Scientists use giant-clam shells as dated archives: they read growth increments to build a timeline, sample carbonate along that timeline, and analyze chemical signals such as oxygen isotopes and trace-element ratios. Those signals can help reconstruct past ocean conditions, including temperature, but they are calibrated proxies—not direct readings of ancient seawater. Biology and shell structure can affect the chemistry, and giant-clam Sr/Ca in particular can vary with the daily light cycle.
How does a giant-clam shell preserve a record?
Giant clams add shell material as they grow. The shell therefore contains both visible growth patterns and carbonate whose chemical composition can be measured. Sclerochronology—the study of growth patterns in accretionary hard tissues—uses those patterns to establish when shell material formed.

Researchers can section and polish a shell, image its growth increments, and measure their spacing. They then align the shell’s physical timeline with a chemical sampling path, or transect, along the growth axis. This alignment matters: a chemical value becomes useful for climate reconstruction only when researchers can relate it to when that part of the shell grew.
Building the timeline
In a 2018 study of Tridacna maxima, researchers prepared and polished sections along the shell’s maximum growth axis, imaged the increments, and matched the growth record to the chemistry transect. They counted increments inward from the shell edge, where the collection date was known, and treated them as daily increments to assign dates to geochemical samples. That is one study’s age-model approach, not a guarantee that every shell increment in every specimen represents exactly one day.

Fossil shells need age control as well as an assessment of preservation. A 2025 review of giant-clam sclerochronology identifies fossil dating and proxy reliability as continuing areas of attention.
What do scientists measure to estimate past ocean conditions?
Researchers combine the chronology with measurements of shell chemistry. Different signals can address different environmental questions, and no single ratio serves as a universal temperature gauge.

The 2025 review also discusses δ13C, Ba/Ca, and Fe/Ca among the signals used in giant-clam reconstructions. Which measurements are useful depends on the question; a fine-scale elemental pattern investigated for daily variability is not automatically comparable to an oxygen-isotope record used to infer temperature.

Methods depend on the proxy and the scale of the question. In the 2018 T. maxima study, researchers measured δ18O and δ13C with a stable isotope ratio mass spectrometer and measured Ba/Ca along a transect using laser-ablation inductively coupled plasma mass spectrometry (LA-ICP-MS). They aligned those chemical results with the shell’s growth increments.

That study reported external long-term analytical reproducibility, assessed through replicate NBS 19 measurements, of 0.03‰ for δ18O and 0.02‰ for δ13C as 1σ standard deviations. These describe the instrument-analysis reproducibility reported in that study, not the total uncertainty of a temperature reconstruction or a universal error range for giant-clam shells.
Why Sr/Ca needs a biological caveat
Sr/Ca is a ratio of strontium to calcium in shell material. It can be investigated as an environmental proxy, but giant-clam studies show that it can also vary over the daily light cycle. That makes it important to distinguish the measured pattern from the cause researchers infer for it.

In a 2012 study of a cultivated giant clam, researchers measured Sr/Ca, Mg/Ca, and Ba/Ca with a high-resolution secondary-ion mass spectrometer at 2 μm resolution. They reported diurnal variation in Sr/Ca and proposed that light-enhanced calcification and elemental transport involving the clam and its symbiotic algae could help explain the pattern. The proposed mechanism is an interpretation of the observations, not a reason to treat every Sr/Ca change as a temperature signal.

What did the fossil-shell example show?
Hori and colleagues studied a fossil Tridacna gigas collected at Shiraho Coast on Ishigaki Island in southern Japan. Radiocarbon dating placed it in the middle Holocene. They measured Sr/Ca along a longer profile at 50 μm resolution and used 2 μm resolution for fine-scale analysis.

The fossil shell showed repeating Sr/Ca variation resembling patterns in cultivated giant clams. The authors interpreted fine alternating bands as potential evidence of daily light-cycle variation at a multi-hour scale. This example demonstrates how a fossil shell can be examined for fine-scale patterns; it does not establish that Sr/Ca directly measures past ocean temperature in all giant-clam shells.
How strong is a temperature reconstruction?
A shell-based reconstruction is an inference built from several connected steps: establish the shell’s age and growth chronology, measure selected chemical signals, and interpret those signals using appropriate calibration. The strength of the conclusion depends on the quality of each step, not just on the precision of a chemical measurement.
- Chronology: How were growth increments identified and dated, and how secure is the fossil’s age?
- Sampling: What species and shell region were sampled, and at what spatial resolution?
- Proxy and calibration: Which chemical signal was measured, what environmental variable is it being used to infer, and what calibration supports that interpretation?
- Biology and structure: Could growth processes, symbiotic algae, or shell microstructure contribute to the observed pattern?
- Preservation: Is the fossil chemistry sufficiently preserved for the intended interpretation?
These considerations also explain why records should not be compared solely because they come from giant clams. Species, location, shell age, growth rate, chronology, sampling resolution, measured proxy, calibration against modern or instrumental observations, and preservation can all affect what a record supports. The 2025 review, which synthesizes literature published from 1986 through 2023, describes giant clams as promising high-resolution palaeoenvironmental archives while emphasizing ongoing needs around calibration, reliability, and fossil dating.
