Salamanders can regrow limbs because an injury can activate a coordinated rebuilding program: a specialized wound covering forms, local cells respond to signals from nerves and immune cells, and a blastema—a temporary mass of activated, tissue-forming cells—develops at the stump. Humans usually repair a wound without forming that kind of blastema. We can regenerate some tissues, but current medicine cannot make a person regrow a whole arm or leg.
How salamander limb regeneration works
Axolotls, a type of salamander widely studied in laboratories, provide a detailed example. After a limb is amputated, a blood clot seals the wound. In the sequence described in an axolotl study, a specialized wound epithelium covers the cut surface within about a day. With nerve input and other local signals, cells in the stump then activate and multiply, forming a blastema that contributes to replacement tissues. The blastema is not simply a pile of generic stem cells: its progenitor cells are substantially restricted in the types of tissue they can form. The new tissues must also be organized into a functioning limb. The axolotl regeneration study describes this sequence.

Several parts of the local wound environment matter. In experimental salamander models, removing nerve input before amputation can prevent blastema formation; depleting macrophages, a type of immune cell, or failing to form an appropriate wound epithelium can also disrupt regeneration. This is why “salamanders have stem cells” is not a sufficient explanation: regeneration depends on cells, signals, and wound conditions working together. The findings are from studied salamanders, especially axolotls, and should not be assumed to apply identically to every salamander species or injury.

Most mammals, including humans, do not form blastemas. Human repair is generally tissue-specific: adult stem cells tend to support the tissue or organ where they reside, and injuries often heal through scar formation. That can close a wound and restore some function, but it does not recreate the coordinated arrangement of bones, joints, muscles, nerves, blood vessels, and skin that a complete limb requires. The U.S. National Institute of General Medical Sciences summarizes the distinction in its overview of regeneration.

Scarring is part of the contrast, but it is not established as the sole reason humans cannot regrow limbs. The broader difference is that salamander regeneration coordinates wound closure, signals, progenitor-cell behavior, and tissue patterning in a way human limb repair does not. The precise mechanisms behind this species difference remain an active area of study; current sources do not identify one evolutionary cause or one molecular switch that would enable human limb regrowth.
Humans do regenerate some tissues
Human regenerative capacity is limited, not absent. Skin and hair are continually renewed; bone can repair after a fracture; and some organs can compensate after injury. The remaining liver, for example, can grow after part of the organ is removed or damaged. There is also evidence of limited regeneration at amputated fingertip tips in children. These cases do not amount to rebuilding an entire limb, with its many interdependent tissues and precise structure.

What salamander research means for medicine
Researchers study axolotls because their regeneration can help reveal how tissues respond to injury. The U.S. National Institutes of Health Office of Research Infrastructure Programs describes the axolotl as a useful model for regenerative-medicine research, while noting that much remains to be learned before regenerative elements might be reproduced in mammals: The Amazing Axolotl: A Valuable Model for Regenerative Medicine.
Whole-limb regrowth in humans remains a research goal, not an established treatment. A 2024 review of limb-regeneration research discusses pharmacological, tissue-engineering, and cell-therapy approaches, but describes significant challenges in translating them: “Mechanisms and translational applications of regeneration in limbs: From renewable animals to humans.” Salamander biology may inform future work, but it does not mean a person can currently regrow a lost limb.

Even salamander regeneration can be disrupted
Regeneration is not an automatic or unlimited response. A 2017 axolotl study found that repeated amputations under its experimental conditions compromised later regeneration. The researchers associated altered gene expression, including aberrant amphiregulin expression, with thickened wound epithelium and severe defects. This shows that the wound-response context can interfere with regeneration even in a species capable of it; it does not establish that this specific mechanism explains human healing. The study was published in npj Regenerative Medicine.
