Horseshoe crabs remain important to pharmaceutical manufacturing because their blood cells supplied the biological basis for a widely used test for bacterial endotoxins. Those toxins can cause serious reactions in patients, so manufacturers must check for them. A recombinant alternative can avoid crab-derived material, but each testing method still has to be shown suitable for the product and purpose.
What makes horseshoe-crab blood useful?
The value lies in an immune response, not in using the animal as a drug ingredient. Horseshoe-crab blood contains amebocytes, cells that rapidly clot when they encounter certain gram-negative bacteria. Researchers adapted that response into Limulus amebocyte lysate (LAL), a laboratory assay for bacterial endotoxins. The U.S. Food and Drug Administration (FDA) describes the biological mechanism and assay formats in its account of bacteria and endotoxin testing.

Endotoxins are substances released by certain bacteria. If they contaminate a medicine, they can trigger serious adverse reactions. FDA describes testing for them as a critical safety and quality step in manufacturing biological products, making endotoxin detection a routine part of quality control rather than an optional research tool (FDA guidance on pyrogen and endotoxin testing).
Two ways to read an LAL test
FDA describes gel-clot and chromogenic formats. In a gel-clot assay, the result is based on whether the reaction forms a clot. In a chromogenic assay, the reaction produces a color change that can be measured. Both use the general biological detection system behind LAL, but the readout differs; assay performance depends on the specific method and sample.

How recombinant testing changes the equation
Recombinant Factor C (rFC) is made using recombinant production rather than horseshoe-crab blood or lysate. It reproduces Factor C, the endotoxin-sensitive component of the crab’s clotting response. A 2020 review in the PDA Journal of Pharmaceutical Science and Technology reported high correlation between rFC and traditional lysate testing in comparative studies. It also noted a difference in detection: lysate reagents can respond through Factor C and a separate Factor G pathway, while rFC does not detect Factor G (Bolden et al., 2020).
| Comparison | LAL | rFC |
|---|---|---|
| Reagent origin | Horseshoe-crab-derived lysate | Recombinantly produced Factor C; no crab-derived material required |
| Detection pathways | Can detect Factor C and Factor G pathways | Detects the Factor C pathway; does not detect Factor G |
| Suitability for a product | Must be verified for the material and intended use | Must be verified for the material and intended use |
Neither method is established as universally better for every product. The appropriate choice depends on the material being tested, the method’s performance under the intended conditions, and the manufacturer’s validation work.

What the 90% figure does—and does not—mean
A 2018 study reported that using rFC to test water and other common manufacturing materials could reduce the use of crab-derived reagents by 90% for that application. The finding is specific to those materials and the study’s testing scope; it does not show that the pharmaceutical industry can eliminate 90% of horseshoe-crab use across all products or tests (Maloney et al., PLOS Biology, 2018).
What FDA’s current position means for manufacturers
FDA’s revised Edition 2 guidance accommodates recombinant reagents for bacterial endotoxin testing. It directs sponsors to verify that a method is suitable for its intended purpose, review the supplier’s validation package, and conduct experiments appropriate to the complexity of the material. FDA also points to USP Chapter <86>, “Bacterial Endotoxins Test Using Recombinant Reagents” (FDA guidance).

That is not a blanket approval for every recombinant reagent in every test. A manufacturer must establish that the chosen assay works for its particular product and intended use.
A documented transition route
On August 31, 2026, FDA reported that an international collaborative assessment had concluded with approval in June 2026 of a post-approval change protocol. The protocol let a manufacturer transition several biological products—from cancer, cardiovascular, respiratory, and rare-disease therapies—from traditional lysate-based testing to recombinant endotoxin testing. It demonstrates that a transition can be approved for specified products, not that every product can switch without its own suitability work (FDA, August 31, 2026).

Why the animal remains part of the story
The crab’s contribution is foundational: its clotting defense gave laboratories a sensitive way to detect a serious contamination risk. Recombinant testing creates a route to perform that safety check without crab-derived reagents where the method is suitable and accepted for the product. That makes horseshoe crabs still relevant to pharmaceutical safety, even as testing options evolve.
