No. Sony says the games in Uncharted: Legacy of Thieves Collection are designed so each can be enjoyed on its own. Playing earlier entries adds character and relationship context, but you do not have to finish them first.
Can you start with an Uncharted game without playing the earlier ones?
Yes. Sony Interactive Entertainment’s Legacy of Thieves Collection FAQ answers “Which UNCHARTED game should I play first?” with: “Whichever one you’d like. While there is a general evolving storyline through the series, each game is meant to stand on its own and has a narrative that’s unique. Any way you choose to play through the series, you’ll enjoy!”

What order should you play the games in?
If you want the most context for Nathan Drake’s story, use release order: play the first three games, then Uncharted 4. If you mainly want to try an adventure without committing to the earlier trilogy, Sony’s guidance supports starting with any game.

Sony identifies the first three single-player campaigns as the contents of The Nathan Drake Collection. The collection is a convenient way to access that trilogy, not a prerequisite for playing Uncharted 4.
Does The Lost Legacy require playing Uncharted 2 or 3?
No. The Lost Legacy follows Chloe Frazer, with Nadine Ross. Sony says Chloe first appeared in Uncharted 2: Among Thieves and last appeared before The Lost Legacy in Uncharted 3: Drake’s Deception. Those earlier appearances provide background, but Sony does not describe either game as a requirement.
