How to Measure Carbon Quantum Dot Emission Spectra and Quantum Yield

A practical guide to measuring carbon quantum dot emission spectra and reporting reproducible photoluminescence quantum yields by relative or integrating-sphere methods.

Measure a carbon quantum dot (CQD) emission spectrum at a stated excitation wavelength and under documented instrument and sample conditions. Determine photoluminescence quantum yield (PLQY) either by comparison with a reference of known yield or with an integrating sphere. In either case, report the method, spectral integration range, absorbance, blank, and corrections: a peak wavelength or peak height alone is not enough to reproduce the result.

What an emission spectrum tells you

A fluorescence spectrum is conditional, not a single intrinsic curve for a material. It depends on the excitation wavelength, the sample’s state and concentration, the solvent or matrix, and the instrument settings and wavelength range collected. State those conditions whenever you present or compare spectra.

Use a calibrated fluorimeter or spectrofluorometer. Blank with the solvent or matrix used for the sample, hold excitation and acquisition settings fixed, and collect emission over a range that captures the relevant band. Choose an excitation wavelength based on the sample’s absorption and the comparison you intend to make; there is no universal CQD setting. Published examples include excitation at 365 nm with an emission scan from 385 to 850 nm, and measurements at 320 nm for a pH series. These are study-specific choices, not default settings for every sample (carbon-dot photophysics study; Scientific Reports study, 2025).

Record the conditions that shape the curve

  • Excitation wavelength and emission scan limits.
  • Slit widths, detector or gain settings, and whether the instrument’s spectral correction was applied.
  • Sample concentration, solvent or matrix, cuvette path length and geometry, and blank composition.
  • Any sample treatment or state relevant to the measurement, such as pH when comparing a pH series.

For comparisons, measure samples with the same excitation wavelength, acquisition settings, sample handling, and spectral correction. Use a corrected spectrum and integrate emission over a stated wavelength band for a PLQY calculation. Do not replace the integrated signal with peak height unless the particular method validates that substitution.

Choose a PLQY method

There are two common routes. A relative measurement compares the sample with a fluorescent reference whose PLQY is known under applicable conditions. An integrating-sphere measurement determines PLQY absolutely in principle, without that fluorescent reference. The relative route requires careful matching and absorbance measurements; the sphere route requires a suitable instrument configuration, blank, and corrections (relative-method study; carbon-dot/sol-gel study; JASCO integrating-sphere application note).

Method External fluorescent reference Main practical considerations
Relative comparison Yes; report the standard, solvent, adopted PLQY, and excitation conditions. Use optically dilute sample and reference solutions, matched acquisition conditions, absorbance at excitation, integrated emission, and the refractive-index correction required by the chosen equation.
Integrating sphere No. Document sphere and instrument configuration, sample and blank setup, excitation wavelength, correction procedure, and calculation method. Geometry and blanking need particular care for scattering samples or suspensions.

Measure PLQY by the relative method

  1. Select and document a reference. Quinine sulfate in 0.1 M H2SO4 is used in published CQD examples. Record the standard’s solvent, the reference PLQY you adopt, and the excitation wavelength associated with that value. The reference and sample conditions must be appropriate for the calculation you use.
  2. Prepare dilute concentration series. Prepare several concentrations of both CQD sample and reference. Measure absorbance at the chosen excitation wavelength. Keep absorbance low to limit reabsorption and inner-filter effects; cited protocols commonly use absorbance below 0.1, including at the excitation wavelength. Treat that as a practical protocol recommendation, not a universal metrology limit (Scientific Reports study, 2025; surface-state-modulated carbon-dot study; microwave-assisted carbon-dot study).
  3. Collect matched emission spectra. Blank each solution with its solvent or matrix. Use the same excitation wavelength and matched instrument settings for sample and reference, and apply the same spectral-correction approach. Define the emission integration range and use corrected, integrated emission rather than peak height.
  4. Calculate from a matched pair or, preferably, concentration-series slopes. For one matched sample/reference pair, a commonly used expression is Φx = Φr × (Ix / Ir) × (Ar / Ax) × (nx / nr)2. Here Φ is PLQY, I is integrated emission, A is absorbance at the excitation wavelength, n is the solvent refractive index, and subscripts x and r identify sample and reference. Follow one cited method consistently: publications may define corrections or equation forms differently, so do not mix a formula with incompatible intensity or absorbance definitions (Scientific Reports study, 2025; relative-method study).

A slope method reduces reliance on any one concentration pair. Plot integrated emission against absorbance for multiple dilute concentrations of each solution, fit the linear region, and calculate the sample-to-reference slope ratio with the reference PLQY and refractive-index correction. A published CQD protocol derives its value from concentration-series slopes (microwave-assisted carbon-dot study).

Measure PLQY with an integrating sphere

An integrating sphere provides an absolute route: the calculation compares emitted photon signal with excitation light absorbed by the sample, using the instrument’s blank and correction procedure. Carbon-dot studies report commercial integrating-sphere accessories, and an instrument application note describes measuring incident light with no sample in the holder (carbon-dot/sol-gel study; JASCO application note).

Follow the validated protocol for the sphere and sample type. In particular, scattering samples and suspensions can make geometry and blanking consequential. Report the sphere and instrument configuration, sample and blank arrangement, excitation wavelength, corrections, and calculation method so the absolute measurement can be interpreted and reproduced.

Choose the reference value carefully

Quinine sulfate is not adequately described by an unqualified statement such as “54%.” The cited publications tie their adopted values to differing study conditions: 54% at 350 nm excitation in a 2025 Scientific Reports carbon-dot study; 54% at 360 nm in a 2021 carbon-dot study; and 54 ± 0% in a 2022 study. These are reported study-specific values, not interchangeable universal constants (2025 study; 2021 study; 2022 study).

Use a reference value that matches the standard, solvent, and excitation conditions of the method you follow, and cite the source of the adopted value. Do not transfer a value reported at one excitation wavelength to another without an appropriate basis.

Cuvettes, blanks, and setup checks

A protocol for absolute CQD PLQY specifies a UV quartz cuvette with a 10 mm path length and ethanol in a quartz cuvette as the blank (Bio-protocol method). This is an example, not a universal cell requirement. Before measuring, verify that the cuvette transmits at the excitation wavelength, is compatible with the solvent, and fits the instrument holder and measurement geometry.

  • Confirm the blank matches the sample’s solvent or matrix and the instrument’s measurement arrangement.
  • Check absorbance at excitation and reduce concentration if optical density is too high for the chosen protocol.
  • Ensure the emission scan includes the band being integrated and that sample and reference use equivalent settings.
  • Record whether spectra are corrected and exactly which wavelength interval was integrated.

What to report with a result

A useful report lets another researcher distinguish a sample property from an acquisition choice. Include the following information for each spectrum and PLQY result:
