The result points to a possible mechanism of vulnerability; it is not a clinical test or a measure of a person’s Parkinson’s risk.
What the study compared
Hare and colleagues compared the SNc with the nearby ventral tegmental area (VTA) and other midbrain regions. They also examined a mouse model in which researchers created a one-sided lesion with 6-hydroxydopamine (6-OHDA), allowing comparison between the lesioned and contralateral sides.

The work was published in Chemical Science in 2014 as “An iron–dopamine index predicts risk of parkinsonian neurodegeneration in the substantia nigra pars compacta”. The word “predicts” in the paper title refers to the study’s experimental index, not a validated prediction for patients.
How co-localisation was measured
The team combined metal labelling with laser-ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) imaging to map iron in mouse midbrain sections. They also used immunohistochemical labelling for tyrosine hydroxylase (TH), an enzyme involved in dopamine biosynthesis, to identify dopamine-related regions.

TH labelling is a marker of dopamine-producing-cell regions; it does not directly image or quantify dopamine molecules. The researchers used the spatial relationship between the iron map and TH signal to calculate an iron–dopamine product, an experimental index of their local combination.
Why the combination mattered
The SNc did not have the midbrain’s highest iron concentration. Instead, the proposed distinction was that substantial iron coincided with substantial dopamine-related signal, giving the SNc a higher iron–dopamine product than the adjacent VTA. In the authors’ interpretation, looking at iron or the TH-related signal alone did not capture the same regional pattern.

The idea is that iron and dopamine-related chemistry together may promote oxidative stress: iron can participate in reactions that generate reactive species, while dopamine chemistry can also contribute to oxidative processes. The study’s abstract describes the concern this way: “The co-localization of iron and dopamine raises the risk of a potentially toxic reaction.” That is the authors’ mechanistic hypothesis, not proof that this pathway causes Parkinson’s disease in people.
What changed in the mouse lesion model
In mice given a unilateral 6-OHDA lesion, the iron–dopamine product was reported as 42.5 ± 18.0% greater in the lesioned hemisphere than in the contralateral midbrain. This is a result from an experimental lesion comparison, not a 42.5% increase in human disease risk, disease severity, or likelihood of developing Parkinson’s.

What the finding does—and does not—show
- It supports a regional hypothesis: the combination of iron and a dopamine-related marker may help explain why the SNc is vulnerable compared with nearby regions.
- It does not establish a human biomarker: the index was measured in mouse tissue and has not been shown here to diagnose Parkinson’s or predict an individual’s prognosis.
- It does not establish treatment: the study tested neither a therapy nor a supplement, and its findings do not justify changing medical care.
- It is one preclinical study: published in 2014, it cannot by itself establish the current consensus across later animal and human research.
