In supersaturated water containing amino acids and short peptides, Liao, Das, Robb, Beveridge and Wynne reported a continuous population of disordered, solute-rich aggregates ranging from molecular dimers to oligomers of at least 30 molecules, nanometre-scale clusters and micrometre-scale particles. Larger members of this population became places where crystals formed spontaneously or after laser stimulation.
The proposed mechanism is straightforward: low-surface-tension amorphous aggregates appear with little effective barrier, concentrate solute internally, and then allow an ordered crystal nucleus to form inside the enriched volume. This helps explain why a crystal nucleus can emerge from a disordered aggregate rather than being built directly from isolated molecules.

What are prenucleation clusters?
Prenucleation clusters are transient, solute-rich groupings that exist before an ordered crystal nucleus appears. The term is often used for small molecular assemblies, but the 2024 Chemical Science study describes a much broader population. It found amorphous aggregates across an almost continuous size range: dimers and other oligomers, species containing 30 or more molecules, nanometre-scale clusters and particles visible at micrometre scales.

“Amorphous” means these assemblies lack the repeating long-range order of a crystal. They are not simply tiny crystals waiting to grow. Their molecules are concentrated and associated, but their arrangement remains disordered. “Prenucleation” indicates their position in the sequence: they are present before detectable crystalline order develops.

This distinction matters because a conventional picture starts with individual dissolved molecules joining into a sufficiently stable crystal nucleus. The new observations instead identify a measurable, disordered environment that can precede and host crystallization.
How amorphous clusters help a crystal nucleate
- Amorphous aggregation: supersaturated solute molecules gather into low-surface-tension, disordered aggregates. The authors describe this initial formation as essentially barrierless compared with making a crystal surface directly.
- Internal enrichment: molecules become more concentrated inside an aggregate than in the surrounding solution. That local concentration changes the conditions experienced by the molecules.
- Crystallization within the aggregate: an ordered crystal nucleus forms in the enriched interior. The aggregate therefore acts as a precursor and physical site for nucleation, rather than being the final crystal structure.
In this model, the difficult step is not necessarily the first gathering of molecules. The crystal-forming barrier is encountered after a dense amorphous phase already exists, where the local composition and interfaces can be more favorable.
What the 2024 experiments found
| Observation | What it contributes |
|---|---|
| Dimers through at least 30-mers and larger species | Mass-spectrometry evidence that association begins at molecular scale and extends to larger oligomers. |
| Nanometre- and micrometre-scale particles | Evidence that the same broad aggregation phenomenon is not confined to one molecular size. |
| Broad, time-dependent particle-size distribution | Dynamic light scattering (DLS) shows aggregates appearing and changing rather than one fixed preferred diameter. |
| Broad, unstructured low-frequency Raman bands | In-situ terahertz Raman spectroscopy identifies an amorphous phase distinct from both solution and crystal spectra. |
| Larger aggregates associated with crystallization | They were observed as sites of slow spontaneous nucleation and as intermediates during laser-induced nucleation. |
The samples were supersaturated aqueous solutions of amino acids and di- and tri-peptides. Aggregate appearance took approximately one day in the reported observations; the paper’s conclusions describe a one-to-five-day timescale, followed by redissolution over hours. These are laboratory observations for the tested systems, not a universal timetable for every crystallizing substance.
Why several instruments were needed
Dynamic light scattering
DLS infers apparent hydrodynamic particle sizes from fluctuations in scattered light. Here it revealed a broad distribution and growth over time. A broad distribution is important: interpreting one reported radius as the size of “the” prenucleation cluster would lose the central result that aggregates span many scales. Earlier DLS studies reviewed by the authors reported hydrodynamic radii of about 75 nm and 500 nm; those values are prior observations, not a single characteristic size established by this experiment.

Mass spectrometry
Mass spectrometry detected molecular clusters from dimers through at least 30-mers and larger species. This supplies molecular-scale evidence that complements DLS, which reports apparent particles and can combine signals from differently sized populations.
In-situ terahertz Raman spectroscopy
Raman measurements made in the solution showed broad, unstructured low-frequency bands associated with an amorphous phase. The spectrum differed from those of the liquid solution and the eventual crystal, providing structural evidence that the aggregates are a distinct disordered state.
No single method proves the entire pathway alone. DLS is sensitive to broad size distributions; mass spectrometry establishes oligomeric association; Raman spectroscopy addresses structural character. Their combination is what supports a molecular-to-micrometre aggregation picture.

How this differs from classical nucleation theory
| Question | Classical description | Aggregate-assisted description |
|---|---|---|
| Precursor | Individual solute molecules assemble into a crystal nucleus. | A pre-existing amorphous, solute-rich aggregate precedes the crystal nucleus. |
| Free-energy path | A crystal nucleus must cross a conventional interfacial free-energy barrier. | Low-interfacial-tension aggregate formation is followed by crystallization inside the enriched phase. |
| Intermediate | No disordered mesoscale phase is required. | Oligomeric, nanometre and micrometre amorphous aggregates are observable. |
| Trigger | Supersaturation drives formation of a sufficiently stable crystal nucleus. | Local enrichment can support spontaneous, laser-assisted or potentially shear-assisted nucleation. |
| Scope | A general theoretical framework for nucleation. | Direct experimental support in the amino-acid and peptide solutions tested. |
The result does not make classical theory useless. Rather, it adds a nonclassical route that classical free-energy ideas can be extended to describe: the system may first enter a disordered dense state, then cross the crystal-ordering barrier within that state. Chemistry World summarized the implication by noting that clusters provide nucleation sites, a finding that challenges a simple classical picture.

Why larger aggregates matter
The study links larger amorphous aggregates with slow spontaneous crystal nucleation. It also identifies them as intermediates in laser-induced nucleation. A laser can trigger crystallization in a population that already contains suitable enriched aggregates; it need not create every nucleus from a uniform solution at the instant of irradiation.
This observation explains why nucleation can be spatially localized and temporally delayed. Aggregates can grow, concentrate solute and redissolve over hours. A solution may therefore remain apparently clear while its molecular organization is changing, with only some larger aggregates reaching conditions favorable for crystal order.
What “universal role” means here—and what it does not
The authors state that their results indicate a universal role for amorphous aggregates in crystal nucleation. In context, that conclusion refers to the amino acids and di- and tri-peptides examined and to the consistent evidence across size scales and measurement methods. It does not prove that every inorganic, organic, biological or industrial crystallization process follows exactly the same sequence.

- The tested chemistry is limited to the reported molecular-solute systems.
- DLS reports apparent hydrodynamic sizes and can be influenced by broad mixtures of particles.
- The observed timescales should not be transferred directly to another solvent, concentration, temperature or solute.
Practical implications for crystal-growth research
Monitor the solution before visible crystals appear
Clear solutions should not automatically be treated as structurally uniform. Time-resolved DLS, molecular mass measurements and spectroscopic monitoring can reveal aggregation before macroscopic crystals are visible.

Separate aggregate formation from crystal ordering
Experiments should ask two questions: when does a dense amorphous phase appear, and when does crystalline order begin inside it? Treating those as one event can obscure the intermediate.
Control aggregate history
Waiting time, mixing, supersaturation and external stimulation can change the population of aggregates. Reproducible crystallization therefore requires recording the solution’s age and treatment, not only its final concentration.
Use complementary evidence
A particle-size signal alone cannot establish that a population is amorphous or that it contains the molecular species detected by mass spectrometry. Combining size, composition and structural measurements provides a stronger mechanistic test.
Common misconceptions
“Prenucleation clusters are tiny crystals.”
Not necessarily. The reported aggregates are amorphous and spectroscopically distinct from the crystal. Their role is to provide a concentrated precursor environment.

“There is one optimal cluster size.”
The central finding is the opposite: aggregates form a broad, continuous distribution from molecular assemblies to micrometre particles. The 75 nm and 500 nm radii cited from earlier work should not be treated as universal targets.
“Every aggregate crystallizes.”
No. Larger aggregates were associated with nucleation, but the observations do not say that every aggregate becomes a crystal. Aggregates can persist, grow or redissolve.
“The finding disproves nucleation theory.”
It challenges a molecule-by-molecule-only pathway and motivates an extension of classical nucleation theory. It does not eliminate thermodynamic barriers or establish one mechanism for all materials.

Frequently Asked Questions
How can amorphous clusters help crystals nucleate?
They concentrate solute in a disordered, low-interfacial-tension phase. Crystal order can then form inside that enriched aggregate instead of assembling directly from isolated molecules in dilute solution.
What are prenucleation clusters?
They are transient molecular or larger solute-rich assemblies that exist before an ordered crystal nucleus. In the reported systems they range from dimers and oligomers to nanometre- and micrometre-scale amorphous particles.
Why do crystal nuclei form inside disordered aggregates?
The aggregate provides higher local concentration and a distinct interface, making crystal ordering more favorable there than in the surrounding solution. The evidence supports this pathway for the tested amino-acid and peptide solutions, not automatically for every material.
