irritation testing and, for microneedles or injectors, device consistency and sterility controls.

Quality-by-design approaches map critical material attributes and process parameters to the finished product’s performance. This is particularly important when changing from laboratory equipment to commercial production: mixing energy, drying conditions, lipid sources, polymer grades and packaging can all change the final exposure profile. “Nano” on a label is not a substitute for a specification or stability study.

Safety, regulation and realistic consumer expectations

Safety must be evaluated for the complete formulation, not only the vitamin or botanical. Lipids, surfactants, polymers, alcohols, preservatives, adhesives and device materials can alter tolerability. Nanomaterials may behave differently from the same substance in bulk form, so exposure, distribution and clearance deserve route-specific assessment.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

Regulatory status also varies by country and by whether a product is marketed as a food, dietary supplement, cosmetic, drug or medical device. A supplement claim about supporting normal nutrition is not equivalent to a disease-treatment claim. Transdermal systems and device-assisted delivery can trigger requirements that do not apply to an ordinary oral supplement. Consumers should look for a complete ingredient panel, serving dose, manufacturer contact information, storage instructions, lot or batch information and warnings relevant to the route.

People who are pregnant, taking medicines, managing kidney or liver disease, or using concentrated botanicals should discuss a new high-exposure formulation with a qualified clinician. Bypassing digestion or increasing exposure can change interaction and toxicity considerations; convenience does not remove those risks.

Where the field is heading

The 2023–2024 review literature points toward combinations of technologies rather than one universal replacement for pills: a film carrying a protected compound, a lipid carrier embedded in an oral matrix, or a transdermal system paired with microneedles or controlled energy. Progress will depend on reproducible manufacturing, clear dose measurement and human studies that connect exposure to meaningful nutritional or clinical outcomes.

At present, oral thin films and oral lipid carriers are the most straightforward proposals for improving convenience or protecting selected ingredients. Transdermal delivery remains promising for molecules that can cross skin or for systems that temporarily bypass the barrier, but it demands the strongest ingredient- and device-specific proof. The evidence base remains heterogeneous, spanning formulation experiments, cell and tissue models, animals and a smaller number of human studies; no single format has established across-the-board superiority for nutraceuticals.
