How Fast DNA Nanomachines Roll and What They Can Detect

DNA-coated particles rolled across RNA-bearing surfaces when RNase H removed hybridized RNA. Here’s what the study measured—and what its mutation-sensing result does not prove.

DNA nanomachines in a 2015 laboratory study moved by rolling DNA-coated particles across a surface, not by taking steps. The researchers reported average speeds near 2 micrometres per minute and a maximum of 5 micrometres per minute under their experimental conditions. They also demonstrated a proof-of-concept method for detecting a single-nucleotide change; it was not a clinical test or a consumer device.

How the DNA roller moves

The motor pairs a spherical particle coated with DNA strands with complementary RNA strands attached to a surface. When the particle binds to the surface, the DNA and RNA form hybrids. The enzyme RNase H selectively hydrolyses the RNA in those hybrids, releasing the particle’s bonds so it can bind at new points and continue rolling.

This is a chemically powered surface motor, not a free-swimming robot. Its movement depends on the particle, the RNA-bearing surface and the enzyme working together. The study describes this design as rolling rather than the stepwise walking used by earlier DNA motors.

How fast it was in the experiments

Yehl and colleagues measured average velocities of 1.8 ± 0.4 μm/min for 0.5 μm particles and 1.9 ± 0.5 μm/min for 5 μm particles. The full paper reports maximum velocities of up to 5 μm/min. These are measurements in the study’s particular experimental system, not a general speed specification for DNA motors.

The authors also described the roller’s maximum speed and processivity as three orders of magnitude greater than the maximum for conventional DNA motors. That is the paper’s comparison with earlier DNA motors; it should not be read as a universal ranking against every molecular motor or as evidence that the systems are interchangeable.

How the path depends on particle shape

Spherical motors moved in a self-avoiding manner in the reported experiments. The authors also found that anisotropic particles—including dimerized and rod-shaped particles—could travel linearly without a track or an external force. These results show that particle geometry affects path behavior; they do not establish one path as the best choice for every application.

What the mutation-sensing demonstration showed

The researchers demonstrated detection of a single-nucleotide polymorphism by measuring particle displacement with a smartphone camera. It was a proof of concept: the reported experiment does not establish clinical accuracy, suitability for diagnosing patients, or availability as a consumer test.

What the result does—and does not—establish

The paper, by Kevin Yehl and colleagues, appeared online on 30 November 2015 and in the February 2016 issue of Nature Nanotechnology. It presents a DNA-based rolling motor and experimental findings about its movement, particle shapes and a sensing demonstration. It does not report a commercially available nanorobot or a validated clinical diagnostic.
