What fermentation does—and what purification does
Manufacturing penicillin has two linked parts. During upstream production, a fungal culture grows under controlled conditions and produces antibiotic in a liquid fermentation broth. During downstream recovery, the process separates that product from the fungus, leftover nutrients and other broth components, then prepares it in a form that can be tested and handled as an active pharmaceutical ingredient (API).

The distinction matters because making penicillin in the fermenter does not make a finished medicine. The broth is a complex mixture, and the antibiotic must be recovered while controlling impurities and protecting product quality. Subsequent steps may include additional formulation and dosage-form manufacturing, which are not the same thing as producing and purifying the API.

1. Establish and expand a production culture
A process begins with a selected production strain and expands it through seed cultures before it enters the main production fermenter. This staged expansion supplies enough viable culture to inoculate the larger vessel in a controlled way. The exact strain and details of the seed train are manufacturer-specific.

For classical fermentation APIs, FDA’s ICH Q7 guidance treats cell-bank establishment and maintenance, inoculation and culture expansion as part of the controlled manufacturing process—not informal preparation. The aim is consistency between batches: the organism entering production must be managed as a defined starting material, and its use must fit the facility’s documented controls.
2. Produce the antibiotic in an oxygenated fermentation
Penicillin production is commonly described as a submerged fungal fermentation: the culture grows in liquid medium while the process supplies oxygen and manages nutrients. Public process overviews describe carbon sources, nitrogen sources such as corn-steep liquor and minerals. Those are examples of media components, not a complete formulation or a standard recipe. Different strains and production strategies can use different media and feeding approaches.
Oxygen transfer is important because this is an aerobic process. The fermenter must support the culture’s growth and production, while operating conditions and nutrient management are controlled for the particular process. A vessel’s contents, the organism and the production conditions interact; changing one element can affect the outcome. Exact operating conditions are process-specific and should not be inferred from a generic description.

The side-chain precursor helps determine which penicillin is produced. Phenylacetate is associated with penicillin G, while phenoxyacetic acid is associated with penicillin V. This is a useful illustration of how fermentation can be directed toward different products, but the two products should not be treated as interchangeable recipes. Product choice influences the process and the required downstream controls.
3. Harvest and clarify the broth
Once the production stage is complete, the broth is harvested and the fungal biomass and other suspended solids are separated, for example by filtration. This clarification creates a liquid stream from which the antibiotic can be recovered. The separation must remove unwanted solids without causing avoidable product loss or compromising quality.

Harvest is therefore more than a mechanical handoff. The recovered liquid still contains substances from the culture medium and fermentation, and the process must manage contamination and the quality of material entering purification. FDA’s API GMP guidance treats harvest as a controlled step within the manufacturing sequence.
4. Extract the antibiotic and remove impurities
In a representative classical recovery route, the clarified broth is acidified and the penicillin is transferred into an organic solvent. The solvent phase can then be handled to separate the antibiotic from water-soluble broth components; further treatment removes impurities such as pigments. A subsequent pH adjustment or addition of a salt can move or precipitate the antibiotic into a form suitable for isolation.
These operations use the product’s chemical behavior to separate it from a mixture. pH changes alter which form of the compound predominates and where it partitions; extraction and later isolation steps exploit those differences. The sequence is not identical for every manufacturer or product, and the publicly described classical route should be read as an example rather than a claim that all current plants use the same solvent or sequence.
Purification has to balance removal of unwanted material with preservation of the product. FDA’s ICH Q7 guidance describes the objective as removing the producing organism, cell debris, media components and other impurities while maintaining consistent recovery quality. A step that improves one aspect of separation still has to be controlled for its effect on the API.
5. Form the product, crystallize and dry
After extraction and purification, processes may form a salt and isolate it by crystallization, followed by drying. These physical operations turn the recovered antibiotic into a manageable solid API and help establish the material for subsequent testing and packaging. Which salt is formed and which operations are used depend on the product and the manufacturing process.

API production also includes physical processing, packaging and quality testing. The release specifications and validated controls are product-specific; a clear-looking crystal or a successful drying step is not, on its own, proof that a batch meets its requirements.
Why penicillin G and penicillin V need not follow identical routes
Penicillin G and penicillin V illustrate how selecting a side-chain precursor affects the penicillin produced: phenylacetate is associated with G, and phenoxyacetic acid with V. That difference is one reason a single public description cannot stand in for every penicillin process. Strains, media, feeding strategies, recovery solvents and purification sequences can also differ.
An academic overview reports that commercial producers regard exact industrial penicillin process technology as highly confidential. Public accounts can explain the broad stages and give historical or illustrative conditions, but they do not establish a present-day standard operating procedure for commercial production. In particular, a temperature, duration or sequence stated in one educational account should remain tied to that account.
How to interpret one published example
The University of Bristol’s educational account describes an aerobic process lasting 4–5 days at 25–27°C. It also describes acidification, solvent extraction, decolorization, potassium acetate precipitation, crystallization and drying. These are the operating details of that historical or illustrative account, not verified universal settings or a confirmed current commercial recipe. They should not be generalized to other strains, products or plants.
Quality controls and facility containment
Penicillin quality is managed throughout production rather than established only by a final test. FDA’s ICH Q7 API guidance addresses cell banks, culture expansion, critical fermentation parameters, harvest, impurity removal and recovery consistency. It also discusses bioburden monitoring where appropriate. Together, these controls help ensure that each stage produces material suitable for the next and that the final API is assessed against its own specifications.

Containment is especially important because penicillins are highly sensitizing materials. FDA’s guidance recommends dedicated production areas for highly sensitizing substances such as penicillins. Separately, FDA explains that U.S. current good manufacturing practice regulations require adequate separation of penicillin-manufacturing facilities and air-handling systems from facilities used for other drugs. This is a facility and cross-contamination control issue, not a purification step.
Regulatory impurity documents address how quality specifications are framed; they do not prescribe one universal manufacturing route. FDA’s April 2026 draft guidance concerns organic impurity specifications for antibiotics made by fermentation and semisynthesis. It is explicitly marked “Not for implementation,” so it is draft guidance rather than an adopted requirement. The European Medicines Agency’s adopted guideline covers related-impurity specifications for human and veterinary antibacterial substances that are fermentation products or semisynthetic derivatives; its page lists the current effective version as first published and last updated on 13 July 2012.
What the historical production figures mean
FDA’s historical account reports that 21 penicillin production plants came online in the United States during the wartime scale-up period. It also reports historical improvements of 100-fold in yield and 85-fold in purity as the mold and its cultivation were improved. These figures describe historical developments, not current plant performance benchmarks, guaranteed yields or a comparison of present-day manufacturers.
The broader lesson is that production improvements involved both the organism and the way it was cultivated. The figures should not be read as a recipe or as evidence that a modern batch will achieve a particular output: the account is historical and does not specify a contemporary process, plant or measurement protocol.

Public sources support a clear high-level account: expand a production culture, run an aerobic fermentation, harvest and clarify the broth, recover and purify the antibiotic, and complete product-specific isolation and testing. They also establish why precursor choice, impurity controls and dedicated containment matter.

They do not establish a single current commercial recipe. Exact strain selection, media composition, operating parameters, solvent choices, process sequence and release specifications vary or are manufacturer-specific. Treating one educational process description as a universal industrial standard would overstate what those public accounts show.
