By comparing ancient genomes with one another and with present-day populations, researchers can test whether populations were related, whether ancestry moved between regions, and whether groups mixed or changed over time. The results are evidence for population history—not a map of a migration route, and not a direct measure of someone’s language, culture, or identity.
How scientists use ancient DNA to study migration
The process begins with human remains recovered from an archaeological context. Their location and estimated age let researchers place genetic data in space and time. DNA preserved in the remains is then compared with data from other ancient individuals and living populations.

Researchers look for patterns of genetic similarity and difference across samples. Statistical models use those patterns to test population-history explanations, such as whether groups shared ancestry, whether people moved between regions, or whether populations mixed. A model can show that a proposed history is consistent with the genetic evidence; it cannot reveal a route directly from a genome.

Archaeological and historical evidence helps interpret the genetic patterns. A genetic shift in a region may be consistent with migration or population turnover, but DNA alone does not explain the social causes of movement or establish which language people spoke.

What different kinds of DNA can reveal
Mitochondrial DNA traces one maternal line
Mitochondrial DNA is inherited through the biological maternal line. It can help researchers study that lineage, but it represents only one narrow strand of a person’s ancestry. It cannot, on its own, describe the full history of a population.
Genome-wide data support broader comparisons
Genome-wide data provide many more points of comparison. When researchers have suitable samples from multiple periods and places, they can use these patterns to investigate broader relationships, population mixture, and changes in ancestry. The conclusions remain dependent on which people were sampled and on the assumptions of the population models used.

What ancient-DNA studies have found in different regions
Ancient-DNA findings are regional rather than one continuous, complete account of human movement. The examples below address different periods and questions, so their sample sizes and conclusions should not be treated as directly interchangeable.

| Region and period | Evidence and reported finding | How to interpret it |
|---|---|---|
| Central and Eastern Europe, sixth to eighth centuries CE | A 2025 Nature study analyzed 555 ancient individuals, including 359 from Slavic contexts dating as early as the seventh century CE. Its model linked large-scale movement from Eastern Europe with the spread of Slavs and estimated more than 80% replacement of the local gene pool in Eastern Germany, Poland, and Croatia. | The authors also reported substantial regional variation. The replacement estimate applies to the regions and period studied; it does not mean every locality or social group changed in the same way. |
| The Americas, early peopling | A 2021 Nature review describes dispersal from northeast Asia involving distinct populations, followed by rapid expansion south of the continental ice sheets and branching into multiple populations. | This is a complex, episodic history, not evidence for one simple, uniform wave of settlement. |
| Europe, across earlier periods | A 2014 study modeled present-day European ancestry as deriving from at least three differentiated ancient populations: western European hunter-gatherers, ancient north Eurasians, and early European farmers. A 2016 study analyzed 51 people dated to approximately 45,000–7,000 years ago and reported recurring migration and population turnover. | These findings describe multiple ancestry sources and changes over time, rather than a single origin for all Europeans. |
| Eastern and south-central Africa, spanning approximately 18,000 years | A 2022 study presented genome-wide data from six individuals alongside previously published samples. The patterns were consistent with deep population structure and past mobility. | Six individuals from these regions cannot provide a complete map of African population history. |
What genetic ancestry does—and does not—say about identity
Genetic ancestry describes patterns of biological relatedness inferred from DNA and the comparison populations available to researchers. It is not interchangeable with ethnicity, nationality, language, or cultural practice. Those identities have social and historical dimensions that a genome cannot independently establish.

The distinction matters even when a study identifies a major population shift. The 2025 Central and Eastern European study connects large-scale movement with the spread of Slavs, but its finding of regional heterogeneity argues against treating the period as one uniform genetic outcome or treating a population label as a fixed biological category.
How to read claims about Neanderthal ancestry
The Smithsonian Institution Human Origins Program summarizes estimates of about 1–4% Neanderthal ancestry in non-African present-day populations, with variation among regions. It also describes evidence of Neanderthal ancestry in African populations. These are population-level estimates that vary with the populations and methods considered; they are not personal identity labels or a complete account of an individual’s ancestry.

Why the evidence has limits
- DNA preservation is uneven. DNA degrades over time, so recoverable material is scarce and not evenly available across regions, periods, or archaeological settings.
- Samples are not whole populations. An ancient individual is evidence about that person and a contribution to a broader model, not a census of everyone living nearby. Uneven sampling can leave gaps in the history researchers can reconstruct.
- Migration is inferred, not read off a genome. Models test explanations against genetic patterns and available samples. Their conclusions should be described as supported by or consistent with the evidence, not as a directly observed route.
- Genetic evidence needs context. Archaeology and history help make sense of population patterns, while DNA by itself cannot establish language, cultural practice, or identity.
- Research involves descendant-community interests. Ancient human remains and genetic data raise ethical questions. The cited sources do not establish a single consent or governance protocol applicable to all studies.
Why a consumer ancestry test is not the same as ancient-DNA research
Ancient-DNA studies compare genetic data from dated archaeological remains with other ancient and present-day data, then use population models to investigate histories over time. A consumer ancestry test is not a way to reproduce those studies: it does not, by itself, provide the dated archaeological samples and population-history comparisons needed to test migration hypotheses.
