How does light disrupt the walls of capped nanotubes?

A 2011 laboratory demonstration showed how light can disrupt the amphiphile assembly in vesicle-capped nanotubes, while keeping experimental results distinct from proposed applications.

Light can disassemble these capped nanotubes by changing the shape of the synthetic amphiphile that builds their walls. In a 2011 laboratory demonstration, that molecular change disrupted the packing holding the nanotubes together; phospholipid vesicles capped the tube ends, but the study did not establish a drug-delivery product or a process tested in living systems.

What are vesicle-capped nanotubes?

They are self-assembled structures made from a synthetic, light-responsive amphiphile—not commercial carbon nanotubes. In water, the amphiphile forms bilayer structures that assemble into nanotubes. Researchers added DOPC, a phospholipid, to create longer hybrid structures. Because the phospholipid and amphiphile phase-separated, DOPC formed vesicles at the nanotube ends.

In the formulation reported by Coleman and colleagues, a 2:1 amphiphile-to-DOPC ratio produced tubes around 10 μm long and 28 nm wide, with a nanotube wall about 3 nm thick. These are measurements from that experiment and formulation, not general specifications for capped nanotubes. The study appeared in Nature Nanotechnology on 14 August 2011.

How does light disassemble the tubes?

The amphiphile contains a light-responsive overcrowded-alkene unit. Irradiation drives a photochemical cyclisation, changing the molecule’s conformation. That change disrupts the molecular packing needed to sustain the amphiphile’s assembly, so the nanotube disassembles. The primary paper reports that the light-induced disassembly was observed in real time.

The trigger is therefore a change in the building block, not a mechanical action that pulls the tube apart. The experiment demonstrated a light-responsive molecular assembly under laboratory conditions; it does not establish how the material would behave in a living system.

Can the caps be removed without breaking the nanotube?

In the reported experimental conditions, the DOPC vesicle caps could be removed and reformed while the nanotube remained intact. This was possible because the caps and tube had distinct bilayers. It describes a laboratory manipulation, not a consumer process or evidence that cap removal works inside the body. Chemistry World’s contemporary report, published 15 August 2011, also described the cap-removal result.

What did the study establish—and what remains a proposal?

The 2011 work established that researchers could assemble the hybrid nanotubes, manipulate the vesicle caps under experimental conditions, and use light to disrupt the amphiphile assembly. It did not demonstrate a clinical treatment, a working drug-delivery product, or a commercial nanoengineering application.

Chemistry World quoted smart-polymers researcher Brent Sumerlin as saying the material’s adaptability “indicates applications in controlled drug delivery and nanoengineering.” That is a statement about potential, not a report of demonstrated performance. The same article attributed to Ben Feringa the aim of demonstrating how a complex nano-object could be controlled and changed significantly.

A 2016 Chemical Communications study later examined phospholipid-vesicle capping under osmotic conditions. It reported that the phase-transition temperature of zwitterionic phospholipids affects phase separation and cap formation; cationic caps could be loaded into nanotubes by osmosis, while anionic caps remained stable under hyper-osmotic conditions. This follow-up adds experimental context about caps, but does not show that the 2011 design became a commercial delivery platform.
