Trifluoromethylation adds a trifluoromethyl group (CF₃) to a molecule. The main approaches transfer CF₃ as a nucleophile, an electrophile, or a radical. TMSCF₃—also called the Ruppert–Prakash reagent—is a familiar starting point for nucleophilic chemistry, particularly addition to carbonyl compounds, but the right method depends on the target molecule and the site where CF₃ must go.
What trifluoromethylation does
A trifluoromethyl group consists of one carbon bonded to three fluorine atoms: CF₃. In a trifluoromethylation, a reagent or reaction sequence installs that group at a chosen position on a molecule. The transformation can change a molecule’s properties, but the practical question is more specific: which site can be modified, and which transfer strategy fits its chemistry?

Synthetic methods are commonly grouped by the behavior of the CF₃ source: nucleophilic, electrophilic, or radical transfer. These are broad reaction families, not interchangeable recipes; substrate type, selectivity goals, conditions, and handling all influence the choice. A methods review surveys the three-way classification and the development of these approaches: Chemical Reviews: Trifluoromethylation of Organic Compounds.
Three ways to transfer CF₃
Nucleophilic transfer: TMSCF₃
Trimethyl(trifluoromethyl)silane, commonly written TMSCF₃ and known as the Ruppert–Prakash reagent, is a widely used nucleophilic CF₃ source. One prominent application is addition to carbonyl compounds, which can form a carbon–CF₃ bond at the carbonyl carbon. In many common transformations, TMSCF₃ requires activation, often by a fluoride source; the details depend on the reaction system, so this should not be treated as a universal mechanism or condition set. MilliporeSigma’s technical overview of the Ruppert–Prakash reagent discusses its nucleophilic role, while a review of asymmetric methods describes commonly used activation cycles and carbonyl applications: Chemical Reviews: Enantioselective Trifluoromethylation of Ketones.

The useful shorthand is not “TMSCF₃ works on anything,” but “TMSCF₃ is a recognizable option when the target and reaction design call for nucleophilic CF₃ addition.” Other substrate classes or desired bond-forming sites may call for another family.
Electrophilic transfer: hypervalent iodine reagents
Hypervalent iodine reagents, including Togni reagent families, offer an electrophilic route to CF₃ transfer. Their reactivity is distinct from nucleophilic TMSCF₃ chemistry, so method selection depends on the substrate and the bond-forming event required. A major review covers this reagent family and its applications through March 2014; its mechanistic discussion reflects the limits of the literature available at that time: Chemical Reviews: Hypervalent Iodine Reagents for Trifluoromethylation.

Radical transfer: selected C–H functionalization
Radical CF₃ sources and redox methods can functionalize C–H bonds in selected arenes and heteroarenes. Photoredox approaches use light alongside a catalyst or other radical-generating conditions. Which position reacts can depend on the substrate’s electronic properties and the reaction design, so a successful example on one arene does not establish broad compatibility. A 2023 study describes visible-light excitation of a Co(III)–CF₃ complex in an arene trifluoromethylation approach: Journal of the American Chemical Society study.
