A proposed mechanism for DNA photostability starts with what happens just after ultraviolet (UV) light excites a base pair: an electron moves from the purine base to its paired pyrimidine, then a proton follows. A 2020 study argues that this electron-driven proton transfer (EDPT) relieves antiaromaticity in the excited purine and helps the pair return quickly to its ground state. The proposal adds an excited-state explanation to established redox-based accounts; it does not show that antiaromaticity relief is the only cause of DNA’s photostability.
What the proposed mechanism says
The proposal concerns canonical Watson–Crick adenine–thymine (A·T) and guanine–cytosine (G·C) base pairs after they absorb UV light. In the authors’ account, excitation places the purine component—adenine or guanine—in a highly antiaromatic, reactive first excited ππ* state. The pair can then deactivate through a sequence of coupled charge and proton movement.

- UV excitation: The base pair absorbs light and reaches a locally excited state.
- Electron transfer: An electron moves from the excited purine to its paired pyrimidine, thymine or cytosine.
- Proton transfer: A proton follows, giving the process its name: electron-driven proton transfer, or EDPT. It is also described as proton-coupled electron transfer (PCET).
- Return to the ground state: The resulting charge-transfer structure reaches a conical intersection, a pathway between electronic states, and the pair returns to its ground state. The paper describes this deactivation as occurring within picoseconds.
The proposed benefit is relief of excited-state antiaromaticity in the purine’s π-system. Although the proton movement follows a coordinate involving an N–H bond, the authors’ explanation centers on how electron transfer changes the electronic character of that π-system.
How this explanation relates to DNA photostability
DNA bases absorb UV energy, but excited molecules do not necessarily remain excited long enough to undergo damaging chemistry. Rapid non-radiative deactivation—returning to the ground state without emitting light—is one way a base pair can dispose of that energy. The 2020 study connects the proposed EDPT route to such rapid deactivation in the canonical pairs it considers.

This account complements explanations based on ground-state oxidation and reduction potentials, which address the energetics of electron transfer. The two perspectives focus on different aspects of the process: redox energetics can help explain whether transfer is favorable, while the antiaromaticity proposal describes a possible excited-state benefit of the transfer. The study does not establish that the latter replaces the former or uniquely explains photostability.
What evidence supports the proposal—and what it does not establish
Karas, Wu, Ottosson, and Wu’s 2020 open-access study links the proposed EDPT pathway to a change in aromaticity character on photoexcitation and discusses structural, magnetic, and electronic indicators. These indicators support the authors’ interpretation, but they are not definitive direct measurements of excited-state antiaromaticity: the paper notes that the MCI and HOMED indices it discusses were not designed to capture excited-state (anti)aromaticity.

The scope also matters. The account addresses canonical A·T and G·C base-pair structures; it does not by itself demonstrate the same pathway across every DNA sequence, in intact DNA, or under biological conditions. A 2023 review places excited-state aromaticity gain and antiaromaticity relief in a broader photochemical context, describing their relevance to photoreactivity and photostability in multiple molecular systems. That broader context is not proof of a universal DNA mechanism.

Why the proposal matters for future research
If excited-state antiaromaticity relief helps steer deactivation, it offers a design idea for studying photostable unnatural base pairs. The primary study presents this as a prospective implication, not as a demonstrated consumer application or a ready-made biological intervention. Its contribution is a mechanistic hypothesis that can guide further investigation of how base-pair structure and excited-state electronic character influence the fate of absorbed UV energy.
