How a Brainless Sea Sponge Uses Sunset to Prepare for Metamorphosis

In one sponge species, fading light is associated with DNA-accessibility changes that prepare swimming larvae for a later settlement cue—not conscious anticipation.

A sea sponge larva does not need a brain to respond to sunset. In the sponge Amphimedon queenslandica, researchers found that the changing light before night is associated with changes in how accessible parts of the larva’s DNA are. This molecular preparation happens while the larva is still swimming; a separate cue from coralline algae can then trigger settlement and the rapid changes that begin metamorphosis.

What sunset changes before the larva settles

The study followed larvae of Amphimedon queenslandica, a demosponge, through competence, settlement and early metamorphosis. It measured both RNA, which reflects gene expression, and chromatin accessibility, which indicates how open regions of DNA are to regulatory proteins.

As light diminished at sunset, chromatin accessibility changed extensively in swimming larvae. Regions near genes involved in settlement and metamorphosis became more accessible before those genes were broadly switched on. The accompanying changes in gene expression at this stage were comparatively modest. In other words, the larva enters a state in which some relevant genes are easier to activate later; sunset does not directly turn on every metamorphosis gene.

Olivia Hewitt, a postdoctoral researcher at the University of Queensland and an author of the study, describes the regions as being “primed (made ‘accessible’) long before the genes themselves get switched on.” Read the University of Queensland explainer.

Sunset and algae play different roles

The light change is a timing cue associated with the larva’s preparatory state. It is not the same as the local signal that prompts a competent larva to settle. In the experiments, the researchers used the coralline alga Amphiroa fragilissima as an environmental inducer. After this cue, many transcription factors—including members of the AP-1/bZIP family—were rapidly and transiently activated near genes whose expression changed. The authors propose that earlier chromatin priming helps make this coordinated response possible.

The distinction matters: light progression is associated with becoming ready, while the algal cue induces settlement. The study reports that larvae kept in constant light did not settle even when exposed to the inductive algae. They instead entered a different chromatin and transcriptional state, including widespread loss of accessibility and repression of competence-associated transcription factors such as CLOCK. This experimental contrast links normal light-dark progression to competence and settlement in this species; it does not show that sunset alone triggers settlement.

How preparation leads to metamorphosis

  1. Light fades. While larvae are still swimming, sunset-associated chromatin remodelling makes some regions near settlement- and metamorphosis-related genes more accessible.
  2. A local inducer arrives. In the study, coralline algae provided the environmental cue associated with settlement.
  3. Gene activity changes rapidly. Following induction, transcription factors and other genes are activated as the larva settles and early metamorphosis begins.
  4. The larval body is remodelled. The University of Queensland explainer says attachment and the start of larval-body disassembly occur in around 30 minutes. That is an approximate description of the studied transformation, not a universal timing rule for all sponges.

The researchers interpret the sequence as anticipatory in a biological sense: the larva’s molecular state changes before the local inducer arrives, making a later response possible. It is not evidence of conscious planning. Sponges have no brain, nerves or neurons; as the University of Queensland explainer notes, they are made up of specialised cells.

What the finding does—and does not—show

The primary evidence concerns one species, A. queenslandica, and the experiments reported in the paper. The authors suggest that chromatin priming could be relevant to other animals with distinct larval and adult life stages, but that broader idea is a proposed model, not a result established across marine invertebrates.

The primary paper, “Light-entrained chromatin priming poises rapid metamorphosis in a marine sponge,” by Huifang Yuan, Oceane Blard, Zac Pujic, Bernard M. Degnan and Sandie M. Degnan, is an eLife revised reviewed preprint, version 2, dated 2026-10-02. Its experiments and molecular measurements are described in the eLife paper. Broader work has also examined light and sponge larval behaviour, but does not establish this chromatin mechanism: see the 2023 PLOS ONE study on photobehaviour, dispersal and settlement.
