A 2011 report on progesterone pointed to impurities as a possible reason one form resisted reproduction, while a separate 2022 co-crystal study showed that a missing form could be recovered through carefully designed experiments.
What is a disappearing polymorph?
A polymorph is one of several distinct crystal arrangements that can be made from the same chemical substance. The molecules are chemically the same, but their arrangement in the solid differs. That difference can affect properties such as stability and solubility, so obtaining one form rather than another can matter scientifically and practically.

A form that cannot be reproduced has not necessarily vanished from existence. It may be that the conditions that produced it are not understood or controlled well enough to make it again.

Why did one progesterone form prove hard to reproduce?
A 2011 Chemistry World report described researchers’ difficulty reproducing a progesterone polymorph. They compared two forms and found 11 impurities in form 2, versus three in form 1. The researchers suspected that impurities—other steroids introduced during synthesis to induce crystal growth—might help explain why form 2 was more stable than form 1.

The impurity explanation remained a hypothesis: the exact structures of the impurities were not identified. Without knowing which substances were present, it was difficult to recreate the original conditions reliably. The reported counts describe this progesterone study; they do not establish a general rule about impurities in pharmaceutical crystals.
Can a missing crystal form be recovered?
Yes, at least in one later case. A 2022 abstract from the International Union of Crystallography describes three polymorphs of urea–barbituric acid co-crystals. After researchers obtained the P21/c form, they could not reproduce the Cc form under the same or modified conditions, either locally or at other research groups.

The authors report that electron-density studies and interaction-energy calculations informed the design of an experiment that reproduced the missing Cc form and established conditions for the other two forms. This is a distinct case from progesterone: it concerns a co-crystal made from two components, and it reports successful recovery using a quantum-crystallography-informed approach. It does not identify the cause of the progesterone form’s behavior.

Crystal form can be important in pharmaceuticals because regulatory approval may apply to a particular form, and a different, more stable form can be difficult to control in production. The Chemistry World report used ritonavir as a separate illustration: it described the appearance of a second, less soluble form that affected the drug’s performance and required a manufacturing response. Ritonavir was not part of the progesterone study.

Is this the same as the story about a crow and shiny stones?
No. “The mystery of the disappearing crystals” also appears as the title of a children’s story about a crow taking shiny pebbles and crystals. The chemistry meaning discussed here is a puzzle about crystal structures that researchers struggle to reproduce, not a tale about stolen decorations.
