Non-enzymatic RNA copying can be accelerated by improving how activated building blocks interact with a template, managing inhibitory reaction products, and making copying conditions compatible with membrane compartments. Researchers have reported mixed-sequence copying with helper oligonucleotides, RNA copying inside model fatty-acid vesicles, and enhanced copying with in-situ activation chemistry. These are important advances in template-directed chemistry—not demonstrations of a self-sustaining protocell that repeatedly copies a functional genome and evolves.
What “non-enzymatic RNA replication” means
In these experiments, an RNA strand acts as a template: complementary building blocks bind along it, and a primer is extended to make a daughter strand. The chemistry does not rely on an RNA-copying enzyme, but it does rely on prepared or activated substrates and controlled reaction conditions. It is therefore more precise to call the process template-directed chemical copying than to say that RNA autonomously replicates itself.

Many experiments use nucleotides activated with imidazole-related chemistry, including 2-methylimidazole- and 2-aminoimidazole-related approaches. Activation makes a nucleotide’s phosphate more reactive. Complementary base pairing positions an incoming building block beside the growing primer, where a bond can form. Short activated oligonucleotides can also help by providing a more favorable surface for interactions between the template and incoming substrates.

Substrates must bind in a productive orientation, copying must proceed through different sequences, and the growing product must not be blocked by reaction byproducts. A complete cycle would also have to separate the copied strand from its template so both could participate in further copying.

What approaches have improved copying?
| Approach | What it changes | Reported result and scope | Evidence |
|---|---|---|---|
| Activated helper oligonucleotides | Short activated strands assist interactions between the RNA template and incoming substrates. | Copying was reported for templates containing all four nucleobases; this does not establish efficient copying of every sequence or solve strand separation. | Prywes et al., eLife, 2016 |
| Citrate with magnesium in fatty-acid vesicles | Citrate can reduce magnesium’s damaging effects on model fatty-acid membranes while retaining conditions that support RNA chemistry. | Mixed-sequence template copying containing all four nucleotides was reported inside model vesicles; this is not repeated copying of a long functional genome. | Adamala and Szostak, 2013; O’Flaherty et al., 2018 |
| In-situ activation of nucleotide mixtures | Activation chemistry is carried out in conditions designed to be more compatible with copying; the mixtures include mono- and oligonucleotides. | A 2023 study reported enhanced copying relative to mononucleotides for arbitrary RNA sequences under its experimental conditions. It does not show a self-sustaining prebiotic replication pathway. | Nucleic Acids Research, 2023 |
| Autocatalytic-system model | A theoretical model examines how RNA templating and an external supply of activated nucleotides could form a protocell reaction cycle. | The analysis treats templating as second-order autocatalysis; it is a theoretical result, not an experimental demonstration of a protocell cycle. | Sanders, Verbeem, and Higgs, Physical Review E, 2025 |
Why helper oligonucleotides matter
A major obstacle is that sequence composition affects how well copying proceeds. A- and U-rich stretches have been particularly difficult: prior work identified slow extension involving A and U, as well as poor copying of such regions. The 2016 study by Prywes and colleagues showed that activated oligonucleotides can catalyze copying of templates containing all four nucleobases. The proposed benefit is improved interaction between the template and incoming activated substrates.
This broadens the demonstrated sequence scope, but “contains all four bases” is not the same as “every arbitrary sequence copies efficiently.” The result does not establish universal sequence coverage, repeated copying, or a way for daughter and template strands to separate reliably.

How researchers address the membrane–magnesium conflict
Magnesium can promote non-enzymatic copying chemistry, but it can also destabilize fatty-acid membranes used as models of primitive compartments. That creates a systems problem: conditions that help the chemistry may harm the boundary that keeps reactants together.

Adamala and Szostak reported that citrate can protect fatty-acid membranes from disruptive magnesium concentrations while allowing RNA copying and protecting single-stranded RNA from magnesium-catalyzed degradation. In the model systems studied by O’Flaherty and colleagues, citrate-chelated magnesium also increased membrane permeability to short RNA oligomers. They reported copying of mixed-sequence templates containing all four nucleotides inside fatty-acid vesicles.

These experiments bring copying chemistry into a compartment, an important step toward protocell models. They do not show repeated replication of long functional RNA, inheritance across generations, or Darwinian evolution in a complete protocell. Citrate-mediated compatibility is a result in particular laboratory models, not a general solution for every plausible early-Earth environment.
What in-situ activation adds
Activated substrates must be available where and when template copying occurs. The 2023 Nucleic Acids Research study explored in-situ activation of mixtures containing mono- and oligonucleotides, reporting enhanced copying compared with mononucleotides for arbitrary RNA sequences under its experimental conditions.

The significance is that substrate activation and copying can be considered together rather than treating activated building blocks as a fixed input. But an experimentally useful activation mixture does not by itself establish that the activation route was available in a prebiotic setting, that it could be replenished, or that it could power complete cellular replication.

Why copying is still difficult
Sequence coverage and reaction products
Copying performance depends on sequence, and A/U-rich regions remain a known challenge. Another obstacle is product inhibition. In a 2011 Nature Chemistry study, Deck, Jauker, and Richert reported that hydrolyzed, “spent” activated monomers inhibit extension and contribute to incomplete daughter strands. Their finding highlights why simply supplying reactive building blocks may not ensure that a growing copy reaches completion.

Substrate supply and environmental fit
Activated nucleotides must be generated and replenished in a way compatible with copying. A proposed chemical route also has to fit its surroundings: for example, a magnesium concentration useful to the reaction may disrupt a fatty-acid membrane, while a compartment must admit suitable substrates without losing its integrity.
Strand separation and repeated cycles
After a daughter strand forms, it must separate from its template. If the strands remain paired or reanneal, they cannot readily serve as independent templates for another round. Primer extension on a template is therefore only one part of replication; it is not evidence that a system can run repeated cycles.
From copying to evolution
A protocell capable of Darwinian evolution would need more than short-template copying inside a vesicle. It would need repeated copying of longer functional sequences, a way to pass sequence information on, and variation that can affect the success of compartments or their contents. The 2018 review by Joyce and Szostak discusses these wider requirements for a complete protocell model.
How to interpret claims about speed or efficiency
The studies use different templates, substrates, and experimental conditions, so their results should not be collapsed into one field-wide copying rate or error rate. A report that one chemistry improves copying relative to another under its own conditions does not establish a general performance ranking. The advances described here concern particular steps—substrate interaction, membrane compatibility, in-situ activation, or system-level modeling—not a single, standardized measure of complete replication.
