An AI-discovered DNA sequence should be treated as potentially dual-use until its biological function, organism, provenance, synthesis accessibility and plausible misuse pathways have been assessed. A benign-looking sequence can carry context-dependent risks, while a novel design may not match existing screening databases.
Before publishing sequence-level details or ordering DNA, use institutional biosafety and dual-use review, trained personnel, appropriate containment, complete-sequence screening, customer and institution verification, and strict controls for human genomic data. Screening is a baseline safeguard—not proof that a design is harmless.

What “safe” means for an AI-discovered DNA sequence
Safety is not a property that can be assigned from the fact that a model generated a sequence. It is a risk decision based on what the sequence does, where it will be used, who can obtain it, and how easily a harmful application could be reconstructed.

- Intended biological function: Is the design for a non-pathogenic, non-toxic purpose, or could it affect replication, host range, immune response, delivery, antimicrobial resistance or toxin activity?
- Organism and system: A sequence that appears harmless in a computational screen may behave differently in a live organism, cell line, vector or environmental context.
- Novelty and uncertainty: AI can propose combinations or sequences with no close match in reference databases. Lack of a match is not evidence of safety.
- Data provenance: Determine whether the model, training data or output contains controlled-access human genomic information, pathogen data or other restricted material.
- Synthesis accessibility: Consider whether the sequence can be ordered from a provider, built on benchtop equipment, or reconstructed from the information you plan to release.
- Misuse pathway: Ask whether publication would lower the expertise, time, cost or equipment needed to create a harmful biological agent or improve one.
Published official guidance does not provide a universal numerical cutoff at which an AI-generated sequence becomes safe. No authoritative source establishes a validated probability that a particular AI design is harmless. A defensible decision therefore documents the evidence, uncertainties and controls rather than assigning a made-up safety score.

Why AI changes the biosecurity profile
Generative models can accelerate protein engineering, genome editing and molecular synthesis. In a 2025 analysis, NIST authors Mengdi Wang and colleagues warned that these capabilities could be misused to enhance viral virulence, design toxins or modify human embryos. They call for safeguards such as watermarking, alignment, anti-jailbreak measures and unlearning of dangerous capabilities.
The risk is not limited to a model answering an obviously harmful prompt. A system may generate a sequence, optimization suggestion, protocol fragment or ranked design that becomes dangerous when combined with other information. Model weights, prompts, datasets and laboratory automation can each change who can reproduce the result.
NIST also identifies a harder problem: a sufficiently capable system could design entirely novel DNA sequences that current screening tools do not recognize. That means a “no hit” result is only one input to a risk assessment. Screening databases, standards and validation methods must evolve as sequence-generation methods change.

Controls expected before DNA is ordered
Sequence screening and customer checks are baseline procurement controls, not optional paperwork. The 2023 U.S. Department of Health and Human Services framework recommends screening orders, checking customer identity and legitimacy, retaining records of orders containing sequences of concern, tracking responsible transfers and verifying customers who operate capable synthesis equipment. UK guidance issued by the Department for Science, Innovation and Technology in 2024 similarly recommends screening and authenticating legitimate users, including users of benchtop synthesizers.
For an AI-discovered design, a responsible order process should include:

- Screen the complete sequence, not only the most obvious region, and retain the provider or validated service result.
- Confirm the institution, principal investigator, end user and stated purpose.
- Document the applicable biosafety approval, containment level and responsible person.
- Use human review for unusual orders, positive or ambiguous results, and any autonomous laboratory action.
- Record transfers of a sequence of concern and limit access to people with a legitimate need.
Screening can identify similarity to known sequences of concern and flag an order for review. It cannot guarantee detection of a novel harmful design, a fragmented order, an altered sequence or a design whose danger depends on a particular host or experimental context.
Publication decision framework
Publication is a governance decision as well as an editorial one. NIH dual-use procedures include manuscript and abstract review, and the National Science Advisory Board for Biosecurity (NSABB) recommends assessing information hazards and considering structured access or other mitigations for dual-use AI models, datasets and results.

| Candidate or release plan | Typical indicators | Prudent action |
|---|---|---|
| Low-risk candidate | Non-pathogenic, non-toxic, non-human sequence with a clear benign function; no restricted data; no plausible enhancement or evasion capability. | Document the assessment, obtain any required institutional sign-off, and screen every synthesis order. Explain the limits of the evidence. |
| Elevated-risk candidate | Association with a pathogen or toxin; possible host-range, immune-evasion, replication, delivery or antimicrobial-resistance function; novel design with uncertain activity. | Seek institutional biosafety and dual-use review before releasing sequence-level details. Consider redaction, controlled access, or publication of high-level methods and validation only. |
| Do not publish openly without review | Information that could directly enable construction, rescue, enhancement or evasion of a harmful agent, or that exposes controlled human genomic data. | Escalate to the institutional biosafety committee, dual-use review body, funder and the journal or publisher’s security process. Do not release the enabling details while review is pending. |
“Low risk” in this table means lower concern under the stated conditions; it is not a guarantee. Reassess if the organism, phenotype, model, dataset, intended audience or release format changes.
Experimentation checklist
Use the following sequence before moving from an AI output to physical work.
- Define the use case. Write down the intended organism or system, expected phenotype, materials, collaborators and a realistic misuse scenario. Include what would happen if the sequence performed better than expected.
- Obtain institutional review. Contact the institutional biosafety committee and the body responsible for dual-use or information-hazard review. Follow the required containment, training, supervision and incident-reporting rules.
- Evaluate the sequence and fragments. Screen the complete construct and relevant fragments with a synthesis provider or validated screening service. Preserve the result, date, sequence version and any reviewer comments.
- Verify people and institutions. Confirm the principal investigator, end user, institution, funding and legitimate purpose. Keep approvals and records for any sequence-of-concern transfer.
- Protect sensitive data. Apply the data-use agreement to human genomic data, model derivatives, prompts, embeddings and intermediate files. Do not upload controlled material to a generative-AI service unless the agreement and institution expressly allow it.
- Use human-in-the-loop decisions. Require a qualified person to approve ambiguous screening results, unusual synthesis requests, automated design changes and autonomous instrument actions.
- Reassess at each release point. Repeat the review before publication, model-weight release, dataset release, transfer to another group or change in experimental system. New evidence can change the classification.
Laboratory biorisk management is broader than sequence screening
The World Health Organization describes three connected pillars of biorisk management: laboratory biosafety, laboratory biosecurity and dual-use oversight. Biosafety addresses accidental exposure or release; biosecurity addresses loss, theft, misuse or unauthorized access; dual-use oversight addresses work that can be used for both beneficial and harmful purposes.

For an AI-designed sequence, those pillars affect practical decisions such as facility access, inventory control, transport, waste handling, personnel reliability, emergency procedures and supervision. A computationally benign prediction can acquire risk when inserted into a vector, combined with another component or tested in a permissive host. Conversely, a sequence with a concerning annotation may be manageable under a tightly controlled, approved protocol. The decision belongs to the competent institutional review process, not to a sequence label alone.
Human genomic data require additional restrictions
Human sequence data create privacy, discrimination and re-identification risks in addition to biological risks. A March 2025 notice from the NIH Office of Intramural Research states that controlled-access genomic data and derivatives may not be distributed outside approved users without written authorization. It also warns that generative-AI tools can inadvertently disclose protected information.
Before using an AI system with human genomic material:
- Check the data-use agreement and institutional policy for model training, inference, storage and onward transfer.
- Separate identifiers and metadata from sequence data where the approved protocol requires it.
- Do not paste controlled sequences, donor information or uniquely identifying variants into consumer-facing tools.
- Track derivatives, including generated embeddings, prompts, summaries and model outputs that could reconstruct sensitive information.
- Obtain written authorization before sharing controlled data or derivatives outside the approved user group.
How to decide what to publish
Publication does not have to be all-or-nothing. Match the release to the information hazard.
Publish the scientific finding, not the enabling recipe
If the sequence itself would materially lower the barrier to harmful construction, report the biological question, evaluation design, aggregate results and limitations without depositing the exact sequence. Explain that sequence-level details were withheld pending review, where appropriate.

Use controlled access for necessary details
A controlled repository, vetted request process, data-use agreement and named responsible person can reduce exposure while allowing legitimate replication. Access criteria should be decided with the institution, funder and journal rather than improvised after publication.
Review model and dataset releases separately
A paper may be acceptable while releasing model weights, prompts, training data or optimization code is not. Assess each artifact for how it changes the ability to generate, select or build a harmful sequence.
Keep an audit trail
Record the sequence version, provenance, screening output, reviewers, decisions, redactions, access approvals and date of reassessment. This makes later correction possible if screening standards or biological knowledge change.
Independent reader supportYour contribution helps us test, update, and keep practical guides available for everyone.Comparing two candidate sequences or release plans
When choosing between designs, compare the same factors rather than relying on a model’s confidence score.
| Comparison axis | Questions to ask |
|---|---|
| Function | What phenotype is intended, and could it affect replication, toxicity, host range, immunity, delivery or resistance? |
| Pathogen or toxin association | Does either design resemble a known agent or a component that could increase harmful capability? |
| Novelty and uncertainty | How far is the design from characterized sequences, and what experiments are needed to resolve uncertainty? |
| Synthesis accessibility | Can it be ordered, assembled from fragments or produced on available benchtop equipment? |
| Screening result | Was the complete sequence screened, when, by whom and with what outcome? Are there ambiguous regions? |
| Data sensitivity | Does the design include controlled human genomic data, restricted pathogen data or protected model derivatives? |
| Containment and review | What facility, training, supervision and institutional approvals are in place? |
| Release format | Will the public receive a sequence, model weights, prompts, code, a dataset or only high-level findings? |
The safer option is generally the one with a clearer benign purpose, less sensitive provenance, lower synthesis accessibility, stronger screening evidence and a release format that preserves scientific value without providing an actionable construction path.

Common failure modes and responses
“The model says the sequence is benign.”
Model confidence is not a biosafety determination. Obtain independent functional analysis and institutional review; document uncertainty rather than treating a probability or label as clearance.
“The provider’s screen returned no match.”
A negative screen means the tool did not flag the sequence under its rules and database at that time. It does not rule out a novel harmful design or a context-dependent effect. Escalate unusual or uncertain designs for human review.
“Only a short fragment is being shared.”
Fragments can become enabling when combined with other published information. Assess the fragment, its assembly context and the recipient’s ability to reconstruct the full design.
Do these 3 things before closing this tab:
1Repair Windows errors before they cause bigger problems2Scan for outdated or missing drivers - takes under a minute3Clear out junk files and repair common Windows errors“The work is computational, so no committee is needed.”
Computational work can create information hazards even before any DNA is ordered. NIH procedures specifically include manuscript and abstract review for potential dual-use issues. Ask the institutional biosafety or dual-use office what review applies.
“A public release can be corrected later.”
Once a sequence, model or dataset is copied, withdrawal cannot reliably remove it. Resolve access, redaction and security questions before release.
What current guidance can—and cannot—establish
Official guidance from NIST, HHS, UK authorities, WHO, NIH and NSABB supports layered governance: screening, identity checks, institutional oversight, containment, data controls and review of information hazards. It does not establish a universal safe sequence threshold or a validated harmlessness probability for an AI-generated design.
That limitation is itself a safety fact. Treat the absence of a warning as incomplete evidence, preserve qualified records, and revisit the decision as screening standards, model capabilities and biological understanding develop.
