A self-assembling tripeptide increased conversion in one laboratory Michael addition, but the result is not a general 74% speed increase. In a 2021 study, D-Pro-L-Phe-L-Phe (D-PFF) formed fibrils in phosphate-buffered saline (PBS); at 35 °C, the reaction reached 74% conversion in PBS, compared with 41% in water and 56% with a non-fibril-forming peptide analogue. The study found no significant change in enantiomeric excess.
What the researchers tested
Sinibaldi and coauthors designed minimal tripeptides that combine an organocatalytic component with the ability to self-assemble. They tested D-Pro-L-Phe-L-Phe, abbreviated D-PFF: proline provides the organocatalytic functionality, while the phenylalanine-containing sequence supports formation of fibrils.

The benchmark reaction was a Michael addition of isovaleraldehyde to β-nitrostyrene. The authors chose the relatively low-reactivity aldehyde partner to make a potential catalytic enhancement easier to detect. They reported D-PFF fibrils in PBS and in HFIP/water; the L-PFF analogue and D-PF derivative did not form the same fibrillar structures in the comparisons described.

At 35 °C, the paper reports 74% conversion for D-PFF in PBS, 41% in water, and 56% for the non-fibril-forming L-PFF comparison. These are conversion measurements under the reported conditions—not reaction-rate increases or yields that can be assumed for other substrates or setups.

| Reported condition at 35 °C | Conversion | How to interpret it |
|---|---|---|
| D-PFF in PBS, where fibrils formed | 74% | Reported conversion in the fibril-forming condition |
| D-PFF in water | 41% | Reported conversion in the water comparison |
| Non-fibril-forming L-PFF comparison | 56% | Reported conversion with the analogue that did not produce the same fibrillar structures |
The authors also tested uncatalyzed reaction conditions, PBS alone, and non-fibril-forming peptide analogues. They report that PBS alone did not explain the enhancement. The paper notes that temperature and substrate equivalents also affected conversion, so the headline figures should be read as a comparison within the reported experimental setup, not as an isolated universal effect of fibrils.
For the reported experiments, conversion was measured by ¹H NMR; diastereomeric ratio was assessed by ¹H NMR analysis of the crude mixture; and enantiomeric excess (ee) was measured by HPLC using a chiral stationary phase. The study includes a 5 mol% catalyst condition among its best results, but comparisons across entries can also reflect differing reagent concentrations and other conditions.

Did the fibrils improve selectivity?
Not significantly, according to the study. The reported increase is in conversion or catalytic activity under the tested conditions, not a demonstrated improvement in ee. The authors also report that raising temperature and substrate equivalents improved conversion without significantly affecting ee.

The authors propose that the assembled fibrils create a more organized, lipophilic environment that helps the catalytic reaction. Their controls support an effect associated with the fibril-forming D-PFF condition rather than PBS alone, but the proposed environment is a rationale, not a fully established molecular mechanism.

What the result does—and does not—show
The paper describes the work as a proof of concept for a simple fibril-forming tripeptide organocatalyst with higher activity in its supramolecular state. It shows that changing the assembly state of this particular peptide was associated with higher conversion in one benchmark reaction.
- It does not show that every self-assembling peptide will accelerate every Michael addition.
- It does not establish broad substrate performance, manufacturing scale-up, or an industrially validated catalyst.
- It does not demonstrate a lifecycle or overall green-chemistry advantage. Aqueous reaction conditions are relevant to that discussion, but a full process assessment would be needed to establish such a benefit.
In a 2021 Chemistry World report, biocatalysis expert Kate Adamala described aqueous media and substrate tolerance as milestones for reducing reliance on oil-based reagents and meeting green-chemistry goals. That is expert commentary on potential significance, not a lifecycle assessment of the D-PFF study. Broader applications remain prospective; the authors identify development of other catalytic fibrils as future work.
