How Chemists Build a 5–8–5 Carbotricycle in Four Steps

A 2018 study reports a four-step, photoinduced route to gram quantities of a fused 5–8–5 carbocyclic framework found in more than 30 natural products.

A 2018 Chemical Science paper reports a four-step route to gram quantities of a fused 5–8–5 carbocyclic scaffold. Its key move is a photoinduced cycloisomerization: light helps turn a readily accessible starting isomer into a strained cyclobutane that can rearrange to form the scaffold’s central eight-membered ring. This is a method for making a molecular framework, not a four-step synthesis of a finished drug.

What “carbotricycle” means here

In this context, the term refers to a fused three-ring carbon framework known as a 5–8–5 system: its three fused rings contain five, eight and five carbon atoms, respectively. The route was reported by Anna E. Salvati, James A. Law, Josue Liriano and James H. Frederich in “Modular access to functionalized 5–8–5 fused ring systems via a photoinduced cycloisomerization reaction,” published in Chemical Science in 2018.

The authors describe the framework as a useful target because the dicyclopenta[a,d]cyclooctene core occurs in more than 30 di- and sesterterpene natural products. The shared core is not, by itself, a guarantee of any particular biological effect: the natural products differ in their peripheral substituents, which help determine their properties.

How the four-step strategy builds the framework

The route’s central idea is to convert a more accessible isomer into a strained intermediate, then use that strain to reorganize the molecule into the desired fused rings. The authors describe the sequence as a proposed mechanistic pathway; it should not be read as proof that every mechanistic detail was directly established.

  1. Prepare the cycloisomerization substrate. The modular route begins with a precursor designed to place the reactive groups in the right relationship for an intramolecular reaction. The paper reports access to the target scaffold in four steps, but the abstracted source details do not specify reagent-by-reagent conditions or the identity of every intermediate.
  2. Use light to form a cyclobutane. The authors propose an intramolecular [2+2] photocycloaddition, in which two alkene units within the molecule join to make a cyclobutane. The photochemical step establishes a stereodefined, highly strained intermediate.
  3. Rearrange the strained ring system. The accumulated strain is proposed to drive a Cope rearrangement, reorganizing bonds to generate the eight-membered ring that is central to the 5–8–5 framework. The strategy also installs a C11 quaternary stereocenter as part of scaffold construction.
  4. Isomerize to the conjugated enone. A final isomerization gives a conjugated enone in the fused-ring product. The paper’s abstract characterizes the overall transformation as a cycloisomerization that forms the 5–8–5 framework directly.

Why the scaffold is useful to natural-product chemistry

A concise route to this shared core can give chemists a platform for studying or preparing related molecules whose differences lie in substituents around the rings. The paper points to natural products with distinct reported biological activities:

  • Fusicoccin A is described as an orthosteric stabilizer of 14–3–3 protein–protein interactions.
  • Ophiobolin A is described as a cytotoxin that modulates calmodulin activity.
  • Cyclooctatin is described as an inhibitor of lysophospholipases.

These examples explain the scaffold’s interest in natural-product synthesis. They do not show that the synthetic core itself is a medicine, nor do they establish clinical efficacy for the named natural products.

What the four-step claim does—and does not—establish

The paper reports that the scaffold can be accessed in gram quantities through four steps. That is a statement about a synthetic route to a molecular framework, not a claim that the authors produced a drug candidate, demonstrated a treatment, or established a universal improvement over every earlier synthesis.

The authors position the approach as a concise, modular alternative within the specific context of earlier methods. Its use of photochemistry and the ability to vary substituents are relevant when considering possible applications, but whether it is preferable depends on the target molecule and the detailed conditions. The available summary does not provide a numerical yield, a full reagent list, or a complete comparison of step counts and conditions against other routes.
