In a 2018 laboratory study, the approach produced up to 8.49 ± 0.31 g/L isobutanol from glucose under micro-aerobic conditions. It shows how light can control microbial metabolism; it does not establish a commercially viable biofuel process.
How can light control biosynthesis?
Optogenetics uses light-responsive proteins to control gene expression. Evan M. Zhao and colleagues engineered Saccharomyces cerevisiae with two circuits: OptoEXP, which induces gene expression in response to light, and OptoINVRT, which represses it. The circuits let the researchers adjust genes involved in ethanol fermentation and the isobutanol pathway rather than leaving those pathways at a fixed level throughout fermentation. The authors’ paper appeared in Nature in 2018.

Why alternate growth and production?
Growing yeast and making a target chemical place different demands on the cell. The team used light-supported gene expression to help the yeast grow, then darkness to shift the engineered cells toward product formation. They also applied periodic light pulses during production to tune enzyme expression rather than relying only on a simple light-on/light-off schedule.

Ilv2 supports the isobutanol pathway but uses pyruvate; Pdc1 directs pyruvate toward ethanol, while also helping regenerate NAD+, which yeast needs to use glucose and grow. Controlling expression over time gave the researchers a way to balance these competing needs as the fermentation moved from growth toward production. Chemistry World’s 2018 account explains this metabolic tension.

What did the yeast produce?
For micro-aerobic production from glucose, Zhao and colleagues reported these study maxima:
| Product | Maximum reported concentration |
|---|---|
| Isobutanol | 8.49 ± 0.31 g/L |
| 2-methyl-1-butanol | 2.38 ± 0.06 g/L |
These are results from the researchers’ 2018 study, not current industry benchmarks or specifications for a commercial process. The contemporary Chemistry World report described the isobutanol concentration as five times the previous academic yeast concentration at the time and reported a yield of 54 mg per gram of glucose fed. Those comparisons belong to that 2018 report, not to a present-day ranking.

How did the control modes differ?
| Control mode | When light was applied | Purpose |
|---|---|---|
| Growth followed by production | Light supported the growth phase; darkness followed for production. | Shift gene expression from supporting cell growth toward making product. |
| Periodic pulses | Light was pulsed during the production phase. | Tune expression of pathway enzymes during production. |
The point is not that light itself supplies the chemical product. Light acts as a control signal that changes which genes the yeast expresses, and therefore which enzymes it makes. In the 2018 Chemistry World report, Avalos described the switching as “immediate and reversible.” Raman Vaidyanathan of the University of Sheffield said the system could provide different levels of expression, rather than only an on-off state.

The study included light-stimulated fermenter experiments and a 5-litre setup used to test light-induced gene expression at high cell density. Its extended data describes blue LED panels for that experimental setup. This is evidence of laboratory-scale control, not proof that ordinary consumer LED panels would reproduce the illumination conditions, or that the process is ready for industrial use.

The paper does not establish commercial-scale economics. The 2018 Chemistry World report noted that a yield of 54 mg per gram of glucose was not economically viable at the time; Jörg Pietruszka of the Jülich Research Centre said economic feasibility still needed testing. The cited sources do not establish whether this specific process has since become commercially viable.
Independent reader supportYour contribution helps us test, update, and keep practical guides available for everyone. Why the result matters—and what it does not show
The study demonstrates a useful engineering idea: a fermentation can be programmed to change its gene-expression regime as the cell’s needs change. Light offered researchers a reversible and adjustable way to coordinate growth and production in engineered yeast. The reported concentrations establish a research result under specified laboratory conditions, not a commercial production rate, cost, or scale-up outcome.
