How Regioselective Nitrogen Insertion Edits Arenol Rings to Make Benzazepines

A 2024 Chemical Science paper describes a one-pot, regioselective route from selected arenols to benzazepines, and explains why substitution at the ortho position limits the method.

A 2024 study reports a one-pot way to insert nitrogen into selected arenol ring skeletons, producing benzazepines. The reaction uses a dearomative azidation and aryl-migration sequence, but it is not a general edit for any aromatic ring: the arenol’s substitution pattern determines whether it works.

What skeletal editing means in this reaction

Skeletal editing changes the atoms or connectivity within a molecule’s core framework rather than merely adding a substituent to its outside. In the method reported by Yi He, Juanjuan Wang, Tongtong Zhu, Zhaojing Zheng and Hao Wei, an arenol starting material is converted into a benzazepine by inserting nitrogen into the ring skeleton. The paper describes the transformation as regioselective: the insertion occurs at a defined position in the substrates examined.

The study, “Nitrogen atom insertion into arenols to access benzazepines,” appeared in Chemical Science, volume 15, pages 2612–2617. The Royal Society of Chemistry records its first publication on 16 January 2024, DOI 10.1039/D3SC05367A. The article is open access and includes supplementary information with experimental procedures and characterization data.

How the reported transformation works

The authors propose that the reaction proceeds through dearomative azidation, followed by aryl migration. In practical terms, an azide-bearing intermediate forms from the arenol, then a rearrangement changes the ring framework to place nitrogen in the expanded heterocycle. The authors report mechanistic experiments that support this sequence; it is their proposed pathway, not a claim that every mechanistic detail is settled.

#1 Best Overall

For the model reaction, He and colleagues report an 80% isolated yield using TMSN3 as the azide source, CuI (5 mol%), Cy3PO (10 mol%), tert-butyl peroxybenzoate (2 equivalents), and toluene at 120 °C for 12 hours. That figure is the yield for the reported research-scale experiment, not an independently replicated result or a general rate of success.

Which substrates and functional groups were demonstrated

The paper illustrates the method with several arenol frameworks, including naphthol, phenanthrol, tetraphenol and benzo[c]phenanthrenol examples. Its reported examples also include compounds bearing esters, methyl ethers, thioethers, trimethylsilyl groups, aryl halides, nitriles and trifluoromethyl groups. More complex examples include ester-linked steroid and carbohydrate structures, as well as a heteroarene-containing structure.

Rank #2

These examples show what the authors tested; they do not establish that every compound bearing one of those functional groups will be compatible. The authors summarize the breadth as follows: “This protocol exhibited a wide substrate scope, encompassing a variety of multi-arenols, including naphthol, phenanthrol, benzo[c]phenanthrenol, and tetraphenol, along with various functional groups.”

Why regioselectivity comes with a substrate caveat

The method’s scope depends on substitution near the reacting site. In the examined naphthol series, a phenyl group or an electron-withdrawing substituent at the 2-position could support the reaction, whereas a 2-alkyl substituent inhibited it. Chemistry World’s February 2024 account describes the broader limitation reported by the authors: an electron-withdrawing or aromatic group at the ortho position was needed. Corresponding author Hao Wei told the publication the team had not resolved the issue despite nearly a year of effort.

This restriction matters when assessing “wide scope.” The paper demonstrates a varied set of frameworks and functional groups, but the reaction is not shown to accept arbitrary arenols. A candidate substrate should be judged first by its substitution pattern at the relevant ortho position, then by whether its overall structure resembles the successful examples.

How it differs from earlier nitrogen-insertion approaches

Chemistry World notes that some earlier nitrogen-insertion strategies required a nitrogen-containing group, such as an azide, to be installed in advance, while some relied on photolysis. This method combines azidation and migration in a one-pot transformation, starting from arenols and using TMSN3 during the reaction. An expert quoted by Chemistry World identified the absence of photolysis as a potential practical advantage.

That comparison is limited to the approaches discussed in the cited coverage; it does not establish that the new procedure is universally simpler, more scalable or superior. A meaningful method comparison depends on the substrate and substitution pattern, the position selectivity obtained, demonstrated functional-group compatibility, number of operations, and whether a pre-installed nitrogen group or photolysis is needed.

What the study establishes—and what it does not

The result is a synthetic method for making benzazepine structures by editing selected arenol skeletons. The paper demonstrates a model reaction, a range of substrate examples and mechanistic experiments consistent with its proposed pathway. It does not demonstrate a commercial process, universal substrate coverage, or industrial-scale performance. The authors discuss possible relevance to N-heteroarene development and materials chemistry as prospective applications, not established products or uses.
