Shaydullin and colleagues used sealed glass capillaries to isomerize N-substituted pyrazoles at temperatures up to 500 °C, reporting yields up to 50% and reaction times as short as five minutes. The result establishes a striking example of high-temperature solution chemistry, not a general method for running arbitrary organic reactions at 500 °C.
What the study demonstrated
The primary study, by Shaydullin, Galushko, Ilyushenkova, Vlasova and Ananikov, appeared in Chemical Science in 2025. Its title asks whether activation barriers of 50–70 kcal mol−1 are accessible for transformations in solution. The authors investigated that question through isomerizations of N-substituted pyrazoles, rather than surveying many reaction classes. Read the study in Chemical Science.

The authors report reactions at temperatures up to 500 °C, product yields up to 50%, and reaction times as short as five minutes. Their conclusion gives an experimental activation energy of 50.7 ± 2.7 kcal mol−1. These are results for the reported system; they are not expected yields, times, or activation energies for other molecules or procedures.

How the sealed-capillary method worked
The reaction mixture used small quantities of reactants and solvent sealed inside glass capillaries. Chemistry World describes the vessels as borosilicate Pasteur pipettes sealed after filling. The researchers heated the vessels using induction, microwaves, or a muffle furnace. Chemistry World’s report describes the setup and its solvent observations.

Solvent and vessel compatibility mattered
At the tested high-temperature conditions, Chemistry World reports that water, dimethyl sulfoxide, and pyridine caused capillaries to rupture. Aromatic or saturated hydrocarbons were more compatible in the reported experiments, and p-xylene was identified as the optimal solvent among those tested. This is not a general compatibility chart: the result does not establish that every hydrocarbon or every glass capillary is suitable for a high-temperature sealed reaction.
What 500 °C does—and does not—mean
The finding challenges a blanket assumption that solution-phase organic chemistry cannot be performed at very high temperatures. It does not show that 500 °C is a broadly useful operating temperature for synthesis. The demonstrated chemistry was a particular N-substituted pyrazole isomerization, with solvent choice and vessel integrity central to the procedure.

Nor does the report establish broad substrate tolerance, universal safety, or successful scale-up. Chemistry World describes scale-up as a future goal for corresponding author Valentine P. Ananikov, not as an outcome already demonstrated. Jaime Portilla, an expert in pyrazole-derivative synthesis at the University of the Andes, characterized the tested reactions as “very specific.”
How to interpret the result alongside conventional solution synthesis
Rather than replacing conventional solution-phase synthesis, the method expands the conditions that have been shown to work for one model transformation. Its significance is proof of feasibility under constrained conditions; whether it helps with another synthesis depends on the chemistry and the pressure and compatibility limits of the complete setup.

Why the result is useful
For chemists, the study provides an experimental example that exceptionally high activation barriers can be accessible in a solution-phase transformation under specially chosen conditions. For readers outside the field, the important distinction is between “possible in this demonstrated setup” and “generally practical.” The paper supports the first claim; it does not establish the second.
