DP5 is an open-source computational method that estimates how consistent one proposed molecular structure is with carbon-13 NMR chemical-shift evidence. It can also flag the atoms that contribute to concern, giving chemists a diagnostic lead—not proof that the structure is right or an answer that identifies the correct alternative.
What DP5 checks
When a chemist has one proposed structure to assess, DP5 addresses a different question from a method that ranks a supplied set of alternatives: how probable is it, under the method’s assumptions, that this single candidate is consistent with the carbon-13 NMR evidence?

In the summarized workflow, calculated chemical shifts for the candidate are compared with experimental shifts. Prediction-error distributions for individual atoms contribute to a probability for the molecule. The method is designed to calibrate that probability for a single candidate, rather than simply declare it the best option among several.

This makes DP5 a probabilistic validation check. It does not draw a structure, determine a molecule from a spectrum on its own, or establish which alternative structure is correct if the candidate appears inconsistent.

What the atom-by-atom output can tell you
An atom-level view can help localize likely trouble spots: it points to sites whose predicted-versus-experimental shifts contribute to concern about the proposed structure. A chemist can use that cue to re-examine the assigned signals and the local structural environment. The cue is not a diagnosis of what is wrong, and it does not independently prove that a particular atom assignment or bond should be changed.
Interpreting the result therefore still requires chemical judgment and the relevant experimental evidence. DP5’s stated scope is carbon-13 chemical shifts; the available sources do not establish that its result incorporates other nuclei, multidimensional NMR data, or fully automated spectrum processing.
How to interpret the probability
A DP5 probability is conditional on the method’s chemical-shift predictions, error model, and calibration. It should be read as an estimate under those assumptions, not as a guarantee that a proposed structure is correct.

The review attributes this limit to uncertainty in atomic environments and errors in computational chemical-shift prediction. That is the review’s description, not an established universal ceiling for every implementation or later revision.
Why automation is relevant—and what is not established
Chemistry World reported in April 2022 that DP5 could be automated for high-throughput robotic synthesis workflows. That describes a potential application, not a measured throughput or evidence of broad deployment. The available sources do not provide a quantitative throughput result or a broad prospective performance benchmark.
Automation could make a single-candidate check useful in workflows that generate many compounds, but an automated probability would still need to be interpreted within the method’s prediction and calibration limits. The report of a possible workflow does not establish how widely it is used or how well it performs across settings.
