If COVID-19 symptoms have started, test immediately. If you were exposed but still have no symptoms, wait until at least five full days after the exposure before taking an at-home test. A single negative rapid antigen result cannot rule out infection, so repeat a negative test 48 hours later. The FDA recommends at least two antigen tests in total for someone with symptoms and three for someone without symptoms.

This schedule reflects current U.S. guidance from the Centers for Disease Control and Prevention (CDC) and the Food and Drug Administration (FDA). Test labeling, expiration information and public-health advice can change, so use the instructions supplied with your test and check the agencies’ current guidance.

COVID-19 testing timing at a glance

Situation When to start If the first at-home antigen test is negative
Symptoms of COVID-19 Test immediately, even if symptoms began only recently. Repeat after 48 hours. FDA recommends at least two tests total for people with symptoms.
Exposure but no symptoms Test at least five full days after the exposure. If symptoms appear sooner, switch to the symptomatic schedule and test immediately. Repeat after 48 hours, then repeat again 48 hours later if still negative, for three tests total.
A more reliable single negative result is important Ask a clinician about a molecular test, such as a PCR or other NAAT. CDC identifies PCR testing as an alternative to serial antigen testing; FDA says molecular tests are generally more accurate than at-home tests.

The timing applies to the situation you are in now. An exposure without symptoms is not a reason to test immediately with a rapid antigen kit, because the test may be taken before there is enough virus to detect. Symptoms change the decision: do not wait for the five-day point if illness begins earlier.

If COVID-19 symptoms have started

Use an at-home test as soon as you notice symptoms that could be COVID-19. Early symptoms can be mild or nonspecific, and a test taken promptly gives you information for decisions about work, school, household contact and medical advice. The CDC’s self-testing guidance specifically says to test immediately when symptoms are present.

If that first antigen test is negative, read the result as preliminary rather than definitive. Antigen tests are designed to detect viral proteins and can miss an infection early in its course. Follow the repeat interval in the timing table instead of assuming that one negative ends the matter. If you need an answer sooner, or if the result will affect an important medical decision, ask a health professional whether a molecular test is appropriate.

Why symptoms matter more than the calendar

The five-full-day wait is for people who remain without symptoms after an exposure. It is not a rule that overrides new illness. A sore throat, cough, fever, unusual fatigue or another symptom that raises concern should move you to immediate testing, even when the exposure was recent.

If you were exposed but have no symptoms

When you feel well after an exposure, wait at least five full days before beginning an at-home antigen testing sequence. Testing earlier can produce a negative result simply because the infection is not yet detectable. Count the interval according to the CDC’s wording and use the date of the known exposure as your reference point.

If symptoms develop during the waiting period, do not continue waiting for the scheduled date. Test at once and use the symptomatic repeat plan if the result is negative. If you remain symptom-free and the first test is negative, continue the serial schedule: another test 48 hours later and a third 48 hours after that if necessary.

When the exposure date is uncertain

If you do not know exactly when the exposure occurred, tell a clinician or testing provider what you do know: the possible dates, the type of contact and whether anyone involved was confirmed to have COVID-19. A professional can help choose a practical testing date. Avoid treating an uncertain exposure as proof that a negative rapid test is conclusive.

What a negative rapid antigen result really means

A negative at-home antigen result means the test did not detect enough target material in that sample at that moment. It does not prove that you are uninfected. The FDA explains that antigen tests can miss infections early or in people without symptoms, while the CDC advises repeat testing after a negative result.

Repeat testing improves the chance of detecting an infection that was below the test’s detection threshold the first time. Keep the interval between tests at 48 hours, use a fresh test when required by the product instructions and perform each test exactly as labeled. Do not combine swabs, reagents or components from different kits unless the manufacturer explicitly permits it.

When to choose molecular testing

Molecular tests, including PCR and other NAATs, look for viral genetic material rather than antigen. They generally have greater analytical sensitivity, particularly when infection levels are low. A laboratory or authorized molecular home test may therefore be useful when you need a more dependable single negative result, when repeated antigen testing is impractical or when a clinician recommends it.

Availability, turnaround time and authorization vary by product and location. CDC guidance identifies PCR as an alternative to repeating antigen tests; it does not mean every molecular test is interchangeable. Confirm the test’s intended use and follow the provider’s collection and timing instructions.

How to test correctly at home

  1. Identify your scenario. Decide whether you have symptoms now, are symptom-free after an exposure, or need a higher-sensitivity molecular option. If symptoms and exposure both apply, follow the symptom-based timing.
  2. Check the product before opening it. Verify that the test is authorized for your intended use, age group and specimen type. Look at the expiration information and any FDA-listed expiration extension for that product. Do not use a damaged package or a kit missing components.
  3. Read the entire insert. Collection depth, swabbing time, buffer volume, waiting period and result window differ among tests. A result read too early or after the stated window may be invalid.
  4. Collect the sample exactly as directed. Wash or sanitize your hands, prepare a clean surface and follow the kit’s instructions for the nasal or other authorized specimen. Poor collection can reduce the test’s ability to detect infection.
  5. Run and read the test on schedule. Add the specified amount of sample, set a timer for the manufacturer’s reading interval and interpret the control line and test line using the supplied diagram. Do not rely on a photo or a result viewed outside the approved time window.
  6. Record the details. Note the date, time, product name, lot information if available, symptoms and result. This record helps a clinician interpret a sequence of tests and helps you avoid losing track of the 48-hour interval.
  7. Plan the next action. A positive result should be treated as evidence of current infection and discussed with a health professional when advice is needed. A negative antigen result may require the next test in the serial schedule. An invalid result requires a new test, because it provides no usable answer.

How to interpret each type of result

Positive antigen result

A positive result on an authorized antigen test is meaningful evidence of infection, even if symptoms are mild or absent. Follow current public-health precautions and contact a clinician if you have medical risk factors, questions about treatment or uncertainty about the result. Do not wait for a later negative test to dismiss a positive one.

Negative antigen result

Use the repeat schedule appropriate to your symptoms and exposure history. Consider molecular testing if a single, more sensitive result is important. Until the situation is clearer, take reasonable steps to reduce exposing other people, particularly those at higher risk, and seek professional advice when your circumstances are complicated.

Invalid or failed test

An absent control line, an error message or a result outside the permitted reading window is invalid, not negative. Discard or set aside the failed device according to the instructions and use a new authorized test. If repeated failures occur, ask a pharmacist, testing site or clinician for help.

Choosing between an at-home antigen test and a molecular test

Factor At-home antigen Molecular test (PCR/NAAT)
Detection method Detects viral proteins. Detects viral genetic material.
Speed and convenience Usually provides an on-site result and is available online, in pharmacies and in retail stores. May require a laboratory, clinic or specialized home kit; turnaround varies.
Early or asymptomatic infection More likely to miss low-level infection, so serial testing is important after a negative result. Generally more sensitive, although performance depends on the specific authorized test and sample.
Instructions and authorization Follow the package directions, eligible-age limits and expiration information for that product. Verify the product’s authorization, specimen requirements and collection procedure before testing.

Neither category eliminates the need to read the individual product labeling. The FDA maintains a changing list of authorized at-home tests and product-specific details. A multi-test pack can be practical because the recommended strategy often requires more than one test, but no particular brand should be assumed to be medically preferred.

Common timing mistakes

  • Testing immediately after an asymptomatic exposure: a negative result at that point may be too early to be informative. Wait the recommended interval unless symptoms begin.
  • Stopping after one negative rapid test: one antigen test cannot reliably exclude infection. Complete the repeat sequence that matches your symptom status.
  • Waiting for five days after symptoms begin: symptoms call for immediate testing; the exposure timetable is not a reason to delay.
  • Ignoring the test insert: different kits use different specimen, volume and reading requirements. A correct calendar date cannot rescue an incorrectly performed test.
  • Using an expired or damaged kit without checking for an extension: confirm the current FDA information and the package labeling before use.
  • Assuming a positive result needs confirmation before taking it seriously: treat it as evidence of infection and obtain clinical advice when needed.

When to contact a healthcare professional

Contact a healthcare professional if you are unsure how to apply the schedule, cannot identify the exposure date, have medical conditions that increase COVID-19 risk, need advice about a molecular test or receive conflicting results. A clinician can consider your symptoms, exposure details, recent infection and the exact test used.

Seek prompt medical care for worsening illness or any symptom that concerns you. Testing guidance is not a substitute for assessment of breathing problems, dehydration or other serious changes. Because CDC recommendations and FDA authorizations can change, recheck the agencies’ current pages and your test’s instructions whenever you test.
