Does the Keto Diet Increase Cancer Risk? What MIT’s Mouse Study Found

A 2026 MIT study found different effects of a ketogenic diet on small-intestinal and colon tumors in genetically predisposed mice. It does not show that keto causes cancer in people.

A 2026 MIT study found that a ketogenic diet increased small-intestinal tumors in genetically predisposed mice, while suppressing colon tumors in the same research. The result points to a possible tissue-specific effect of dietary fat metabolism—not proof that keto causes cancer in people.

What did the MIT study find?

The study, “Ketogenic diet mediates intestinal tumorigenesis through lipids not ketones,” was published in Nature on July 15, 2026. Researchers tested dietary, genetic, and metabolic changes in mouse models prone to developing intestinal adenomas. The Nature abstract reports that a ketogenic diet accelerated tumor burden and shortened survival in these models, independently of ketone metabolites. Read the Nature paper.

MIT’s summary says the mice were genetically predisposed to intestinal cancer and were assigned ketogenic, control, or high-fat, high-calorie diets. Mice on the ketogenic diet developed more small-intestinal tumors than controls, at rates similar to or higher than those on the obesogenic diet, despite not becoming obese. Does the keto diet increase cancer risk in people?

This study does not establish that keto increases cancer risk in humans. The reported tumors arose in genetically predisposed mice; the sources provide no human cancer-risk estimate or quantified effect size. The findings therefore cannot tell an individual whether their diet changes their cancer risk.

MIT’s senior author, Ömer Yilmaz, cautioned against generalizing across tissues: “Ketogenic diets have distinct effects on different tissues even within the gastrointestinal tract. I think the message here is that we need to be very careful in generalizing the effects that these diets can have, because what might be beneficial for one tissue may be detrimental for another tissue.”

Why might the effects differ between the small intestine and colon?

Tissue in the mouse models Reported result on the ketogenic diet
Small intestine More tumors than in controls; tumor burden accelerated.
Colon Colon tumors were suppressed.

The contrast matters: the study does not support a blanket claim that keto promotes tumors throughout the digestive tract. Yilmaz told MIT that more active intestinal stem cells may help the small intestine repair after injury, while also creating more opportunity for tumor formation. These are results and explanations from mouse models, not a demonstrated human mechanism.

Was the effect caused by ketones or by dietary fat?

The paper attributes the effect to metabolism of dietary lipids rather than ketone bodies themselves. It implicates fatty-acid oxidation, a process by which cells use fatty acids for energy. In the mouse models, this pathway was linked to intestinal stem-cell expansion and tumorigenesis; genetic interference with PPAR signaling or CPT1A-dependent fatty-acid oxidation limited parts of the effect.

Because the proposed driver was fat metabolism rather than ketones, MIT says commercial ketone supplements or drinks would not be expected to reproduce the effects observed in the study.

Should you change your diet or cancer treatment because of this study?

The mouse findings do not provide a basis for changing cancer treatment or an individual’s diet. The study did not establish clinical guidance for people. If you have cancer, a hereditary cancer risk, or questions about a ketogenic diet, discuss your situation with your clinician rather than using this animal study to make treatment decisions.
