What a Polystyrene Nanoplastic Study Found About Ubiquitination in HeLa Cells

A laboratory study reported reduced overall ubiquitination in HeLa cells exposed to polystyrene nanoparticles. The result is specific to its model and conditions, not evidence of effects in people.

A 2022 laboratory study found that polystyrene nanoparticles lowered the overall ubiquitination signal in HeLa cells: the authors reported a 40% reduction after 24 hours at 0.2 mg/mL. That is evidence from a specific cell experiment—not proof that nanoplastics disrupt ubiquitination in people, at environmental exposure levels, or in every type of cell.

What did the HeLa cell study find?

Della Valle and colleagues exposed HeLa cells to polystyrene nanoparticles (PS-NPs), then measured ubiquitination using western blotting. At 0.2 mg/mL after 24 hours, the overall ubiquitination signal was significantly lower than in untreated cells; band-intensity analysis indicated a 40% reduction. The result applies to that cell model and those experimental conditions.

For cell proliferation and metabolic assays, the researchers also tested 0.2 ng/mL and 0.2 μg/mL. They reported lower proliferation beginning at 0.2 μg/mL, with a larger decrease at 0.2 mg/mL. Those assays are separate from the ubiquitination measurement: the ubiquitination analysis tested 0.2 μg/mL and 0.2 mg/mL, and the reported significant reduction was at 0.2 mg/mL.

What is ubiquitination, and why might a change matter?

Ubiquitination is a cellular process in which ubiquitin—a small protein—is attached to other proteins. Depending on the target and context, this marking can affect a protein’s activity, location, interactions, or breakdown. It is one part of how cells regulate and manage proteins.

#1 Best Overall

An overall ubiquitination signal is a broad measurement. It does not identify which proteins changed, whether every type of ubiquitination changed in the same way, or what downstream effects followed. The HeLa result therefore signals an experimental change in a cellular process; it does not, by itself, establish a specific malfunction or health consequence.

How did the researchers investigate a possible mechanism?

In experiments outside the cells, the 2022 study used circular dichroism, NMR, and transmission electron microscopy to examine interactions between ubiquitin and PS-NPs. The authors reported structural rearrangement of ubiquitin and formation of a hard protein corona around the particles.

These in-vitro observations offer a possible route by which particles could interact with proteins. They do not establish the full causal pathway inside a cell or show that the structural changes caused the lower HeLa-cell ubiquitination signal.

How do other cell studies compare?

Later cell studies examined different models and outcomes. Their results add context but are not direct replications of the HeLa experiment.

Rank #3
Study and model Particles and exposure details Reported finding How it relates to the HeLa result
2022 study, HeLa cells Polystyrene nanoparticles; ubiquitination analysis at 0.2 μg/mL and 0.2 mg/mL for 24 hours At 0.2 mg/mL, the overall ubiquitination signal was significantly lower; band-intensity analysis indicated a 40% reduction. Measured an overall ubiquitination signal in a human-derived cell line.
2023 study, primary human nasal epithelial cells 50 nm and 500 nm polystyrene nanoplastics; dose and duration not stated in the supplied study summary Reported particle internalization, increased intracellular reactive oxygen species, decreased mitochondrial membrane potential, and accumulation of the autophagy markers LC3-II and p62. Examined autophagy-related effects, not the same global ubiquitination endpoint.
2026 study, mouse spermatocyte-derived cell lines 50 nm and 90 nm polystyrene nanoplastics; dose and duration not stated in the supplied study summary Reported increased ubiquitination and proteasome-dependent degradation of ferroportin 1 (FPN1), a specific protein. Measured ubiquitination of a particular protein in mouse-derived cells, not an overall signal in HeLa cells.

The different directions reported across these studies are not necessarily contradictory. They measured different things in different cell types: a broad ubiquitination signal in HeLa cells versus the modification of one specific protein in mouse-derived cells. The particle sizes, exposure conditions, and endpoints also differ.

Does this show that nanoplastics cause disease in people?

No. The HeLa experiment was conducted in cultured cells, not in people, and it does not establish what happens at ordinary environmental exposure levels. The other cited cell studies likewise do not show that exposure disrupts ubiquitination in people or causes a particular disease. These findings do not establish a human exposure threshold or a consumer remedy.
