Choose a platform for the specific question—electrical impedance, electrode contact, insertion force, membrane rupture, or viscoelastic response—and treat its results as evidence for that endpoint, not complete device validation.
Choose a phantom for the measurement, not the tissue label
“Brain phantom” or “nerve simulator” does not tell you what a model can predict. A material may match electrical impedance while behaving unlike tissue mechanically, or reproduce a selected insertion response without matching electrical, optical, or chemical properties. Define the device, loading conditions, and measurement first.

- Electrical testing: conductivity and frequency-dependent impedance, including electrode contact.
- Mechanical testing: stiffness, viscoelastic relaxation, insertion force, dimpling, and rupture.
- Geometry: layers, membranes, channels, and the placement of the device relative to them.
- Repeatability: how consistently the phantom can be made and tested, and whether it remains usable over time.
These are distinct validation questions. A favorable result on one axis does not establish equivalence on the others.
Compare the main nonliving options
| Platform | Best-supported use in the cited studies | Main limitation |
|---|---|---|
| Saline or conductive hydrogel | Electrical conduction, electrode contact, and impedance tests. | Electrical similarity alone does not establish mechanical equivalence. The artificial-nerve study below is one specific conductive formulation. |
| Gelatin | Selected soft-tissue insertion-force testing. | In the comparative study, its relaxation was nearly elastic and differed from brain; matching stiffness did not match every interaction. |
| Composite hydrogel | Mechanical tests where viscous response matters. | The evidence concerns a particular formulation, not composite hydrogels as a class. |
| Agarose layers with a PVC dura surrogate | Probe insertion, dimpling, and membrane-rupture experiments. | The cited model targets selected rat insertion mechanics and does not reproduce all tissue features. |
| Conductive artificial nerve hydrogel | Plug-electrode contact, signal transmission, insertion, and impedance tests. | Its reported electrical values apply to that simulator design and test setup, not to nerve models generally. |
For brain-interface insertion, use a layered model when layers matter
A 2026 Frontiers in Neurology study developed a benchtop phantom with agarose layers representing pia and cortex and a thin PVC film representing dura. It was designed for pia-only and dura–pia electrode insertion tests against selected in-vivo Sprague–Dawley rat measurements.

The authors report that phantom tests preserved insertion trends across wire sizes and tip geometries, with lower repeat-trial variability. They conducted more than 600 insertion trials while finalizing the formula and fabrication protocol. The study also reports R² > 0.82 for listed microwire-diameter correlations in its phantom insertion data; neither that fit nor the trial count is a general performance benchmark.
The model has important boundaries. The authors report deviations for larger wires and dura–pia conditions, and say it does not fully reproduce nonlinear viscoelasticity, anatomical heterogeneity, membrane fibers, or arteries. It was not designed to match all electrical, optical, or chemical properties. Its intended role is benchtop insertion testing and device development, not full performance qualification; overall neural-interface evaluation still requires in-vivo results.

Published agarose concentrations
The study reports 0.5% w/v agarose for the cortex layer and 1.01% w/v for the pia layer. Its cortex preparation used agarose powder in deionized water, heated to boiling, cooled, molded, and refrigerated. These are study-specific formulations, not universal recipes; grade and preparation can matter when reproducing a phantom.
For soft-tissue mechanics, decide whether force or viscous response is the priority
In a 2015 comparative study, Leibinger and colleagues tuned gelatin and a composite hydrogel to porcine-brain stiffness. Gelatin’s insertion forces agreed closely with brain, while the composite hydrogel better reproduced viscous behavior. Gelatin’s relaxation profile differed and approached elastic behavior; the phantom responses also diverged under failure conditions despite small-strain matching.

The practical lesson is to match the interaction and loading regime, not just a stiffness value. If the experiment concerns insertion force, gelatin may be informative under conditions like those studied. If time-dependent deformation matters, the tested composite hydrogel may be a better candidate. Neither result establishes that either material reproduces brain comprehensively. The study’s abstract puts the distinction plainly: “Both materials match different characteristics of brain, but neither of them is a perfect substitute.”

In that design, the reported impedance between electrode needles was approximately 2.4–2.9 kΩ at 1 kHz, compared with approximately 2 kΩ in the pig-nerve measurements cited by the authors. These are configuration-specific study results, not universal target values for artificial or living nerves. The comparison supports use for specified bench measurements; it does not establish that the gel reproduces living peripheral nerve in general.

A phantom can isolate a question and improve control over early experiments, but it cannot establish how a device will perform across the biological variability and properties absent from that model. The 2020 primer Guidelines to Study and Develop Soft Electrode Systems for Neural Stimulation frames characterization as spanning bench and in-vivo evaluation. In practice, use a phantom to refine a relevant test, document the material and test conditions, and keep claims limited to what the setup measures.
Quick Recap
- Report formulation, geometry, preparation, temperature, insertion speed or loading conditions, and electrode configuration where relevant.
- Measure the endpoint directly: impedance for electrical behavior, force and displacement for insertion, or time-dependent response for viscoelasticity.
- Do not infer biological safety, chronic performance, or overall neural-interface efficacy from phantom testing alone.
