Light-field microscopy (LFM) uses a microlens array to record not only where light lands on a camera sensor, but also a sample of the directions from which it arrived. Software uses those directional clues to reconstruct a 3D volume or computationally refocused views from one camera exposure. The capture is scanless; the finished volume still requires processing, and the technique trades some spatial detail for angular information.
What does a light-field microscope capture?
A conventional wide-field microscope records light intensity across a two-dimensional image plane. In doing so, it largely combines light arriving from different directions. LFM preserves some of those directional differences by placing a microlens array in the optical path before the camera sensor.

Each lenslet directs light onto a small group of sensor pixels. The lenslet positions represent spatial samples, while pixels behind each lenslet sample different angles. Together, these measurements form a reduced, sampled version of a light field, commonly described using two spatial and two angular coordinates. A complete description of light could include additional dimensions such as wavelength, depth and time; a practical camera does not record every possible dimension.

How does one exposure produce a 3D volume?
- The objective collects light from the specimen and forms an image within the microscope’s optical path.
- A microlens array divides the light into small sub-images, preserving samples of its direction as well as its position.
- The camera records those samples together in one exposure, instead of scanning through successive focal planes to acquire a stack.
- Software uses a model of the microscope’s point-spread function and an inverse reconstruction—commonly 3D deconvolution—to estimate the volume or computationally refocus planes.
The exposure is the scanless acquisition step, not the whole process. In the 2013 method described by Broxton and colleagues, reconstruction is iterative and GPU-accelerated; processing cost and time depend on the implementation and data.
What does LFM trade for single-exposure capture?
A camera sensor has a finite number of samples. Using some of them to distinguish ray directions leaves fewer for fine lateral spatial detail. Angular information helps the system infer depth, but the balance among angular sampling, lateral resolution and axial reconstruction depends on the optical layout, objective, microlens array, camera, specimen and reconstruction method.

Broxton and colleagues modelled diffraction, which affects the measured light field at microscope scales. In a particular configuration, they reported up to an eightfold improvement in lateral resolution over the earlier computational-refocusing limits used as their comparison, based on a planar USAF test target. That result is specific to the test and configuration; it is not a general claim of eightfold improvement over other microscopes. The paper also states that its method did not reach the diffraction-limited spatial resolution of confocal, two-photon or wide-field deconvolution microscopy.
When is light-field microscopy useful?
LFM’s strongest rationale is capturing a dynamic three-dimensional scene without sequentially scanning planes. A volume reconstructed from one exposure can preserve events that might change during a scanned stack. Review literature describes research applications including neuronal activity, live-cell imaging, worm locomotion and single-molecule observations. These examples do not mean every LFM configuration can image every specimen with equivalent resolution or signal quality.

For a particular experiment, the practical question is whether the benefit of simultaneous volumetric acquisition outweighs the loss of spatial detail and the work of reconstruction.
What should you consider before comparing LFM with other methods?
- Temporal sampling: Determine whether the event requires one-exposure volumetric capture or whether a sequential stack is fast enough.
- Resolution: Specify the lateral detail and depth separation needed, including how performance changes away from the native object plane.
- Signal and sample: Consider how much signal is available per angular sample, whether the specimen is fluorescent, and whether thickness or scattering may complicate reconstruction. Conventional LFM reviews identify spatial resolution and signal-to-noise as limitations; scattering can also matter.
- Illumination and specimen health: In biological imaging with fluorescent labels, photobleaching and phototoxicity remain relevant considerations.
- Reconstruction resources: Account for processing time and computing requirements after the exposure, rather than treating acquisition speed as total turnaround time.
These are practical constraints, not universal failure modes: their importance varies with system design, specimen and reconstruction.

What does building an LFM setup involve?
The microlens array is a central optical component, but it is not a generic add-on for an ordinary microscope. Broxton and colleagues describe matching the array’s f-number to the objective’s numerical aperture and accounting for sensor placement or relay optics. The appropriate component therefore depends on the optical design; selecting an array without that design does not establish compatibility.
