How Nanopore Structures Could Tune Drug Crystallisation

Nanopore geometry and surface chemistry may alter how drugs nucleate and crystallise, but results depend on the drug, host and laboratory conditions.

Yes—laboratory studies suggest nanopore shape, size and surface chemistry can influence whether a drug begins to crystallise, what form it takes, and how quickly it dissolves. The effect is conditional, not universal: different pore architectures can hinder or promote nucleation, and confinement can also help preserve a drug in an amorphous state. These are research strategies, not evidence that nanopore-engineered medicines are in routine clinical use.

How can a pore change crystallisation?

Crystallisation begins when molecules arrange into an ordered structure. Inside a nanoscale pore, the available space and the pore wall can affect that process: geometry may influence molecular ordering, while surface chemistry can strengthen or weaken interactions between the drug and its host. Pore dimensions and manufacturing conditions also matter. As a result, confinement may encourage nucleation in one system and suppress it in another.

Researchers are investigating these effects for different aims: controlling nucleation, influencing crystal form, producing nanocrystals, or stabilising amorphous drug material. Those aims are related, but not interchangeable. A result for one drug and host material does not establish what will happen with another.

What experiments show about pore shape and size

Aspirin: angular and spherical pores behaved differently

In a 2011 study, Diao and colleagues patterned polymer films with spherical and angular nanopores ranging from 15 to 120 nm in diameter. Spherical pores hindered aspirin nucleation, while angular pores promoted it under the tested conditions. The angular-pore effect required favourable interactions between the surface and the aspirin molecules; the authors suggested that changes in molecular orientational order near pore angles may help explain the result. Fenofibrate: a size threshold in controlled-pore glass

Dwyer and colleagues studied fenofibrate in controlled-pore glass with ten pore sizes from 12 to 300 nm. They reported drug loading above 20 wt% for pore sizes larger than 20 nm. Nanocrystalline fenofibrate formed above that size, while the smaller pores did not yield the same reported crystalline result. The confined crystals showed melting-point depression consistent with a Gibbs–Thomson relationship, as well as enhanced dissolution rates in the study. These are experimental findings for fenofibrate in that host, not a general threshold for other drugs. Royal Society of Chemistry journal article (2015).

Nanopores can also help stabilise amorphous material

Not every strategy aims to create crystals. Rengarajan and colleagues described confinement in nanoporous hosts with strongly interacting pore walls as a way to extend the lifetime of amorphous drugs by changing thermodynamics and crystallisation kinetics. That makes the host–drug interaction central: confinement can favour crystallisation in one setup and help preserve a non-crystalline state in another. Journal of Materials Chemistry study (2008).

MCE Gridded Membrane Filter, 47 mm 0.45 µm, Sterile Individual Pack, 100/Pk
  • APPLICATION: Mixed Cellulose Ester (MCE) membrane Filter is composed of cellulose acetate and cellulose nitrate, which is higly biologically inert and widely used in Microboilogical analysis of water, wastewater, cosmetic, pharmaceuticals and beverages.
  • HIGH QUALITY MEMBRANE: Sterile gridded membrane filter is without support layer naturally hydrophilic membrane, ideal for Colony Counting and Sterility Testing, and our MCE membrane with ISO 9001 certificated and meet ISO 7704 requirements.
  • GRID LINES: Gridded filters have clearly defined grid lines spaced at 3.1 mm intervals. Contrasting grid lines facilitate counting colonies on the filter surface and are tested to assure freedom from grid line inhibition.
  • HIGH POROSITY: Microbail membrane filter have high porosity allows gases and liquids to provide high filtration rates in every area. High porosity also provides a large surface area for adsorption and binding.
  • STERILE INDIVIDUAL PACKAGE: Membrane filter disc filters sterilized by 25 kGrey gamma radiation, sterile-sealed without protective paper, placed on a band in individual packages. This special setting ensures each membrane filter will stay perfectly flat when they are dispensed.

What determines whether the approach works?

Experimental comparisons need to account for more than pore diameter. Relevant variables include the pore’s geometry, host material and surface chemistry; the strength of drug–surface interactions; process conditions such as temperature and pH; and whether crystallisation happens within the pores or on their exterior. Researchers also need to define the intended outcome—nucleation, a particular crystal form, nanocrystal formation or amorphous-state stabilisation—before judging whether a result is useful.

Mesoporous silicon is one material discussed for its large loading capacity, adjustable pore size and adaptable surface. A review also identifies crystallisation outside pores, along with processing variables such as temperature and pH, as challenges for translation. Mesoporous silicon review (2020).

Beyond rigid nanopores

Porous solids are not the only templates being studied. Hydrogel microparticle templates have also been investigated as a way to crystallise small-molecule drugs, offering a different material setting from rigid nanoporous hosts. Hydrogel microparticle study.

What these findings do—and do not—show

The studies establish that pore architecture can change crystallisation behaviour under laboratory conditions. They do not establish a universal method for improving drug solubility, a routinely prescribed nanopore-engineered medicine, or better outcomes for patients. Even the enhanced fenofibrate dissolution rate is an in-vitro experimental result; it cannot by itself show that a medicine would work better in people.
