In a 2014 proposal, researchers adapted term frequency–inverse document frequency (TF-IDF), a technique for identifying informative words in documents, to score chemical bonds as candidate disconnections in retrosynthetic analysis.
How does a molecule become a “sentence”?
The approach treats a molecular structure like a sentence and recurring molecular fragments like words. The team assembled a dictionary of fragments found across molecules and compared how often those fragments occurred. The report says the frequency distribution across all structural fragments resembled the distribution of text fragments in English more closely than a dictionary limited to functional groups did. Bartosz Grzybowski of Northwestern University described the connection this way: “Modern computational linguistics is pattern recognition,” Chemistry World reported on 17 July 2014.

What did TF-IDF do with chemical bonds?
In information retrieval, TF-IDF helps distinguish terms that are informative in a document from those that are common across many documents. The proposal applied that statistical idea to molecular bonds: bonds with higher information content could be prioritized as promising places to disconnect a molecule when reasoning backward from a target structure.
This is a ranking signal, not a complete synthesis planner. The report describes it as something that could complement chemical heuristics, helping suggest which bond breaks deserve attention rather than determining a full route or establishing that a route is practical.

What does the reported 97% result mean?
Chemistry World’s 2014 account of the Cadeddu et al. study reported that in around 97% of manual assessments, at least one chemist selected one of the computer’s top three bond choices. This measures agreement with a computer-selected option among its top three; it is not a 97% synthesis success rate, evidence that every suggestion was experimentally tested, or proof that the method outperformed expert planning.
What questions did chemists raise?
The report presented the method as an early step and highlighted two reservations: whether it would be useful to skilled practitioners, and whether it would stand up to the challenges of complex molecules. University of Bristol chemist Varinder Aggarwal said, “The proof will be provided when it’s tested against complex molecules.” Phil Baran of the Scripps Research Institute questioned whether a method based on cataloguing past patterns could support creative planning, warning that it might remain “inside the box.” These were expert reactions in the 2014 coverage, not assessments of current retrosynthesis software.

How does the proposal relate to SYNTHIA today?
The 2014 report said Grzybowski’s team integrated the approach into Chematica. Merck now markets SYNTHIA as retrosynthesis software and offers SYNTHIA Lab, a SaaS subscription for academic and nonprofit teams; its product page lists a six-month subscription. Merck’s SYNTHIA product page and SYNTHIA Lab page describe the current offering, but do not establish that it still uses the 2014 TF-IDF method. The available information therefore does not support a direct performance comparison between that proposal and the current product.
