A 2024 study describes a way to store phosphazene superbases as air-stable carboxylate salts, then release the reactive freebase in solution by adding an epoxide. Opening the epoxide ring creates an alkoxide strong enough to free the superbase; changing the epoxide structure can tune how quickly activation occurs. The approach offers researchers more control over when a strong base appears in a reaction, but the reported examples do not establish universal compatibility or commercial-scale economics.
How does an epoxide activate a superbase?
The method starts with a carboxylate salt of a phosphazene superbase such as BTPP or P2-t-Bu. The authors report that these salts are stable under ambient storage and handling conditions, unlike the reactive freebase, which is generated only after the components are combined in solution.

- Combine the salt and epoxide in solution. The carboxylate attacks and opens the strained epoxide ring.
- Form an alkoxide. Epoxide ring opening produces an alkoxide intermediate.
- Release the freebase. The alkoxide deprotonates the protonated superbase in the salt, generating the active phosphazene base.
The reported approximate values are pKa′ ∼24 in MeCN for the carboxylate and pKa′ ∼43 in MeCN for the alkoxide intermediate — Sujansky, Hoteling and Bandar, 2024. These are solvent-specific values in acetonitrile, not universal acidity or basicity figures that can be transferred unchanged to other solvents. The mechanism and values are described in the 2024 Chemical Science paper.
Why use a salt instead of handling the freebase?
Strong organic superbases can be difficult to prepare, store and handle because the freebase may be sensitive to air. Keeping the superbase in a carboxylate salt form separates storage from use: the salt is the bench-stable starting material, while activation in solution generates the freebase when the reaction needs it. The paper presents the approach as improving preparation, shelf stability, handling and recycling; it does not mean that the activated freebase itself becomes air-stable.

How can the activation timing be controlled?
The epoxide is not just an on-switch. Its structure can alter the rate of ring opening and therefore the timing of freebase generation. This can provide an induction period or mimic slow addition of a strong base, which is useful when a reaction is sensitive to the concentration or timing of base. Study corresponding author Jeffrey S. Bandar described the design principle this way: “It is this modulation of the epoxide structure that allows control of the rate of this reaction.”
This is control over base release within the reported solution chemistry, not evidence that one epoxide gives the same activation profile in every solvent or reaction. Researchers would need to consult the paper’s supplementary information for condition-specific procedures and characterization.

What reactions did the authors demonstrate?
The study reports the salt systems as precatalysts or stoichiometric prereagents in superbase-promoted addition, substitution and polymerization reactions. Demonstrations cited in the paper and coverage include:

These are reported examples, not a claim that substrates, conditions or yields are interchangeable across the list. The authors specifically point to reactions sensitive to strong base, including palladium-catalysed coupling, as settings where control over base generation may matter. Compound characterization and experimental procedures are provided in the paper’s supplementary information. Garrett A. Hoteling, a PhD student on the team, said: “We tested the superbase precatalysts in Michael-type addition reactions, amidation reactions and polymerisation reactions.”

The paper establishes a research strategy and reaction demonstrations, not universal scope or broad manufacturing readiness. For a prospective application, the practical questions are whether the salt and epoxide work with the target substrate and solvent, whether the activation rate fits the reaction, and whether reagent costs are acceptable at the intended scale. Chemistry World reported University of Michigan medicinal chemist Tim Cernak’s concern that the cost of superbase carboxylate salts could constrain production at very large scale; this is an expert concern, not a published economic analysis.
The primary report is Sujansky, Hoteling and Bandar, “A strategy for the controllable generation of organic superbases from benchtop-stable salts,” Chemical Science 15 (2024), 10018–10026, first published 29 May 2024. Its article page links to the supplementary information for experimental detail. Chemistry World’s 11 June 2024 report provides the quoted expert comments and scale-up qualification.
