Mitochondrial DNA (mtDNA) testing follows one direct maternal line: your mother, her mother, and the women before her. It can identify a haplogroup and find people with similar mtDNA in a testing company’s database, offering clues about that line. It does not test every branch of your mother’s family, provide a complete family tree, or prove that you descend from a particular named ancestor.
What mtDNA testing actually follows
Mitochondrial DNA is found outside the cell nucleus. In genealogy, its usefulness comes from its pattern of inheritance: both men and women receive mtDNA from their mother, but only women pass it on. A man can take an mtDNA test, but he does not pass his mtDNA to his children. FamilySearch summarizes the pattern this way: “Mitochondria are passed from a woman to all her children, regardless of whether they are male or female.” (FamilySearch)

The line being followed is mother → her mother → her mother’s mother, continuing backward. This is only one thread through a family tree. Your mother’s father, your father’s mother, and other ancestors on your mother’s side are not part of this direct maternal line unless they also happen to be ancestors along that exact chain.

Your maternal haplogroup
A haplogroup places your direct maternal line on a branch of a broad genetic family tree. It can provide context about ancient population history and migration, but it is not an ethnicity estimate for your whole family. A haplogroup or map should not be read as proof that all your ancestors came from one present-day country or belonged to one ethnic group. A provider’s haplogroup map represents broad lineage history; locations reported for individual matches are a different kind of information and depend on what participants provide. Possible matches on the same maternal line
A match means the tested sequences are similar under the company’s matching rules. It is consistent with a shared direct maternal-line ancestor, but the ancestor may be hundreds or thousands of years in the past because mtDNA changes slowly. A match by itself usually cannot identify which person in a documented family tree is shared.
Match lists are also limited by the company’s database and its settings. They do not include every living relative or everyone who shares an ancestor with you. FamilyTreeDNA says testers who have not signed a release or have opted out of matching are not displayed, and it distinguishes matching levels based on which parts of mtDNA were tested. Its closer matching levels are more likely to reflect a more recent maternal-line connection, but they do not give a precise date for that connection. (FamilyTreeDNA)

A result whose detail depends on sequence coverage
Test coverage affects how much sequence can be compared. FamilyTreeDNA describes its mtFull Sequence test as covering HVR1, HVR2, and the coding region; it describes HVR1-only and HVR1/HVR2 results as lower resolution. Broader coverage can make comparisons more informative, but it cannot turn genetic distance into a reliable calendar date. (FamilyTreeDNA)
Matching thresholds are provider-specific, not universal scientific rules. For example, FamilyTreeDNA’s full-sequence matching rules allow up to three differences, including certain heteroplasmy cases, and exclude two high-frequency insertion/deletion locations, positions 309 and 315, from the count. Other providers may use different methods. (FamilyTreeDNA)

What mtDNA testing cannot establish
- Your whole ancestry or ethnicity: the test represents one direct maternal line, not all the lines that make up your family.
- A complete list of relatives: results depend on who tested, what they tested, and whether they appear in the company’s matching database.
- A named shared ancestor from a match alone: similar mtDNA may point to a shared maternal-line ancestor, but matching does not identify that person or establish when the connection occurred.
- Every branch on your mother’s side: the test does not follow your mother’s father or other maternal relatives outside the direct mother-to-mother chain.
- A precise date or generation for a shared ancestor: sequence similarity is not a timestamp.
Which DNA test fits your family question?
Different tests follow different inheritance paths, so the right choice depends on the question. There is no universally best DNA test for every genealogy project.
| Test type | Inheritance path | What it is suited to | Important limit |
|---|---|---|---|
| mtDNA | Direct maternal line: mother to child, then passed on by daughters | Investigating a particular maternal lineage and its haplogroup or matches | Covers one narrow line, not all maternal relatives or the whole family tree |
| Autosomal DNA | DNA inherited across many family lines | Finding and comparing relatives across multiple branches | Does not isolate one direct maternal line in the way mtDNA does |
| Y-DNA | Direct paternal line in people with a Y chromosome | Investigating a particular father-to-son lineage | Follows one paternal line, not the whole family tree |
These tests provide different kinds of evidence; none replaces records when the goal is to identify a specific person or relationship. FamilySearch and the International Society of Genetic Genealogy describe the distinctions and the limited reach of lineage tests. (FamilySearch; ISOGG)

How to use a match in genealogy
- Check what was tested. Confirm whether the result covers HVR1, HVR1 and HVR2, or the full mitochondrial sequence, and learn the provider’s matching rules.
- Compare documented maternal lines. Look for a plausible connection by tracing each person’s mother-to-mother ancestry through records rather than assuming that a match points to a recent or famous ancestor.
- Use relevant testers. If a particular branch is in question, compare people whose documented direct maternal lines represent that branch. A match is a clue to investigate alongside genealogy records.
- Choose another test if the question is broader. For relatives across many branches, autosomal testing is usually a better fit. mtDNA is most useful when the question is specifically about the direct maternal line.
Privacy and consent matter
DNA testing can reveal unexpected family information that may affect relatives as well as the person tested. Get permission before managing or testing another person’s DNA, and review the company’s current consent, matching, and data-use choices before submitting a sample. FamilyTreeDNA documents optional matching settings, but its policies and options can change. (FamilySearch; FamilyTreeDNA)
