The “new anti-aging drug” is most likely topical rapamycin, also called sirolimus. A small exploratory human trial reported changes in skin-aging markers and improvements in appearance for some participants, but it did not prove that rapamycin reverses skin age or establish it as an approved anti-aging treatment.
What drug is the headline referring to?
Rapamycin, also known as sirolimus, is a drug that inhibits the mTOR pathway. It is not a newly established skin-rejuvenation medicine. The human evidence behind claims that it may affect aging skin is a single exploratory study published in GeroScience in 2019: Chung and colleagues’ trial.

What did the human study test?
The Drexel University College of Medicine team enrolled 36 people older than 40 who had signs of photoaging and loss of dermal volume. Participants applied a topical rapamycin formulation to the back of one hand and a placebo to the other every 24 to 48 hours.

Nineteen participants discontinued or were lost to follow-up, leaving 17 who completed the study. Tissue analysis was possible for only a subset of those completers, so the biological findings came from fewer people than were originally enrolled.

What changed—and what did not?
Measured skin markers
The researchers reported a reduction in p16INK4A protein, a marker associated with cellular senescence, with P = 0.008. They also reported an increase in collagen VII, a protein important to the skin’s basement membrane, with P = 0.0077. These are findings from this study; they are not percentages of rejuvenation or a quantified reduction in skin age.
Visible appearance
The authors described clinical improvement in skin appearance for many of the participants who completed the study. That observation does not establish how large or durable the improvement was for a typical user, and it is distinct from the tissue-marker results.

What the findings cannot show
The study does not demonstrate whole-body rejuvenation or prove that topical rapamycin reverses biological age. Its exploratory design, small number of completers, substantial attrition, and limited tissue samples make it difficult to generalize the findings. The evidence presented here does not establish long-term safety, broad effectiveness, or independent replication.
Is topical rapamycin approved or widely available for anti-aging?
The evidence here does not establish current regulatory approval or commercial availability of topical rapamycin for anti-aging use. The 2019 trial is evidence of an early research signal, not proof of a generally approved treatment. Do not infer that a formulation sold or prepared for another purpose has the same evidence or safety profile as the formulation used in the study.

How to interpret the “turn back the clock” claim
“Turn back the clock” suggests a proven reversal of aging. The study supports a narrower statement: in an exploratory trial, researchers observed changes in two skin-related markers and reported visible improvement in some completers. Larger studies with better retention, fuller tissue analysis, and independent replication would be needed to establish whether those effects are reliable, meaningful, and safe over time.
