“Making allowances for anthrax” was a 2004 Chemistry World news item about a possible cancer-research application of anthrax toxin. The key finding was a crystal structure showing how the toxin’s protective antigen binds a human cell receptor. That structural insight suggested ways to engineer toxin components for research into tumor-targeted therapies; it did not show that anthrax treats cancer or establish a treatment for patients.
What did “making allowances for anthrax” mean?
The phrase was wordplay about the potential to adapt components of anthrax toxin for cancer research—not a suggestion that anthrax infection should be used as a therapy. The indexed standfirst for Emma Davies’s 1 August 2004 Chemistry World item says, “Crystal structure brings researchers closer to using anthrax to fight cancer.” The full article is not available to verify its detailed reporting, so the standfirst is the limit of what can be attributed to that news item. The underlying scientific result was more specific: researchers determined the structure of anthrax protective antigen bound to the human-cell receptor CMG2. Their paper described this as a potential source of insight for both antitoxins and cancer-therapeutic design, not evidence of a tested cancer medicine.
What did the protective-antigen structure show?
Anthrax toxin is composed of protective antigen (PA), lethal factor (LF), and edema factor (EF). PA binds a host-cell receptor, is processed, and assembles into a prepore that can form a membrane pore. LF and EF can then enter the cell. The 2004 study examined PA’s interaction with CMG2 and discussed the closely related receptor TEM8 as well.

Santelli, Bankston, Leppla, and colleagues reported the PA–CMG2 crystal structure at 2.5 Å resolution. Their structural analysis and modeling suggested how receptor binding could contribute to pH-sensitive pore formation. Understanding that entry mechanism could help researchers look for ways to block toxin activity—and could inform attempts to modify the system for a different purpose.
The authors wrote in their Nature paper: “The structure provides new leads for the discovery of anthrax anti-toxins, and should aid the design of cancer therapeutics.” The distinction matters: a structure can reveal useful molecular features and guide experiments, but it does not demonstrate that a proposed therapy works in people. How have researchers adapted the toxin system for tumor research?
Later research explored engineered anthrax-toxin components as a delivery platform: the goal is to make toxin activity more selective for tumors and use it to deliver a cytotoxic effect. Two strategies described in a 2016 review are changing when PA is activated and redirecting it toward receptors associated with tumors. The review reports work in animal models, which is preclinical evidence—not proof of human benefit. 2016 review of anthrax toxin-based cancer therapy research
Make activation depend on tumor-associated proteases
In this approach, researchers alter PA’s activation requirements so that enzymes associated with a tumor environment can activate it. Experimental studies of PA variants with a urokinase-sensitive activation site reported selective activation and tumor-cell killing in laboratory systems. A later report described a recombinant urokinase-activated toxin with activity against tumor cell lines and in vivo tumor models. These findings support a research concept; they do not establish a safe or effective treatment for patients. Redirect PA toward tumor-associated receptors
A second strategy changes the targeting step: engineered PA is directed toward receptors associated with tumor cells rather than relying on its usual receptor interactions. The 2016 review describes receptor retargeting among the approaches explored in preclinical tumor therapy. The choice of target and the evidence for selectivity are central questions; the existence of a targeting design alone does not show that it is safe, effective, or suitable for clinical use.
What the evidence does—and does not—establish
- Established structural result: the 2004 paper reports a 2.5 Å structure of PA bound to CMG2 and discusses implications for toxin entry and therapeutic design.
- Preclinical research: later studies explored engineered toxin components in cell systems and animal tumor models, including protease-dependent activation and receptor targeting.
- Not established by these sources: safety or efficacy in people, regulatory approval, availability as a cancer treatment, or current clinical-trial status.
Accordingly, “using anthrax to fight cancer” is shorthand for investigating engineered components of anthrax toxin as a possible platform. It should not be read as using anthrax infection itself, or as a claim that a patient can receive this as an established therapy.

Why the 2004 finding still matters
The structure connected a detailed view of how PA recognizes a host receptor with two research goals: disrupting toxin entry and learning how to redesign toxin components. That is why a basic structural result could be relevant to cancer research even though the 2004 paper did not report a cancer treatment. The subsequent engineered-platform studies show how that possibility was explored, while their preclinical status sets a clear boundary on what can be concluded.
