Researchers isolated two closely related fluorescent coumarins from the roots of orange climber (Toddalia asiatica): 5-methoxyseselin (5-MOS) and 6-methoxyseselin (6-MOS), also known as braylin. Their striking difference appears in water-rich mixtures: 6-MOS grows brighter as it aggregates, while 5-MOS dims. In laboratory cell-imaging experiments, that water-sensitive dimming helped 5-MOS produce images without a wash step.
Where the fluorescent molecules come from
Orange climber is the common name for Toddalia asiatica, a plant whose roots show fluorescence. In a 2023 study, Shan-Shan Chen and colleagues isolated two fluorescent compounds from those roots: 5-methoxyseselin and 6-methoxyseselin, the latter also called braylin. The molecules are close structural isomers, meaning they have the same general molecular components arranged differently. The authors described them as rotor-free aggregation-induced-emission luminogens and, to their knowledge, the first natural coumarins reported to show aggregation-induced emission. The paper appeared in ACS Central Science in 2023.

Why the two isomers behave differently in water
Aggregation-induced emission (AIE) describes a class of luminogens whose light emission changes when molecules gather into aggregates. The two orange-climber compounds do not respond to water-rich conditions in the same way:

The results are a reminder that two isomers can have very different photophysical behavior despite their nearly identical structures. The proposed explanations are interpretations of these experiments, not a universal rule for coumarins or a settled mechanism beyond the tested systems.

How 5-MOS enabled wash-free cell imaging
The authors tested both compounds in live-cell imaging. In their reported experiments, 5-MOS stained MHCC97H cells with negligible background fluorescence, so the cells could be imaged without washing away excess dye. By contrast, background fluorescence from 6-MOS made the cells difficult to see without a wash step.
The study’s explanation is that 5-MOS is dim in aqueous aggregates but becomes emissive after entering cells and interacting with cellular biomolecules. The authors wrote in the 2023 paper’s abstract: “More interestingly, the unique water-sensitive fluorescence property of 5-MOS enables its successful application for wash-free mitochondria imaging.” This describes their laboratory cell-imaging experiments, not an approved diagnostic or established clinical technique.

What the cell-viability result does—and does not—show
In a CCK-8 assay under the study’s conditions, the authors reported no significant variation in cell viability up to 20 μM. That result is limited to the assay and experimental conditions reported; it does not establish general safety in other cells, organisms, or potential uses.
What the finding means
The study connects a naturally occurring plant compound to a practical imaging behavior: 5-MOS’s low background in water-rich conditions can help distinguish cell-associated signal without a washing step. The contrasting response of 6-MOS also shows why fluorescence properties cannot be inferred from a molecule’s broad chemical family alone. The findings are early laboratory research; the cited paper does not establish commercial availability or clinical application.
