How Scientists Study Cell Adhesion in the Lab

Scientists combine imaging and force measurements to study where cells attach, how adhesions change and how cells interact mechanically with their surroundings.

Scientists study cell adhesion by combining microscopy, which shows where adhesive structures form and how they change, with force measurements, which quantify mechanical interactions. Traction force microscopy estimates forces a cell transmits to its substrate; atomic force microscopy (AFM) single-cell force spectroscopy measures forces as one cell contacts and detaches from a surface. These methods answer different questions, so the right choice depends on what the experiment needs to reveal.

What cell adhesion experiments reveal

Cell adhesion is the attachment of a cell to another cell or to its surroundings, including the extracellular matrix (ECM). Experiments can examine the structures and molecules involved, the timing of their assembly and disassembly, or the forces associated with an interaction. These are complementary forms of evidence: a force measurement does not by itself show the full biological mechanism, and an image alone does not quantify the force involved.

Adhesion is closely connected to the cytoskeleton and cell behavior. During migration, many cells form adhesions near the front, couple them to actin, generate traction and disassemble adhesions toward the rear. Adhesions can also participate in sensing substrate mechanics and signaling. The precise pattern varies by cell and context; there is no single universal migration cycle. Parsons, Horwitz and Schwartz review the relationship between adhesion, cytoskeletal dynamics and cellular tension.

How microscopy shows adhesive structures

Microscopy lets researchers locate adhesive structures in cells and investigate which molecules associate with them. Depending on the imaging approach, they can follow components in living cells to study exchange and changes over time, or relate structure to cell behavior. Some approaches can also perturb actin-based structures locally.

Imaging is useful when the central question is where adhesions form, what they contain, or how they change during a process such as migration. It is not interchangeable with a mechanical measurement: observing an adhesive structure does not, on its own, establish how much force it transmits. How traction force microscopy estimates cell-generated forces

Traction force microscopy (TFM) estimates forces a cell exerts on a compliant substrate by measuring how the substrate deforms. In bead-based implementations, fluorescent beads embedded in the substrate shift as the cell pulls; researchers image their displacement and use computational analysis to estimate cellular traction. The result is an estimate derived from substrate deformation, not a direct reading from a force sensor attached to the cell.

Implementation matters: the substrate, imaging and analysis determine what a particular TFM experiment can resolve. In one specific STED-TFM protocol, Colin-York, Eggeling and Fritzsche use functionalized polyacrylamide gels loaded with fluorescent beads, STED images and open-source analysis software. That protocol reports spatial resolution up to 500 nm and a preparation, acquisition and analysis workflow taking 2–3 days; neither figure is a general specification for all TFM setups. Three-dimensional TFM is also an evolving methods area. A perspective published online in 2025 and assigned to the 2026 issue addresses guidance for 3D TFM, but its detailed recommendations are not established here. See Barrasa-Fano and colleagues’ perspective.

How AFM single-cell force spectroscopy measures attachment

In AFM-based single-cell force spectroscopy, an atomic force microscope (AFM) force probe is used to bring an individual cell into contact with a substrate and then measure forces during detachment. The substrate can be an ECM protein or another cell, depending on the experiment. Unlike ordinary fluorescence imaging, this approach records mechanical interactions during contact and separation; broader AFM force-spectroscopy applications can also examine cell-surface receptors and adhesive or mechanical properties at different scales.

A Nature Protocols example examines integrin-mediated adhesion of HeLa cells to collagen type I. Its procedure includes functionalizing an AFM cantilever with concanavalin A, preparing collagen-coated supports, attaching and handling a cell on the cantilever, measuring adhesion forces and analyzing the data. The authors say the protocol can be modified for other cell lines and ECM proteins, and give 2–3 days as its completion time. Those details apply to that protocol, not every AFM experiment. Choosing a method for the biological question

Question Suitable approach What it measures or reveals
Where do adhesions form, what components associate with them, and how do they change? Microscopy suited to the structure and time scale Location, molecular association and dynamics in cells.

These approaches are not a ranking. Select based on the scale of interest—adhesion structure, whole-cell interaction or molecular bond—and whether the experiment needs dynamic observation or an endpoint measurement. Also consider substrate or probe preparation, equipment access, spatial and force resolution, and the expertise needed to analyze the data. The cited sources do not establish comparable prices, throughput or head-to-head performance across these platforms.
