How Scientists Recover Ancient DNA from Cave Walls Without Damaging the Site

Scientists can recover traces of ancient DNA from decorated cave walls by removing extremely small, carefully chosen samples under archaeological permits and conservation controls. The method is designed for minimal impact, not zero impact: a minute amount of calcite, pigment or wall material is taken away, then processed in an ancient-DNA clean room. A 2026 study shows that human mitochondrial DNA can survive on cave walls for thousands of years, but recovery is uncommon and the DNA cannot, by itself, identify the people who made nearby art.

What the 2026 cave-wall study found

The First Art study examined 24 rock-art panels in 11 caves on the Iberian Peninsula. Researchers collected 120 samples from several kinds of context, including pigmented surfaces, unpigmented wall areas and comparative materials. Only five samples produced authentic ancient human mitochondrial DNA.

One positive sample came from a pigmented calcite crust at Escoural. Four came from unpigmented wall samples at Escoural and Covarón. This distribution matters: DNA was not recovered routinely from every painted surface, and a positive result did not require visible pigment.

The Max Planck Society summary of the study reports the same headline result: five of 120 samples yielded authentic ancient human mitochondrial DNA. The small proportion is a warning against treating cave-wall DNA recovery as a guaranteed test that can be applied to every panel or cave.

Alba Bossoms Mesa, the study’s first author, described the significance precisely: “Although we cannot directly connect the traces of ancient human DNA we have found to the creation of rock art, this is the first evidence for human DNA preservation on cave walls for thousands of years”.

Why “without damaging the site” needs a qualification

Sampling a wall is different from photographing it. Researchers do remove material, but they plan the intervention around the site’s conservation rules and take the smallest practical amount from an approved location. Permit-granting authorities can restrict which panel is sampled, where on that panel a sample may be taken and how the work is documented.

The study’s authors characterize the impact as minimal and describe site-specific controls. That language should not be translated into “non-invasive.” A DNA result requires physical contact with the wall and the removal of a tiny sample. The conservation achievement is reducing the intervention while obtaining information that cannot be gained from a purely contact-free survey.

Researchers also distinguish the sampling area from the visible artwork. An unpigmented portion of wall may be selected so that a painted outline or recognizable feature is not disturbed. In other cases, a naturally occurring calcite crust associated with pigment may be the approved target. The decision is made with conservators and authorities rather than by applying one universal sampling pattern to every cave.

How the workflow minimizes risk

The procedure combines archaeological planning with methods developed for highly contaminated ancient-DNA work. Each stage addresses a different risk: physical alteration of the panel, introduction of modern DNA, or misinterpretation of molecules that are too damaged to authenticate.

  1. Map the panel and define an approved target. Researchers document the art and surrounding surface, then adapt a sampling plan to the cave, panel and permit conditions. Pigmented and unpigmented contexts can both be considered, along with associated sediments or artefacts when the research design calls for them.
  2. Take a minute sample under conservation controls. Personnel use protective equipment and follow the restrictions set by the relevant authorities. The objective is to remove as little material as possible while retaining enough for laboratory analysis. The exact amount and tool choice depend on the wall material and the approved site plan.
  3. Separate ancient-DNA work from ordinary laboratory traffic. Ancient DNA is present in very small quantities, while modern human DNA is abundant. Processing therefore takes place in a dedicated clean room with contamination precautions. Protective clothing, controlled handling and procedural controls help prevent a worker’s DNA from overwhelming the sample.
  4. Release DNA from the mineral or pigment matrix. The study used silica-based extraction methods optimized for ancient DNA. Such methods are intended to recover short, chemically damaged fragments that may be trapped in calcite, pigment residues or porous wall material.
  5. Sequence the recovered fragments. Researchers sequence DNA extracts and use targeted enrichment to increase the chance of detecting human and other mammalian sequences when the total DNA is sparse. Enrichment does not create DNA that is absent; it helps identify relevant fragments in a mixture.
  6. Authenticate an ancient signal. A reported result must be consistent with characteristic ancient-DNA damage and other authenticity checks, while controls are examined for modern contamination. The conclusion is therefore based on a pattern of evidence, not merely on finding a human sequence.

Matthias Meyer, the study’s senior author, said the findings “fundamentally change how we think about where ancient DNA can be found.” The statement refers to preservation in an unexpected mineral surface, not to a new permission to sample every decorated wall.

Where the DNA was found

Context Location Result Interpretation supported by the study
Pigmented calcite crust Escoural Ancient human mitochondrial DNA detected Human DNA can persist in a pigment-associated mineral crust, but the source person’s role in making the art is not established.
Unpigmented wall Escoural Ancient human DNA detected in two samples; no detectable faunal DNA in those two samples The absence of detected faunal DNA is consistent with direct human contact, although it is not proof of contact during painting.
Unpigmented wall Covarón Ancient human DNA detected in two samples, with human and faunal DNA in the broader set of unpigmented contexts Mixed DNA is compatible with indirect movement through sediment or water as well as contact.
Airbrush pigment from a bird bone Altamira No ancient human DNA signal detected A plausible saliva-related source produced a negative result; the sampled amount was very small and modern contamination complicates interpretation.

The table shows why context matters more than a simple positive-versus-negative count. A calcite crust, a bare wall and an artefact can preserve different amounts of DNA and can receive molecules through different routes.

How DNA can reach and remain on a cave wall

Direct contact

People can leave cells and other biological material when they touch a surface. The two Escoural samples with human DNA but no detectable faunal DNA are consistent with this route. “Consistent with” is deliberately narrower than “proves”: the evidence does not establish when the contact occurred or why the person touched the wall.

Indirect transfer

DNA can also move with sediment, droplets or water. Some unpigmented samples contained both human and faunal DNA, a combination the authors discuss as compatible with environmental transfer. A mixture can therefore record activity in the cave without representing the hand, breath or saliva of an artist.

Mineral preservation

Calcite can form a crust over or near pigment and may trap biological molecules as it develops. The Escoural pigment-associated result demonstrates that a mineral surface can preserve ancient human DNA, but it does not show that the DNA and pigment were deposited at the same moment.

What the genetic evidence cannot identify

A DNA fragment from a wall is not a signature of the person who painted, engraved or blew pigment there. Someone might have entered the cave centuries later, touched the same surface, or contributed DNA that arrived through sediment or water. The authors explicitly caution that the DNA and the art may have been deposited thousands of years apart.

Nor does mitochondrial DNA provide a named individual. Mitochondrial genomes are inherited through the maternal line and can help characterize a biological lineage, but a cave-wall fragment is usually too limited and too environmentally displaced to assign authorship. The study demonstrates preservation and possible pathways of transfer, not an identification of an artist or a census of cave visitors.

How old are the recovered molecules?

The paper estimates minimum ages of about 1,000 years for one Covarón sample and about 2,000 years for the other positive samples. These are damage-pattern-based estimates, not direct measurements of the moment when DNA was deposited. The authors note that substantially older ages remain possible.

That distinction is essential. Ancient-DNA damage can indicate that molecules are old relative to modern contamination, but it does not automatically date a wall’s pigment layer, the formation of a calcite crust or the act of touching the surface. Establishing the age of the artwork requires independent archaeological or geological evidence.

Why the Altamira airbrush result was negative

An airbrush made from a bird bone at Altamira was a plausible candidate for saliva-derived DNA. However, pigment sampled from it yielded no ancient human DNA signal. The negative result does not demonstrate that no one ever used the tool or that saliva cannot survive on archaeological objects.

Only a very small amount was available for sampling, and modern contamination can make a weak ancient signal impossible to interpret. Ancient-DNA experiments therefore treat a negative extract as a result for that sample and method, not as proof that the object never carried DNA.

Contact-free cave imaging is a different method

Some cave programs document decorated surfaces without touching them. The French Ministry of Culture’s Chauvet program, for example, describes surveys based on photography and image processing. These techniques can record color, shape and deterioration while avoiding contact with decorated walls.

Contact-free imaging and DNA sampling answer different questions. Imaging documents what is visible on the surface; it cannot collect molecules trapped in calcite or wall pores. DNA sampling can reveal biological traces but requires a permitted physical intervention. A conservation plan may use both approaches, with non-contact documentation covering the whole panel and a tiny approved sample reserved for molecular analysis.

How to interpret a future cave-wall DNA claim

Readers, curators and journalists can evaluate a result by checking five points:

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.
  • Sampling permission: Was the location approved by the authority responsible for the cave, and were the restrictions described?
  • Sample context: Was the material pigmented, unpigmented, a calcite crust, sediment or an artefact?
  • Contamination controls: Did the work use ancient-DNA clean-room procedures and report controls for modern DNA?
  • Authentication: Were ancient-DNA damage patterns and other checks used to distinguish old molecules from contamination?
  • Transfer pathways: Does the study separate direct contact from movement through sediment or water, and does it avoid claiming that DNA identifies an artist?

A result that omits these details may still be interesting, but it cannot support the strongest conclusions about who made a painting or when the contact happened.

Why this approach matters for archaeology

The main advance is not a way to extract unlimited DNA from art. It is the discovery that cave walls themselves can preserve ancient human genetic material in places researchers may previously have treated as purely geological. That expands the list of archaeological surfaces worth considering, while the low recovery rate argues for selective, question-driven sampling.

For conservation, the lesson is equally practical: molecular research must be subordinate to the site’s protection plan. Researchers can combine detailed photography, surface analysis and carefully authorized micro-sampling rather than choosing between information and preservation. The 2026 findings show that this compromise can produce genuine evidence, provided the result is presented with its uncertainty and its very small success rate.
