In a 2016 proof of concept, researchers screened 22,293 natural products and derivatives against synthetic mirror-image MDM2, then synthesized a mirror-image version of a hit and found that it inhibited the natural MDM2–p53 interaction in a biochemical assay. The result was a drug-discovery lead—not a cancer treatment.
How does a mirror-image screen stand in for a virtual library?
Many molecules, including proteins, are chiral: their mirror-image forms have the same atoms and connections but a different three-dimensional handedness. That difference can determine whether two molecules fit together. A natural protein may recognize one handedness of a compound but interact weakly or not at all with its mirror image.

The researchers used that symmetry to search chemical space without first making a large collection of mirror-image compounds. They chemically synthesized a mirror-image version of the target protein, screened available compounds against it, and then synthesized mirror-image versions of selected hits for testing against the natural target. The “virtual” library was therefore a proxy inferred from the screen, not a physical collection of compounds that the researchers had already made.

As the study authors put it, “We established a facile access to an unexplored mirror-image library of chiral natural product derivatives using D-protein technology.” The strategy still requires chemical synthesis: first for the mirror-image target, and later for mirror-image hit compounds worth validating.

What did the MDM2–p53 experiment test?
MDM2 negatively regulates p53, a tumor-suppressor protein. The team focused on the p53-binding domain of MDM2, residues 25–109, and chemically produced its mirror-image form, D-MDM2. In surface plasmon resonance experiments, the synthetic proteins bound the corresponding mirror-image p53 peptides with high affinity, while binding between mismatched handedness was practically nil. This provided a basis for using D-MDM2 to screen compounds that might have mirror-image counterparts capable of engaging natural MDM2.
What did the 22,293-compound screen find?
The team used a chemical array containing 22,293 natural products and derivatives from RIKEN’s NPDepo collection. The 2016 study reported 43 initial selective binding hits. In follow-up competitive binding assays, four compounds showed inhibitory activity against the natural L-MDM2–L-p53 interaction, its mirror-image D-MDM2–D-p53 counterpart, or both.

NP843 and ent-NP843
One hit, NP843, is a chiral α-tocopherol derivative. It selectively inhibited the D-MDM2–D-p53 interaction, with a reported IC50 of 6.5 ± 0.5 μM. The researchers then synthesized its mirror image, ent-NP843. In a biochemical assay, ent-NP843 inhibited the natural L-MDM2–L-p53 interaction with a reported IC50 of 7.6 ± 1.9 μM. These values describe the compounds’ activity in the reported assays; they are not measurements of effects in animals or people.

Follow-up analogue tests showed that activity depended on precise structure, not just on a molecule being a mirror image. The stereochemistry at a tetrasubstituted carbon in the chromane scaffold mattered, and shortening the three-isoprene-unit side chain eliminated inhibitory activity in the derivatives tested. This helps explain why a screening hit is a starting point for further work rather than proof that any related compound will have the same effect.

How does the approach differ from direct screening?
| Question | Direct screening of available compounds | Mirror-image strategy in the study |
|---|---|---|
| What is screened? | Available compounds against the natural target. | Available compounds against a synthesized mirror-image target, as a proxy for mirror-image compounds against the natural target. |
| What must be synthesized? | No mirror-image target is needed for the direct screen; the study does not establish other synthesis requirements for a general direct-screening workflow. | The mirror-image target protein is synthesized before screening, and selected mirror-image hit compounds are synthesized for follow-up against the natural target. |
| What chemical space can it probe? | The compounds actually present in the screened collection. | Potentially, otherwise unavailable enantiomeric chemical space inferred through stereochemical symmetry. |
| What validation did this example reach? | The study did not report a head-to-head comparison with direct screening. | Binding and competitive inhibition in biochemical assays; no animal or human treatment outcome was reported. |
| What limits extension to another target? | No special mirror-image biomolecule is required for ordinary screening. | A corresponding mirror-image biomolecule must be accessible, and the mirror-image target and selected hits require chemical synthesis. |
The authors presented the method as a way to avoid synthesizing numerous mirror-image compounds before knowing which are worth pursuing. The study did not quantify time or cost savings against conventional screening, so it does not establish that the approach is faster or cheaper in practice.

The 2016 paper demonstrates a screening concept and reports biochemical inhibition of the MDM2–p53 interaction by NP843 and ent-NP843 in assays. It does not show that either compound treats cancer, is a validated medicine, or benefits patients. Moving from an assay hit to a treatment would require substantial additional evaluation, including whether enough mirror-image compound can be made and whether it is effective and safe.

At the time, Brian Cox, a chemical-synthesis and drug-discovery researcher at the University of Sussex, described the approach to Chemistry World as “a very exciting approach, assuming technologies can provide the proteins for screening”. That comment reflects the promise and practical dependency identified in 2016, not a later clinical assessment.
