Intelligent drug design combines machine-learning models with medicinal-chemist judgment to search chemical space more deliberately. The approach profiled by Chemistry World in July 2017 centered on Andrew Hopkins, the University of Dundee researcher who founded Exscientia. Its promise was not to replace drug hunters, but to help them choose which molecules to make and test, potentially reducing wasted synthesis and shortening early discovery cycles. The evidence available from 2017–2019 includes a retrospective tadalafil test, a reported 12-month development-candidate program, and pharmaceutical collaborations. Those results are important demonstrations of workflow and commercial interest, not proof that AI-designed medicines will succeed in clinical trials.
What “intelligence amplification” means in drug discovery
In this context, intelligence amplification (IA) means extending the reasoning and pattern-recognition of experienced scientists with software. Machine learning handles large, connected datasets and proposes promising chemical structures; medicinal chemists decide whether the suggestions make biological and synthetic sense. The team then makes selected compounds, tests them, feeds the results back into the models, and repeats the cycle.

Andrew Hopkins summarized the philosophy by saying: “As a company, we are now equally interested in IA – intelligence amplification – as we are in AI.” The distinction matters. A fully autonomous system would imply that software independently defines a target, invents a molecule, validates it and advances it. The workflow described by Hopkins is a human-led loop in which algorithms expand the number and quality of options that scientists can evaluate.
Who Andrew Hopkins and Exscientia are in this story
Hopkins moved from Pfizer to the University of Dundee in 2007. He spun Exscientia out of research in his Dundee laboratory in 2012. The company’s early identity was therefore a hybrid: academic work on computational drug design organized as a commercial organization able to work with pharmaceutical partners.

How the AI-assisted workflow works
The core idea is to make each design cycle learn from the last one instead of treating every molecule as an isolated experiment.
1. Connect the data
Hopkins described building a data warehouse that linked screening results to the compounds tested. Instead of keeping assay outcomes, structures and related measurements in disconnected systems, the warehouse made the chemical space queryable. A scientist could ask how structural changes affected a target or property and use the answer to define the next experiment.
2. Model targets and risks
Machine-learning models used the integrated information to help predict protein targets and implications for efficacy and safety. Such predictions are prioritization tools: they estimate which hypotheses deserve laboratory attention. They do not replace binding assays, cellular tests, animal studies or clinical evaluation.

3. Generate chemically realistic molecules
The design system was trained on medicinal-chemistry data, not only on abstract mathematical representations of molecules. That training was intended to favor structures that chemists could actually synthesize and that might have useful properties. A molecule can score highly in a model yet be impractical to make, unstable, toxic or impossible to optimize; incorporating chemistry experience helps filter out some of those dead ends before synthesis.
4. Select, make and test
Scientists choose a subset of suggestions, synthesize them and measure their activity and other properties. The new results become additional training data. This is an iterative “design, make, test and learn” loop, with human judgment applied at every stage.
5. Refine the next round
Results from the laboratory alter the model’s view of the chemical landscape. The next proposals can focus on potency, selectivity, exposure, safety liabilities or synthetic accessibility, depending on the program’s bottleneck. The value of the system therefore comes from shortening the feedback cycle and improving which experiments are attempted, rather than from generating a single miraculous structure.

What the tadalafil test actually demonstrated
One of the strongest examples in the profile was a retrospective test using historical tadalafil discovery data. The system was given information that would have been available up to the discovery hit, then allowed to evolve hundreds of thousands of candidate structures. Tadalafil emerged as the highest-ranked compound.

This result supports a specific claim: under a reconstruction of the historical information available at that point, the model could navigate chemical space toward a known successful molecule. It is a useful validation because the answer was not supplied as the target output. However, it remains retrospective. The experiment used a known discovery outcome and did not show that a new, unknown medicine would be safe or effective in patients. It is also not equivalent to a prospective clinical trial, regulatory approval or an independently replicated benchmark across many programs.
The reported 12-month development-candidate example
Hopkins also described a program involving a bispecific dual agonist acting on two GPCR subfamilies. He said the team reached a development-candidate stage in 12 months using fewer than 400 molecules. In the same account, he characterized that result as less than a quarter of the time and cost normally expected.
Those figures should remain attributed to Hopkins. The reporting does not provide an independently audited cost ledger, a definition of the comparison set, or a universal industry baseline. “Development candidate” is an important milestone, but it is still an early stage: a candidate must pass further preclinical work, manufacturing development, regulatory review and human clinical testing before anyone can know whether it becomes a medicine.

AI-assisted design versus conventional discovery
Traditional medicinal chemistry already uses computational tools, structure-based reasoning and expert intuition. The difference in the described approach is the degree of integration and iteration.

Why faster molecule selection could matter
Early drug discovery fails frequently because a compound may be weak, selective in the wrong way, unstable, difficult to synthesize or unsafe. Every unproductive synthesis consumes time, materials and specialist attention. A model that helps a team reject poor ideas before making them, or identifies a better-balanced series earlier, could increase the information gained from each laboratory cycle.
Speed alone is not the objective. A very fast program that optimizes only potency can produce molecules with poor exposure or unacceptable toxicity. The workflow’s stated advantage is that efficacy, safety implications and synthetic realism can be considered together, allowing chemists to trade properties deliberately rather than discovering late that a potent molecule cannot become a drug.

Exscientia’s reported partnerships show how computational drug-design companies can commercialize a platform without operating every development program alone. Dundee reported a €15 million investment from Evotec and a strategic collaboration with GSK in 2017. In 2019, Dundee reported a Roche collaboration using the Centaur Chemist platform, with potential consideration of up to CHF 67 million plus tiered royalties.
These arrangements indicate that large pharmaceutical companies saw value in applying the methods to real targets and sharing development risk. They do not establish that every collaboration produced an approved medicine, nor do the reported financial terms describe guaranteed payments. The Roche figure was potential consideration, and the available account does not provide a current status for those agreements.

How reliable are AI-designed drug candidates?
Reliability has several layers, and the evidence should be separated accordingly:
- Data reliability: Models inherit errors, missing values and inconsistent assay conditions in the datasets used to train them.
- Prediction reliability: A high model score is a hypothesis about activity or properties, not a measurement.
- Synthetic reliability: A generated structure must be made, purified and characterized; training on medicinal-chemistry data improves plausibility but cannot guarantee a workable route.
- Biological reliability: In-vitro activity may not translate to exposure, efficacy or safety in animals and humans.
- Clinical reliability: Only well-designed clinical trials and regulatory review can establish whether a candidate benefits patients.
The tadalafil exercise addresses retrospective search performance. The sub-400-molecule, 12-month example addresses an early development workflow. Neither result measures clinical success. A careful reader should therefore treat the approach as a way to choose experiments and molecules more intelligently, not as an oracle that removes uncertainty.
Questions readers commonly ask
Is Exscientia’s approach fully autonomous?
No. The account presents a partnership between machine intelligence and experienced drug hunters. Chemists set objectives, assess synthetic and biological plausibility, select compounds for testing and interpret laboratory results.
Do these 3 things before closing this tab:
1Fix the driver behind crashes, sound loss and screen glitches2Repair Windows errors before they cause bigger problems3Scan for outdated or missing drivers - takes under a minuteDid the tadalafil result prove that AI discovered a better medicine?
No. It was a retrospective reconstruction in which tadalafil ranked highest among hundreds of thousands of generated compounds. That supports the ability to reproduce a known discovery path, not proof of clinical efficacy or superiority to existing medicines.
What does “development candidate” mean in the 12-month claim?
It refers to an early candidate selected for further development after discovery work. Additional preclinical, manufacturing, regulatory and clinical steps remain before approval is possible.
Can the reported speed and molecule count be expected for every drug target?
No. The figures came from one program and were reported by Hopkins. Target biology, assay quality, therapeutic area, safety requirements and manufacturing constraints can all change the duration and number of molecules needed.
