Assembly Theory: How Chemistry Could Explain Molecular Complexity and Life’s Origins

Assembly theory links a molecule’s minimum construction path with its abundance to investigate selection. Experimental measurements exist, but its claims about life’s origins and biosignatures remain under debate.

Assembly theory treats a molecule not only as a structure but as the result of a possible construction history. Its central proposal is that the minimum number of steps needed to build an object, considered alongside how many copies of it occur, can help describe selection in a collection of objects. That offers a chemical way to investigate complexity and life’s origins—but it is a research framework, not a demonstrated explanation of how life began.

What assembly theory is trying to explain

Many ways of describing molecules focus on what they are made of: their atoms, bonds and arrangement. Assembly theory (AT) adds a different question: how much construction would be needed to produce an object from simpler parts?

In “Assembly theory explains and quantifies selection and evolution,” published in Nature on 4 October 2023, Sharma and colleagues present objects as associated with possible formation histories. In a chemical example, forming bonds can be treated as elementary operations. The framework aims to use those histories to reason about how complex objects arise and how selection may affect which objects become common.

This makes chemistry central to the proposal. Rather than treating the transition from nonliving matter to life as a boundary that can only be discussed at the level of biology, AT seeks a way to describe relevant changes in chemical systems. That is an intended use of the framework, not evidence that the transition has been reconstructed.

What is the molecular assembly index?

The molecular assembly index (MA) is the minimum number of steps in a construction path that produces a given molecule, under the assembly rules being used. It is a measure of a proposed minimal construction history, not a count of every reaction the molecule has actually undergone in nature.

It is important to distinguish the index for one object from the paper’s broader assembly quantity, denoted A. The latter considers both the assembly indices of objects and their copy numbers in an ensemble. In the authors’ account, the index describes construction complexity; the ensemble quantity is intended to capture the degree of causation or selection represented by the collection.

Measure What it describes
Molecular assembly index (MA) The minimum number of steps in a path that produces an object.
Assembly quantity (A) An ensemble-level quantity that uses assembly indices together with the number of copies of observed objects.

The distinction matters because, in the authors’ framework, a single molecule with a high assembly index does not by itself show that selection occurred. Copy number matters to their proposed ensemble measure. The index and A therefore answer related but different questions.

How could copy number point to selection?

Imagine observing a collection that contains repeated copies of some objects. AT proposes that the construction complexity of those objects, considered together with their abundance, can help characterize how much selection or causation is reflected in the ensemble. The idea is not simply that “complex means alive”; it is that a pattern of complex objects and their repetition may contain information about the processes that produced the pattern.

This is a proposed way to quantify selection and novelty across physical and biological settings. The 2023 paper does not establish that assembly theory replaces evolutionary biology, nor does its framework alone identify the historical process that created any particular molecule. Applying the measure to a sample still requires interpretation of what was measured and how it relates to the system that produced it.

What has been measured experimentally?

A 2024 study by Jirasek and colleagues in ACS Central Science reported measuring molecular assembly indices using three techniques: nuclear magnetic resonance (NMR), tandem mass spectrometry (MS/MS) and infrared spectroscopy. The use of multiple measurement approaches is significant because it puts MA measurement into experimental practice rather than leaving it solely as a theoretical proposal.

That result supports the narrower claim that assembly-index measurement can be investigated with spectroscopy. It does not independently validate every broader interpretation of AT, including its claims about the origin of life or a universal way to identify biological signatures. Measuring an index and establishing what that index means for life detection are separate steps.

Can molecular complexity show that a molecule came from life?

Not on its own, based on the evidence described here. The prospect of using molecular assembly as a biosignature is one of the framework’s most consequential applications, but the proposed threshold for distinguishing life-related molecules from nonliving chemistry remains debated.

A 2024 paper, “Experimentally measured assembly indices are required to determine the threshold for life,” argues that actual measurements are needed to assess such a threshold. Its discussion notes theoretical inorganic molecules with assembly indices above 15. That does not establish 15 as a universal dividing line: theoretical examples and a proposed threshold are not the same as a validated test that reliably distinguishes living from nonliving origins.

For a biosignature claim to be persuasive, a high index would need to be interpreted in context. Researchers would need to establish how the measured value was obtained and whether plausible nonbiological processes could produce comparable results. The cited threshold-focused paper emphasizes the need for measured values; the broader discrimination question is not settled by the existence of a measurement method.

What are the criticisms of assembly theory?

A 2024 review in npj Systems Biology and Applications, “On the salient limitations of the methods of assembly theory and their classification of molecular biosignatures,” challenges aspects of the method and its application to molecular biosignatures. Among its criticisms, the authors argue that the assembly pathway method is an encoding scheme used by statistical compression algorithms.

This is a published methodological critique, not a final resolution of the debate. It raises questions about how assembly pathways are defined and what conclusions the resulting index can support. Experimental measurement helps make the framework testable, but does not by itself answer those methodological questions.

Reactions to the 2023 paper also combined interest with caution. Palli Thordarson, a chemical self-assembly researcher at the University of New South Wales, said AT “clearly has merit” and has advanced thinking about complex chemical systems, particularly in origin-of-life research and astrobiology. He also cautioned that stronger experimental confirmation in prebiotic chemistry would be needed before accepting a deeper fundamental role for the theory. Lee Cronin of the University of Glasgow acknowledged that the theory could be wrong, while suggesting it may help investigate the connection between physics and biology through chemistry. These are researchers’ views reported in 2023, not a consensus verdict.

What assembly theory can—and cannot—say about life’s origins

AT offers a framework for asking whether the abundance and construction complexity of molecules can reveal something about selection in chemical systems. Its chemistry-first perspective makes it relevant to origin-of-life questions, where researchers seek to understand how increasingly organized chemistry could precede biology.

But proposing a measure is not the same as demonstrating a path from nonliving chemistry to life. The 2023 paper sets out a framework and its intended significance; the 2024 spectroscopy study reports measurement work; and the published critiques question methods and interpretations. Taken together, these developments make assembly theory a testable and contested research approach—not a settled account of abiogenesis or a proven universal biosignature test.
