Why Earth’s Life Shares the Same 20 Amino Acids

Earth’s organisms largely share an inherited genetic code, but scientists have not settled why its standard protein alphabet contains these particular 20 amino acids.

Life on Earth shares a nearly universal genetic code because organisms inherited it from common ancestors, and changing a code already used across thousands of genes can be costly. But that does not explain why the code settled on these particular 20 amino acids. Their origin remains unresolved: prebiotic chemistry, evolving metabolism, protein chemistry and historical chance may all have contributed.

What does the genetic code do?

Cells build proteins by reading messenger RNA in groups of three nucleotides called codons. Each codon specifies an amino acid or a stop signal. There are 64 possible codons but only 20 standard amino acids, so several codons can specify the same amino acid. This redundancy is a basic feature of the code described in Molecular Biology of the Cell.

The “20” refers to the canonical amino acids directly assigned by the standard code—not every amino acid found in nature or every chemical form that can occur in a protein.

Why do organisms share the code?

The strongest explanation is common ancestry. Early cellular life had a translation system, and descendants inherited it. The standard genetic code is “virtually universal among extant life forms,” Koonin and Novozhilov write in their 2017 review of its origin and evolution (Annual Review of Genetics).

Once many genes and proteins depend on particular codon assignments, changing those assignments can alter many proteins at once. A changed code can also make genes transferred between organisms harder to interpret. These costs help explain why the inherited mapping remained broadly stable, even though a few lineages have modified it.

Why these 20 amino acids?

Common ancestry explains why organisms largely share a code; it does not identify why these particular amino acids became its standard alphabet. Several ideas may account for parts of that history, but none establishes a complete, settled account of every amino acid’s entry.

Prebiotic chemistry supplied some building blocks

Amino acids can form without life, and they have been found in meteorites. Such sources could have made material available for early peptides. But availability alone is a weak explanation for the final alphabet: the relationship between likely prebiotic abundance and membership in the standard code is not strong, according to Knight, Freeland and Landweber’s review, “On the evolution of the standard amino-acid alphabet”.

That review estimates that about 80 amino acids were probably produced by abiotic synthesis before life evolved. This is the authors’ estimate, not a definitive inventory of all prebiotic chemistry. It also describes about 900 amino acids produced by natural biosynthetic pathways; that figure illustrates biochemical variety, not the number encoded in proteins.

Metabolism may have expanded the code

One proposal is that the genetic code expanded as organisms developed metabolic pathways capable of making new amino acids. Under this scenario, amino acids available early could have been joined by products of evolving biosynthesis. It offers a possible process, not a confirmed timeline: proposed orders depend on assumptions about ancient metabolism, and modern organisms make many amino acids that are not directly encoded.

Kirschning’s 2022 review surveys prebiotic and metabolic scenarios while emphasizing that why this particular number and set became encoded remains open (Chemistry – A European Journal).

Protein chemistry made the set useful, not demonstrably perfect

The 20 amino acids offer a range of chemical properties that proteins can use. That usefulness does not prove the set is uniquely optimal or was deliberately selected as a complete design. Some amino acids have overlapping properties, and proteins can perform related functions with different sequences. The standard alphabet may reflect a workable chemical repertoire shaped by availability and evolutionary history.

Historical fixation preserved the assignments

The “frozen accident” idea describes how an initially contingent set of assignments could become difficult to change after it was woven into translation and many genes. This helps explain the code’s persistence, not the exact origins of its first assignments. The hypotheses can complement one another: chemistry and metabolism may have influenced which amino acids entered, while inheritance and the costs of reassignment helped preserve the result.

Is the genetic code perfectly optimized?

Codon assignments have some error tolerance: codons differing by one nucleotide often specify the same or chemically similar amino acids. This can reduce the consequences of certain copying or translation errors. However, that pattern is not proof that the code was designed or selected from the outset to minimize errors. Koonin and Novozhilov argue that error robustness may have emerged as a by-product of early code expansion.

Are there really only 20 amino acids in life?

No. Twenty is shorthand for the standard encoded set. Some organisms use specialized translation systems to incorporate selenocysteine or pyrrolysine, and some organisms and organelles have modest variations in codon assignments. Cells also produce many amino acids that are not directly encoded, while chemical modifications after translation can change residues in proteins. These exceptions do not alter the broad pattern of a nearly universal standard code.

What the evidence can—and cannot—tell us

  • Well supported: The standard code is nearly universal, consistent with inheritance from common ancestry.
  • Plausible contributors: Prebiotic supply, metabolic evolution, useful protein chemistry and the later costs of changing codon assignments.
  • Not established: One definitive sequence showing how and why each of the 20 amino acids entered the code, or that this set is the only or uniquely optimal one.
