How to Tell If a Microscope Image Has Been Altered or Misrepresented

Repeated landmarks, seams, or selective tonal changes can flag a microscope image for review. Original files, metadata, and processing disclosures are needed to assess what happened.

To tell whether a microscope image may have been altered, compare distinctive details across panels, inspect seams and tonal changes, and look for selective edits that hide or invent evidence. These clues warrant questions, but they do not prove misconduct. The strongest check is whether the figure can be traced to original image files, acquisition metadata, and a clearly described processing workflow.

How can I tell if a microscope image has been altered?

Start with the claim the figure makes: are panels supposed to show different samples, fields, times, or experimental conditions? Compare details that are unlikely to recur in the same arrangement—not just common biological shapes. Then consider routine transformations such as cropping, resizing, rotation, mirroring, or color mapping. Similarity that persists after accounting for these changes may mean panels share a source, but the original files and context are needed to interpret it.

  • Compare cell outlines, vessel branches, organelle shapes, debris, scratches, speckles, and background texture.
  • Look at the exact spatial relationships among several distinctive landmarks, not one generic feature.
  • Check whether the suspected match could be explained by a crop, resize, rotation, flip, or different color mapping.

A low-resolution screenshot can obscure details and introduce compression artifacts. Treat it as a reason to request better evidence, not as a forensic determination.

Which visual clues deserve closer inspection?

Repeated structures or backgrounds

Identical arrangements of cells, organelles, scratches, or background marks in panels labeled as different samples or fields may indicate reuse. A panel may have been repositioned or mirrored, so compare landmarks after allowing for those transformations. A repeated image is not automatically proof of wrongdoing: a control may legitimately be reused under a study design or journal policy, and figure assembly errors can happen.

Cloned, stamped, or patched areas

Repeated small structures, abruptly different texture, rectangular patches, or an unusually clean region can justify closer examination. Skilled cloning may not be apparent to the eye and can require image analysis to reveal. The U.S. Office of Research Integrity (ORI) says retouching tools are inappropriate for scientific images; adding objects that were not present can constitute fabrication or falsification. See ORI’s Guideline #8 – Cloning Degrades Data.

Seams and composites

A straight boundary, abrupt change in background noise, or discontinuity in texture can suggest that separately acquired images were joined. A composite is not necessarily improper, but its parts should be visibly demarcated and explained. Nature Portfolio says images from different times or locations should not be combined as one image without stating that they were combined; when juxtaposition is essential, boundaries should be marked and described. Those are Nature Portfolio’s editorial requirements, not universal law. Its current image integrity and standards policy explains the rules.

Selective brightness, contrast, or color changes

Brightening, darkening, thresholding, or changing an individual color channel can make a signal stand out—or cause relevant features to disappear. Nature Portfolio’s policy says adjustments should be applied to the whole image and warns against thresholds, expansion or contraction of signal ranges, and alteration of high signals. It calls for disclosure of pseudocolor, nonlinear adjustments such as gamma changes, and necessary individual-channel adjustments in merged images. ORI likewise cautions that contrast changes can hide relevant features and that color manipulation is inappropriate when color carries information central to the conclusion. See ORI’s Questionable Practices.

Crops, scale, and distorted shape

A crop can be acceptable if it does not change the inference or remove context needed to interpret the remaining image. A changed aspect ratio distorts shapes, while downsampling can introduce pixelation or other artifacts. ORI’s guidance puts the issue this way: “The line dividing good editing from bad practices is crossed when an image is cropped in a way that changes the context of what remains.”

What is acceptable image processing, and what is concerning?

Microscope images are often processed for presentation or analysis. Processing alone does not make a figure misleading. The important distinctions are whether the processing preserves the observation, is applied consistently, and is disclosed when it materially changes how the image appears.

Check More consistent with fair presentation Potential concern
Adjustment Whole-image changes that preserve relevant features and are applied consistently to comparable images. Selective edits that hide, add, or emphasize evidence, or make important features disappear.
Comparability Experimental and control images handled using a consistent, stated workflow. Different or unexplained processing that makes conditions appear more distinct or alike.
Composite Joined parts are visibly separated and the figure legend explains the assembly. A seamless join presented as one continuous acquisition, especially across different times or locations.
Traceability The figure can be compared with source images, acquisition metadata, and the stated methods. No clear path from the displayed panel to the original file or processing steps.

Nature Portfolio states: “Digital images submitted with a manuscript for review should be minimally processed.” Its policy also recommends retaining source images and metadata. Requirements vary by journal, so check the relevant publication’s current instructions.

How to check a suspicious figure step by step

  1. Identify the claim. Note which panels are said to represent different fields, samples, times, or conditions, and record the labels and figure legend.
  2. Compare landmarks. Look for multiple distinctive details and their spatial relationships. Account for crop, resize, rotation, mirroring, and color mapping before judging a possible match.
  3. Inspect boundaries and tone. Check for seams, abrupt texture or noise changes, patched regions, and local brightness or color differences.
  4. Read the methods and legend. Look for the acquisition and processing software and settings, and for descriptions of projections, filtering, thresholding, deconvolution, rendering, composites, or other transformations.
  5. Request source material when needed. Compare the published panel with original, unprocessed image files and acquisition metadata—not only a compressed figure. Relevant records can include instrument and acquisition details, bit depth, image dimensions, experimental conditions, and display lookup tables.
  6. Seek an accountable review if the concern remains material. Provide the specific panels and explain the basis for comparison through the journal’s correction or research-integrity process rather than making a public accusation based on appearance alone.

What duplication statistics do—and do not—show

Bik, Casadevall, and Fang’s 2016 study screened biomedical publications and classified 782 papers as containing at least one inappropriate image duplication. Within that flagged set, the authors classified 230 papers (29.4%) as simple duplications, 356 (45.5%) as duplications with repositioning, and 196 (25.1%) as duplicated figures with alteration. These percentages describe only the 782 papers identified under the study’s screening criteria; they are not estimates of how often microscope images generally are altered. The authors also noted that simple duplication can arise from honest assembly mistakes and that intent could not be definitively determined. Read the study in mBio.

What a visual anomaly cannot establish

A similar-looking panel, an abrupt seam, or an unusual tonal change is a lead to investigate—not proof that someone deliberately manipulated an image. The same appearance can have innocent explanations, including allowed reuse, ordinary transformations, or a figure-preparation mistake. Stronger conclusions require the source data and an accountable review of the publication record. Use cautious descriptions such as “appears similar,” “may share a source,” or “requires explanation.”
