The result is an early research proof of concept—not a treatment shown to work in people.
What STP3 is and how it works
STP3 is a sulphonatopropoxy terphen[3]arene: a ring-shaped host molecule designed to hold smaller “guest” molecules. Junyi Chen and colleagues reported its synthesis as the first water-soluble terphen[3]arene of this kind. Sulfonated side chains make the macrocycle soluble in water, a useful property for a proposed oral formulation. The authors describe the work as direct host–guest complexation and identify pharmaceutical research as a potential application (Chen et al., Chemical Communications, first published 11 February 2022).

The idea is to temporarily associate STP3 with a bitter alkaloid. If less of the drug is available to interact with taste receptors in the mouth, bitterness may be reduced. This is not a coating or flavoring; it is a molecular binding strategy.

Why the pH response matters
For an oral taste-masking approach, binding needs to be strong enough to limit bitterness in the mouth but not so strong that the medicine remains trapped. Chemistry World reports that the researchers found different binding strength at pH 6.8, used to represent the mouth, and pH 1.2, used to represent gastric acid. The complex became unstable in acidic conditions and released the alkaloid (Blake Baker, Chemistry World, 23 February 2022).

That pH-dependent release is the proposed handoff: complexation to help mask taste, followed by release in acid. The measurements do not show how STP3 behaves during digestion in people, how much medicine reaches the bloodstream, or whether the timing and amount of release would be suitable in a human dose.
What the experiments found
The team reported complexes with six alkaloid medicines; the Chemistry World account highlights an atropine-related medicine and colchicine as examples. An electronic-tongue taste-sensing system detected a substantial reduction in bitterness when STP3 was co-administered. The paper’s abstract also reports alleviation of aversive alkaloid responses in vitro and in vivo.

Animal studies described in the report found no toxicity from the host in those experiments and no reduction in the activity of the carried drugs. These findings are encouraging for further investigation, but they are not evidence of human safety, effectiveness, or preserved therapeutic action in patients.

What remains unproven
The study does not establish clinical efficacy, human safety, regulatory approval, commercial availability, or use in patients. It supports a taste-masking strategy for a subset of bitter alkaloids under experimental conditions, not a general solution for bitter medicines.

Translation also requires balancing drug binding against the need to retain the medicine’s intended activity, as well as solving practical formulation and scale-up questions. In the 2022 Chemistry World report, supramolecular chemist Ana Castilla of the University of Greenwich said, “To the best of my knowledge, there are no applications yet of synthetic containers such as the one reported in this work [for use] in the pharmaceutical industry.” That was her assessment at the time, not a current survey of the market.
The authors themselves framed the application cautiously, writing that “STP3 has potential applications as an efficient taste-masking macrocyclic container for use in pharmaceutical research.” STP3 is therefore best understood as a promising research concept, not a medicine or a product currently established for consumers.
