Scientists can screen inactive molecules as potential partners for an active pharmaceutical ingredient (API), looking for combinations that form a cocrystal. A 2011 approach proposed ranking candidates by calculated hydrogen-bond strengths and other electrostatic interactions—an early filter, not proof that a cocrystal will form or improve a medicine.
What “speed dating” means in pharmaceutical chemistry
In this metaphor, the API is looking for a molecular partner: an inactive molecule that can crystallize alongside it. The resulting solid is called a cocrystal. Changing an API’s crystal form may alter pharmaceutical properties and potentially affect drug delivery, but the concept alone does not establish a benefit for any particular medicine or patient.

Finding suitable partners experimentally can take time. The 2011 account also describes computationally evaluating full crystal structures atom by atom as potentially laborious, motivating a simpler way to rank candidates before experimental work.

How the proposed screening approach ranked candidates
The approach calculated hydrogen-bond strengths and other electrostatic interactions between the API and candidate partner molecules. Those interaction energies were used to help identify pairings worth investigating, rather than to establish a finished solid form.

The method depended on simplifying assumptions: that the relevant interactions would remain similar between calculations for molecules in the gas phase and their behavior in a solid crystal, and that electrostatic interactions would matter more than crystal packing. These are model assumptions for screening, not universal rules about how crystals form.
What a favorable calculation can—and cannot—tell you
- It can help prioritize candidates. A calculated interaction provides a clue about which partner molecules may merit experimental attention.
- It does not establish that a cocrystal forms. The proposed ranking is not a substitute for experiments that determine whether the intended solid form actually exists.
- It does not establish pharmaceutical or clinical improvement. Even if a cocrystal forms, relevant pharmaceutical testing is needed to determine its properties; the cited account does not show improved clinical outcomes for a specific pairing.
In practical terms, the proposal aimed to make candidate selection less laborious, not to eliminate solid-form experiments or demonstrate that every promising molecular interaction produces a useful drug product.

How this fits among ways to find cocrystal partners
The 2011 account presents experimental searching and computational analysis as different sources of effort: experiments can be time-consuming, while calculating full crystal structures may be laborious. The simplified interaction-based approach sought to reduce the computational burden by ranking pairwise interactions instead of relying on atom-by-atom crystal-structure calculations.

The Royal Society of Chemistry’s Chemical Science blog associated the work with Chris Hunter and colleagues at the University of Sheffield, the University of Barcelona, and AstraZeneca. The cited 2011 coverage describes a proposed screening approach and potential ways cocrystals might alter pharmaceutical properties. It does not establish current validation, adoption, regulatory status, or commercial availability.
